Abstract

PURPOSE:To obtain an immune detection device which obviates the exfoliation from a substrate and has high sensitivity by providing a light interference layer which is optimized in thickness and refractive index and chemically bonding an antibody layer selectively to the surface of the light interference layer except in the recognized active section of an antibody. CONSTITUTION:This simple device for immune detection is constituted of the light reflective substrate 1 which consists of gold, silver, etc., and has substantially no irregular reflection, the light interference layer 2 which is laminated on the substrate 1, a reactive intermediate layer 3 which is provided on the light interference layer 2 and consists of a compd. reacting selectively with the carboxyl group or thiol group contained in the antibody molecules or fragmented antibody molecules mainly by pH adjustment, and a layer 4 of an antigen material and/or antibody protein which is provided on the intermediate layer 3 and consists substantially of a monomolecular layer. The light interference layer 2 has no reflection characteristic to visible light and the increase in the thickness arising from the antigen-antibody reaction of the antibody protein layer appears as a change in the interference color of light. Further, the surface of said layer is required to have sufficient affinity or reactivity to or with the compd. forming the intermediate layer 3 and is formed by controlling long chain carboxylic acid and the metal salt thereof or silicon oxide, etc., to a desired film thickness.
