Abstract
Description
Claims (13)
- 백혈구 유착 병소를 타게팅하기 위한 키메라 항체-시약 접합체 (chimeric antibody-agent conjugate)로서, 상기 키메라 항체는 항원-결합 초가변 영역(antigen-binding hypervariable region) 및 인간 면역 글로불린의 불변 영역(constant region)을 갖고 있으며, 상기 항원-결합 초가변 영역은 과립구에 특이적으로 결합하는 특징을 갖고, 상기 불변 영역은 단핵 림포이드 세포상의 수용체에 대한 친화력이 높은 Fc 부위를 갖는 특징을 가지며, 상기 시약은 50-500KeV 범위의 감마선을 방출하는 방사성 동위원소, 양전자-방출 방사성 동위원소 또는 자기 공명 영상 증강제(magnetic resonance imaging enhancing agent)에서 선택된 적어도 하나의 진단용 영상화제 ; 또는 방사성 동위원소 또는 항미생물제에서 선택된 적어도 하나의 치료제임을 특징으로 하는 키메라 항체-시약 접합체.
- 제1항에 있어서, 상기 진단용 영상화제로 사용되는 방사성 동위원소는 테크네튬-99m(Tc-99m), 요오드-123(I-123), 갈륨-67(Ga-67), 인듐-111(In-111) 또는 요오드-131(I-131)에서 선택된 것임을 특징으로 하는 접합체.
- 제1항에 있어서, 상기 자기 공명 영상 증강제는 가돌리늄 또는 망간 이온을 함유하는 것임을 특징으로 하는 접합체.
- 제1항에 있어서, 상기 초가변 영역이 Ia(DR) 항원에 특이적으로 접합되어 있음을 특징으로 하는 접합체.
- 제1항에 있어서, 상기 치료제로 사용되는 동위원소는 구리-67, 요오드-125, 요오드-131, 레늄-186, 레늄-188, 비스무트-212 또는 아스타틴-211에서 선택된 것임을 특징으로 하는 접합체.
- 제1항에 있어서, 상기 치료제는 방사성 동위원소와 항미생물제의 조합물(combination)임을 특징으로 하는 접합체.
- 제1항에 있어서, 상기 항미생물제는 항균제, 항바이러스제, 항진균제 또는 항기생충제로부터 선택된 것임을 특징으로 하는 접합체.
- 제1항에 있어서, 상기 치료제는 텍스트란 또는 아미노덱스트란에서 선택된 고분자 담체를 이용하여 상기 키메라 항체에 접합되어 있음을 특징으로 하는 접합체.
- 진단용 영상화제를 염증 또는 감염성 병변에 타게팅하는데 사용되는 조성물로서 특허청구의 범위 제1항, 2항, 3항 또는 4항에 따른 키메라 항체-시약 접합체를 함유함을 특징으로 하는 조성물.
- 치료제를 염증 또는 감염성 병변에 타게팅하는데 사용되는 조성물로서 특허청구의 범위 제1항, 5항, 6항, 7항 또는 8항에 따른 키메라 항체-시약 접합체를 함유함을 특징으로 하는 조성물.
- 제10항에 있어서, 키메라 항체의 초가변성 영역이 과립구의 Ia(DR) 항원에 특이적으로 결합함을 특징으로 하는 조성물.
- 제1항, 2항, 3항 또는 4항에 있어서, 상기 접합체에 특이적으로 결합하는 비(非) 표지화된 이차 항체 또는 항체 단편을, 비경구적으로 투여할 때 2-72시간 이내에 상기 접합체의 집중화율을 적어도 약 20% 증가시키는 양으로 함께 사용함을 특징으로 하는 접합체.
- 제1항, 5항, 6항 7항 또는 8항에 있어서, 상기 병변 부위에 있는 백혈구가 선택적으로 최대한의 접합체를 포획할 수 있는 시간이 지난 후, 상기 접합체에 특이적으로 결합하는 비(非) 표지화된 이차 항체 또는 항체 단편을, 비경구적으로 투여할 때 상기 치료제의 치료지수를 적어도 약 10% 증가시키는 양으로 함께 사용함을 특징으로 하는 접합체.
