Abstract

PURPOSE: To obtain a prolonged release type composite having improved prolonged release properties and the desired shape, by mixing a powdery polypeptid with a powdery physiologically active substance, and treating the resultant mixture under pressure.
CONSTITUTION: A powdery polypeptide, e.g. poly-alanine or polyglycine, is mixed with a physiologically active substance, e.g. beomycin drochloride, growth hormone, or antihistaminic agent, and the resultant mixture is then treated under a pressure of 50W20,000kg/cm2 to give the aimed prolonged release type composite of desired shape. According to the method, the polypeptide becomes a film in appearance, and the physiologically active substance is uniformly dispersed and covered with the film-like polypeptide to give the prolonged release type composite having a long duration of drug effects. The two components must be finely pulverized prior to the mixing thereof, e.g. by a method of pulverizing the components in a forzen state at -190°C (liquid nitrogen temperature).
