Abstract

PURPOSE: An analytical composition for choline, betaine or a derivative thereof containing a novel choline oxidase, 4-aminoantipyrine, peroxidase, etc. as a color reagent.
CONSTITUTION: A novel choline oxidase is collected from cultivated Arthrobacter globiformis B-0577 (FERM-P No.3518) by the conventional cultivating and collecting method. The resultant choline oxidase is then added to a solution of choline or a derivative thereof, e.g. lysolecithin, sphingomyelin or bytyrylcholine, or betaine aldehyde or a derivative thereof in an aqueous medium. The resultant hydrogen peroxide formed by the enzymic reaction is then determined by a peroxidase, etc. or the formed betaine is similarly determined by the Reinecke salt method. Alternatively, the reduced oxygen may be determined by an oxygen electrode.
