Abstract

PURPOSE:To obtain a nontoxic red natural pigment containing a substance, such as crocin or crocetin, by transplanting a dedifferentiated callus induced from a saffron tissual piece into a culture medium containing alanine added thereto and cultivating the callus. CONSTITUTION:A bulb, stem, root, etc., of a saffron are sterilized and sliced to provide a tissual piece thereof, which is then planted to a culture medium, such as MS culture medium, containing an added pant hormone to carry out induction of a callus and afford a dedifferentiated callus. The resultant dedifferentiated callus is subsequently transplanted to a culture medium containing alanine added thereto and cultivated to produce a red pigment in the obtained culture. The obtained pigment is then extracted from the afore- mentioned culture with a solvent, such as butanol, to provide the aimed red natural pigment suitable as a colorant for jam, wine, etc.
