Claims (7)
- 키토산아제를 생산하는 바실러스 속(Bacillus sp.) GM 44(KCTC 0377BP)
- ⅰ) 제1항의 바실러스 속(Bacillus sp.) GM 44(KCTC 0377BP)균주를 LB한천배지에서 배양하고 LB 액체배지에 접종한 다음 30℃에서 15시간 진탕배양하여 접종용 배양액을 수득하는 공정;ⅱ) 전기에서 수득한 접종용 배양액을 1내지 3%의 탄소원, 0.5내지 3.5%의 질소원을 함유한 MS배지에 4내지 6%의 비로 접종하고, 100내지 400rpm, 25내지 37℃, 초기 pH 6.8 및 1vvm의 통기조건에서 48내지 72시간 동안 배양하는 공정; 및,ⅲ) 전기 공정의 배양액을 원심분리하여 상등액을 농축하고, 세척한 다음, 이온교환크로마토그래피를 수행하여 0내지 0.5M의 NaCl의 직선농도구배에서 용출하며, 겔여과 크로마토그래피를 수행하고, 탈이온화수로 투석하는 공정을 포함하는 키토산아제의 제조방법
- 제2항에 있어서,LB배지는 박토-트립톤 1.0%, 박토-효모추출물 0.5% 및 NaCl 1.0%를 함유하며 pH가 7.0으로 조정된 것을 특징으로 하는 키토산아제의 제조방법.
- 제2항에 있어서,MS배지는 0.2% K2HPO4, 0.1% KH2PO4, 0.07% MgSO4·7H2O, 0.05% NaCl, 0.05% KCl, 0.01% CaCl2를 함유하며 pH가 6.8로 조정된 것을 특징으로 하는 키토산아제의 제조방법
- 제2항에 있어서,탄소원은 글리세롤 포도당, 글루코사민, 설탕, 가용성 전분 및 키토산으로 구성된 그룹으로 부터 선택되는 1종 이상의 물질을 사용하는 것을 특징으로 하는 키토산아제의 제조방법.
- 제2항에 있어서,질소원은 암모늄클로라이드, 암모늄설페이트, 옥수수 침전물, 폴리펩톤, 펩톤, 트립톤 및 효모추출액으로 구성되는 그룹으로 부터 선택되는 1종 이상의 물질을 사용하는 것을 특징으로 하는 키토산아제의 제조방법.
- 바실러스 속(Bacillus sp.) GM 44균주(KCTC 0377BP)로부터 생산되며, 다음과 같은 특성을 가지는 키토산아제,ⅰ) 키토산에 작용하여 β-1,4결합을 내부절단형으로 절단하며 키토산을 분해하여 3당 내지 8당의 키토산 올리고당을 생성하고,ⅱ) 최적 pH는 6.0, 최적 반응 온도는 70℃이며;ⅲ) 분자량은 약 45,000달톤이고; 및,ⅳ) 아미노-말단 1번부터 20번까지의 아미노산 서열은 알라닌-(미정)-(미정)-리신-글루탐산-메티오닌-리신-프로린-페닐알라닌-프로린-글루타민-글루타민-발린-아스파라진-티로신-알라닌-글리신-발린-이소루이신-리신이다.
