Abstract

다음 일반식(Ⅰ)의 신규한 3-프로페닐 세팔로스포린 화합물 및 그의 제조방법이 제공된다. 본 발명의 암모니오프로페닐 세팔로스포린 화합물은 특히 세팔로스포린계 항생제에 내성이 강한 균주인 엔테로박터 클로아케 P99에 대해 기존의 광범위 항생제로서 널리 알려진 세프타지딤보다 월등한 항균력을 보일 뿐 아니라 그램 양성균에 대해서도 세프타지딤보다 우수한 항균력을 나타낸다.
식중, P는 상기 정의된 바와 같다.

Description

암모니오세팔로스포린 계열 항생제 및 그의 제조 방법
본 발명은 신규한 암모니오세팔로스포린 화합물 및 그 제조 방법에 관한 것이다. 더욱 상세하게는 광범위하게 항균력을 나타내며, 특히, 슈도모나스 균주 및 엔테로박터 클로아케 P99를 비롯한 그램 음성균에 탁월한 활성을 나타내어 항생제로 유용한 다음 일반식(Ⅰ)로 표시되는 세팔로스포린 화합물 및 그 제조 방법에 관한 것이다.
상기 일반식(Ⅰ)에서 P는 하나의 질소를 갖는 동시에 한 개 또는 두 개의 히드록실기를 갖는 비고리형 아민 또는 헤테로시클릭아민으로 메조-3,4-디히드록시-1-메틸피롤리딘,(3S,4S)-3,4-디히드록시-1-메틸피롤리딘, (3R, 4R)-3,4-디히드록시-1-메틸피롤리딘, (2S, 4R)-4-히드록시-1-메틸-2-피롤리딘메탄올, N-메틸-비스(2-히드록시에틸)아민, 3,4-시스-디히드록시-1-메틸피페리딘, 3,4-트란스-디히드록시-1-메틸피페리딘, 4-히드록시-1-메틸피페리딘, 2-히드록시메틸-1-메틸피페리딘 또는 트로핀을 나타낸다.
또한, 이들 세팔로스포린 화합물들의 약학적으로 허용되는 염, 예를 들면 나트륨염이나 칼륨염과 같은 알칼리염, 테트라에틸암모늄염, 베타인염, 칼슘염이나 마그네슘염과 같은 알칼리토금속염, 염산염, 브롬산염, 황산염과 같은 무기염, 탄산염, 중탄산염, 메탄술포네이트, 벤젠술포네이트, 타우린, 톨루엔술포네이트와 같은 유기 술포네이트, 아세테이트, 말리에이트, 락테이트, 타르타레이트와 같은 유기산 카르복실레이트, 아르지닌염, 라이신염, 세린염, 아스파르테이트, 글루타르메이트와 같은 아미노산 염, 트리에틸아민, 피리딘, 디시클로헥실아민과 같은 아민 염, N,N-디벤질에틸렌아민 염, 트리에탄올 염, 트리스(히드록시메틸아민)메탄염, 펜에틸벤질아민염 등의 무독성인 염 형태도 본 발명에 포함된다.
일본공개특허공보 제54-154,786호에는 세팔로스포린의 3위치에 4급암모늄염을 포함한 메틸기, 즉 피리디니오메틸기가 도입된 세프타지임(ceftazidime)의 합성방법이 기술되어 있다. 세프타지딤은 슈도모나스균과 같은 그램-음성균에 대해서는 높은 광범위 항균력을 나타내지만 엔테로박터 클로아케 P99 및 그램-양성균에 대해서는 상대적으로 낮은 항균력을 보이고 있다. 이 화합물의 개발을 계기로 보다 향상된 항균력을 지닌 항생제의 개발을 위한 시도가 이루어졌으며, 그 중에 세피롬(cerfpirome, Drugs of the Future 13,369-371, 1988) 및 세페핌(cefepime, BMY-28142, Drugs of the Future 10, 805-808, 1985) 등의 화합물들은 슈도모나스균 뿐만 아니라 스타필로코코스균에 대해서도 강력한 항균력을 나타내고 있다.
특히, 본 발명과 유사하게 3-위치에 암모니오프로페닐 그룹이 도입된 세팔로스포린 유도체들의 합성이 유럽특허 제462,009호, 유럽 특허 제528,343호 및 Journal of Antibiotics 1990년, 43권, 제5호, 533-543 페이지에 자세히 기술되어 있다.
본 발명자들은 그램 양성균 및 그램 음성균에 광범위한 항균작용을 가진 동시에 강력한 항균력을 보유한 세팔로스포린계 항생제를 개발하기 위해 연구해 오던 중, 7-위치에 아미노티아졸기를 가지고 있으며, 3위치에 히드록실기가 치환된 아민을 도입한 3-암모니오프로페닐 세팔로스포린 화합물이 슈도모나스 균주 및 엔테로박터 클로아케 P99를 비롯한 그램 음성균에 탁월한 항균작용을 나타낼 뿐만 아니라 그램-양성균에 대해서도 강력한 항균력을 나타낸다는 사실을 발견하였다. 따라서, 항생제로 유용하게 사용될 수 있는 세팔로스포린 유도체를 제조하고, 이들 화합물들을 효과적으로 제조하는 것이 본 발명의 목적이다.
이하 본 발명을 상세히 설명하면 다음과 같다.
본 발명의 상기 일반식(Ⅰ)의 세팔로스포린 화합물 및 약학적으로 허용되는 이들의 염은, 일반식(Ⅱ)로 표시되는 화합물과 일반식(Ⅲ)으로 표시되는 3급 아민화합물 p를 반응시킨 후에 트리틸기와 p-메톡시벤질기를 제거하여 제조할 수 있다.
일반식 (II)에서, X는 할로겐 원자이다.
일반식(Ⅲ)에서 P는 메조-3,4-디히드록시-1-메틸피롤리딘, (3S, 4S)-3,4-디히드록시-1-메틸피롤리딘, (3R, 4R)-3,4-디히드록시-1-메틸피롤리딘, (2S, 4R)-4-히드록시-1-메틸-2-피롤리딘메탄올, N-메틸-비스(2-히드록시에틸)아민, 3,4-시스-디히드록시-1-메틸피페리딘, 3,4-트란스-디히드록시-1-메틸피페리딘, 4-히드록시-1-메틸피페리딘, 2-히드록시메틸-1-메틸피페리딘 또는 트로핀이다.
일반식(Ⅱ)에서 X로 표시되는 할로겐 원자는 또 다른 할로겐 원자로 전환될 수 있다. 예를 들면, X가 요오드인 일반식(Ⅱ)의 화합물은 X가 염소인 일반식(Ⅱ)의 화합물을 아세톤 용매에서 1-5당량의 Nal 또는 KI와 반응시켜 얻을 수 있다. 요오드화하는 반응에서 일반식(Ⅱ)의 화합물은 거의가 E-이성체로 얻어진다.
일반식(Ⅲ)로 표시되는 아민 화합물 P가 가끔 반응용매에 잘 녹지 않는 경우에는 일반식(Ⅱ)와의 반응이 잘 진행되지 않는다. 따라서, 1-5 당량의 실릴화제를 사용하여 화합물 P를 실릴화되면 반응용매에 잘 녹게 되어서 반응이 잘 진행된다. 바람직한 실릴화제는 N-메틸트리메틸실릴트리플루오로아세트아미드(MSTFA)이며, N,O-비스트리메틸실릴아세트아미드(BSA), 비스트리메틸실릴유레아, N-메틸트리메틸실릴아세트아미드 등의 다른 실릴화제를 사용할 수도 있다. 실릴화 반응은 일반식(Ⅲ)으로 표시되는 아민 화합물 P를 반응용매에 현탁시킨 후, -15℃∼60℃, 바람직하게는 -15℃∼0℃의 온도에서 실릴화제를 가하고 교반하여 주면, 실릴화 반응이 진행되어 아민 화합물이 반응용매에 녹아 들어간다. 이때 사용하는 반응용매는 아세톤, 아세토니트릴, 디메틸술폭사이드, 디클로로메탄, 클로로포름, 사염화탄소 등과 같은 탄화수소, 디에틸에테르, 테트라하이드로푸란, 디옥산과 같은 에테르, 벤젠, 톨루엔, 크실렌과 같은 방향족 화합물, 또는 이들의 혼합물들을 사용할 수 있으며, 톨루엔이 바람직한 용매이다.
일반식(Ⅰ)로 표시되는 새로운 세팔로스포린 화합물은 -15℃∼40℃의 온도에서 전술한 실릴화된 아민용액을 일반식(Ⅱ)로 표시되는 화합물과 반응시킨 후, 트리틸기와 p-메톡시벤질 그룹을 트리플루오로아세트 산과 아니솔을 사용하여 제거하면 얻을 수 있다. 실릴화된 아민용액을 일반식(Ⅱ)와 일반식(Ⅱ)로 표시되는 화합물의 반응시에 사용하는 용매는 바람직하게는 일반식(Ⅱ)로 표시되는 아민 화합물 P를 실릴화시킬 때 사용하는 용매와 동일한 용매이다.
한편, 본 발명에서 제조된 세팔로스포린 화합물들은 그램 양성균 및 그램 음성균을 포함한 광범위한 균주들에 대하여 높은 항균력을 보인다. 그 유용성은 공지의 화합물인 세프타지딤(ceftazidime)을 비교 물질로 하여 최소억제농도(Minimum Inhibitory Concentration)를 조사하여 평가하였다. 최소억제농도는 뮐러-힌튼 한천 (Mueller-Hinton Agar)에서 한천희석법으로 조사하였고, 배양접시에 5×104cfu의 균주를 37℃에서 24시간 동안 배양하여 구하였으며, 그 결과를 표에 나타내었다.
표에서 알 수 있는 바와 같이, 본 발명의 세팔로스포린 화합물들은 모두 광범위하고 우수한 항균력을 나타내고 있다. 특히, 세팔로스포린 항생제에 내성이 강한 균주인 엔테로박터 클로아케 P99(Enterobacter cloacae P99)에 대해 광범위 항생제로 널리 알려진 대조약제인 세프타지딤의 최소억제농도가 100㎍/㎖ 이상인 반면, 본 발명의 화합물들은 3.13-12.5㎍/㎖의 최소억제농도를 나타내어 세프타지딤보다 16배 이상의 월등한 항균력을 보이고 있을 뿐만 아니라, 녹농균(슈도모나스균)을 포함한 이외의 표준균주에 대해서도 우수한 항균력을 나타내고 있다. 또한, 포도상구균(스타필로코코스 아우레우스)에 대해 세프타지딤의 항균력이 3.13∼12.5㎍/㎖인 반면, 본 발명의 화합물들은 0.2∼0.78㎍/㎖의 최소억제농도를 나타내어 세프타지딤보다 30배 이상의 월등한 항균력을 보이고 있는 것으로 보아 본 발명의 화합물들이 대조약제로 사용된 세프타지딤보다 더 광범위하며, 유용하다는 사실을 시사하고 있다. 한편, 본 발명의 화합물들은 통상의 세팔로스포린 화합물의 경우와 같이 멸균용 주사용수에 희석하여 주사제로서 투여할 수 있다.
다음 실시예는 본 발명을 더욱 상세히 예증하여 줄 것이나 본 발명의 범위가 이들 실시예로 한정되는 것은 아니다.
[실시예 1]
7β-[(Z)-2-(아미노티아졸-4-일)-2-메톡시이미노아세트아미도]-3-[(E)-3-메조-3,4-디히드록시-1-메틸-1-피롤리디니오)-1-프로펜-1-일]-3-세펨-4-카르복실레이트(I-a)의 제조
p-메톡시벤질 7β-[(Z)-2-(트리틸아미노티아졸-4-일)-2-메톡시이미노아세트아미도]-3-[(Z)-3-클로로-1-프로펜-1-일]-3-세펨-4-카르복실레이트 (300㎎, 0.47밀리몰)를 아세톤(15㎖)에 녹인 용액에 Nal(165㎎, 1.10 밀리몰)를 한번에 모두 가하고 15℃∼25℃에서 2시간 동안 교반하였다. 반응혼합물을 농축한 후 에틸 아세테이트로 희석한 후 10% NaSO수용액과 소금물로 차례대로 씻어주고 건조, 증발시켰다. 이때 얻어지는 잔사를 톨루엔(5㎖)에 녹인 후, 이 용액에 메조-3,4-디히드록시-1-메틸 피롤리딘(86㎎, 0.73밀리몰)을 톨루엔(1㎖)에 현탁시키고 N-메틸트리메틸실릴아세트아미드(436㎎, 2.19 밀리몰)를 -10℃에서 가하여 얻은 용액을 가한 후 냉장고 (약 -10℃)에서 12시간 동안 방치하였다. 이때 생성되는 고체를 여과하고 에테르로 여러차례 씻어주어 약 290㎎의 흰색 고체를 얻었다. 이 고체를 디클로로메탄(0.5㎖)과 트리플루오로아세트산(1㎖)과 아니솔(0.5㎖)에 가한 후 15℃∼25℃에서 2시간 동안 교반하였다. 반응 혼합물을 농축한 후 이소프로필 에테르(20㎖)를 가하여 고체상의 물질을 얻었다. 이 고체를 이소프로필 에테르로 여러 차례 씻어주고 중탄산나트륨 용액으로 중화한 후 실리카겔 컬럼 크로마토그라피(CHCN/HO=4:1-2:1)로 정제하여 표제화합물(74㎎, 38%)을 얻었다.

[실시예 2]
7β-[(Z)-2-(2-아미노티아졸-4-일)-2-메톡시이미노아세트아미도]-3-[(E)-3S, 4S-3,4-디히드록시-1-메틸-1-피롤리디니오)-1-프로펜-1-일]-3-세펨-4-카르복실레이트(I-b)
3S, 4S-디히드록시-1-메틸피롤리딘을 사용하고, 실릴화제로서 N-메틸 트리메틸실릴아세트아미드 대신 N,O-비스트리메틸실릴아세트아미드(BSA)를 사용한 것을 제외하고, 실시예 1과 동일한 방법으로 표제화합물을 얻었다. 수율 25%, IR(KBr)3400, 1766, 1606, 1534mc ; H NMR(DO) δ7.03(1H, s), 6.93(1H, d, J=15.1Hz), 5.97(1H, m), 5.83(1H, d, J=4.5Hz), 5.27(1H, d, J=4.5Hz), 4.55(2H, m), 4.10(1H, d, J=7.3Hz), 4.00(3H, s), 3.65(2H, s), 3.60-4.05(4H,m), 3.24(3H, s).
[실시예 3]
7β-[(Z)-(2-아미노티아졸-4-일)-2-메톡시이미노아세트아미도]-3-[(E)-4R-히드록시-2S-히드록시메틸-1-메틸-1-피롤리디니오)-1-프로펜-1-일]-3-세펨-4-카르복실레이트 (I-c)
(2S, 4R)-4-히드록시-1-메틸-2-피롤리딘메탄올을 사용하여, 실시예 2와 동일한 방법으로 표제화합물을 얻었다. 수율 28%, IR(KBr)3422, 1766, 1606, 1534mc 1; H NMR(DO) δ7.03(1H, s), 6.94(1H, d, J=15.5Hz), 6.02(1H, m), 5.84(1H, d, J=4.6Hz), 5.28(1H, d, J=4.6Hz), 4.30(1H, m), 4.20(2H, m), 4.00(3H, s), 3.06(3H, s), 2.15-2.50(2H, m).
[실시예 4]
7β-[(Z)-(2-아미노티아졸-4-일)-2-메톡시이미노아세트아미도]-3-[(E)-3-비스(2-히드록시메틸)메틸암모니오)-1-프로펜-1-일]-3-세펨-4-카르복실레이트 (I-d)
p-메톡시벤질 7β-[(Z)-(2-트리틸아미노티아졸-4-일)-2-메톡시이미노아세트아미도]-3-[(Z)-3-클로로-1-프로펜-1-일]-3-세펨-4-카르복실레이트 (457mg, 0.56밀리몰)를 아세톤 (15㎖)에 녹인 용액에 Nal(252㎎, 1.68밀리몰)을 한번에 모두 가하고 15℃∼25℃에서 2시간 동안 교반하였다. 반응혼합물을 농축한 후 에틸 아세테이트로 희석한 후 10% NaSO수용액과 소금물로 차례대로 씻어주고 건조, 증발시켰다. 이때 얻어지는 잔사를 톨루엔(5㎖)에 녹인 후, 이 용액에 N-메틸-비스(2-히드록시메틸)아민(131mg, 1.0밀리몰)을 톨루엔(1ml)에 현탁시킨 용액을 가한 후 냉장고(약-10℃)에서 12시간 동안 방치하였다. 이때 생성되는 고체를 여과하고 에테르로 여러 차례 씻어주어 흰색 고체를 얻었다. 이 고체를 디클로로메탄(0.5㎖)과 트리플루오로아세트산(1㎖)과 아니솔(0.5㎖)에 가한 후 15℃∼25℃에서 2시간 동안 교반하였다. 반응 혼합물을 농축한 후 이소프로필 에테르(20㎖)를 가하여 고체상의 물질을 얻었다. 이 고체를 이소프로필 에테르로 여러 차례 씻어주고 중탄산나트륨 용액으로 중화한 후 실리카겔 컬럼 크로마토그라피(CHCN/HO=4:1-2:1)로 정제하여 표제화합물(78㎎, 26%)을 얻었다. 3,4-트란스-디히드록시-1-메틸피페리딘을 사용하여 실시예 1과 동일한 방법으로 표제화합물을 얻었다. 수율 49%; IR(KBr)3414, 1766, 1604, 1536cm ; H NMR(DO) δ7.01(1H, s), 6.93(1H, d, J=15.5Hz), 5.95(1H, m), 5.82(1H, d, J=4.5Hz), 5.26(1H, d, J=4.5Hz), 4.13(2H, m), 1.86-4.16(8H, m), 3.98(3H, s), 3.033.15(3H, two s).

4-히드록시-1-메틸피페리딘을 사용하여 실시예 1과 동일한 방법으로 표제화합물을 얻었다. 수율 40%; IR(KBr)3414, 1766, 1606, 1536mc ; H NMR(DO) δ7.09(1H, s), 6.95(1H, d, J=15.5Hz), 6.05(1H, m), 5.90(1H, d, J=4.5Hz), 5.34(1H, d, J=4.5Hz), 4.07(3H, s) 1.95-4.15(11H, m), 3.083.11(3H, two s).

7β-[(Z)-2-(2-아미노티아졸-4-일)-2-메톡시이미노아세트아미도]-3-[(E)-3-(2-히드록시메틸-1-메틸-1-피페리디니오)-1-프로펜-1-일]-3-세펨-4-카르복실레이트 (I-h)
2-히드록시메틸-1-메틸피페리딘을 사용하여 실시예 1과 동일한 방법으로 표제화합물을 얻었다. 수율 28%; IR(KBr)3408, 1766, 1604, 1536cm ; H NMR(DO) δ7.03(1H, s), 6.93(1H, d, J=15.6Hz), 5.93(1H, m), 5.83(1H, d, J=4.6Hz), 5.27(1H, d, J=4.6Hz), 4.62(1H, m), 4.20(2H, m), 4.00(3H, s),3.70(3H, s), 1.50-3.93(10H, m), 3.033.13(3H, two s).

7β-[(Z)-2-(2-아미노티아졸-4-일)-2-메톡시이미노아세트아미도]-3-[(E)-3-트리피니오-1-프로펜-1-일]-3-세펨-4-카르복실레이트 (I-i)
트로핀을 사용하여 실시예 1과 동일한 방법으로 표제화합물을 얻었다. 수율 61%; IR(KBr)3422, 1764, 1618, 1534cm ; H NMR(DO) δ7.03(1H, s), 6.95(1H, d, J=15.5Hz), 5.98(1H, m), 5.84(1H, d, J=4.3Hz), 5.28(1H, d, J=4.3Hz), 4.18(2H, m), 4.01(3H, s), 1.95-4.13(11H, m), 3.03(3H, s).

Claims (4)

  1. 다음 일반식 (Ⅰ)의 신규한 3-프로페닐 세팔로스포린 화합물 또는 이들의 약학적으로 허용되는 염.
    식중 P는 메조-3,4-디히드록시-1-메틸피롤리딘,(3S,4S)-3,4-디히드록시-1-메틸피롤리딘, (3R, 4R)-3,4-디히드록시-1-메틸피롤리딘, (2S, 4R)-4-히드록시-1-메틸-2-피롤리딘메탄올, N-메틸-비스(2-히드록시에틸)아민, 3,4-시스-디히드록시-1-메틸피페리딘, 3,4-트란스-디히드록시-1-메틸피페리딘, 4-히드록시-1-메틸피페리딘, 2-히드록시메틸-1-메틸피페리딘 및 트로핀 중에서 선택된다)
  2. 다음 일반식(Ⅲ)의 아민 화합물 P를 다음 일반식(Ⅱ)의 화합물과 반응시키는 것으로 이루어지는 제1항에 따른 일반식(Ⅰ)의 세팔로스포린 화합물의 제조 방법.
    (식 중, X는 할로겐 원자이다)
    (P는 메조-3,4-디히드록시-1-메틸피롤리딘,(3S,4S)-3,4-디히드록시-1-메틸피롤리딘, (3R, 4R)-3,4-디히드록시-1-메틸피롤리딘, (2S, 4R)-4-히드록시-1-메틸-2-피롤리딘메탄올, N-메틸-비스(2-히드록시에틸)아민, 3,4-시스-디히드록시-1-메틸피페리딘, 3,4-트란스-디히드록시-1-메틸피페리딘, 4-히드록시-1-메틸피페리딘, 2-히드록시메틸-1-메틸피페리딘 및 트로핀 중에서 선택된다)
  3. 제2항에 있어서, 일반식(Ⅲ)의 아민 화합물 P를 일반식(Ⅱ)의 화합물과 반응시키기 전에 2 내지 5당량의 실릴화제에 의해 실릴화시키는 것이 특징인 방법.
  4. 제3항에 있어서, 실릴화제가 N-메틸트리메틸실릴트리플루오로아세트아미드(MSTFA), N,O-비스트리메틸실릴트리플루오로아세트아미드(BSTFA), 헥사메틸디실라잔(HMDS), N,O-비스트리메틸실릴아세트아미드(BSA), 비스트리메틸실릴유레아, 및 N-메틸트리메틸실릴아세트아미드 중에서 선택되는 것이 특징인 방법.
