Abstract

PURPOSE: To make possible the determination of L-lysine simply and precisely by sealing a microorganism germ having L-lysine decarbonization enzyme activity and using a microorganism electrode having dissolved carbon dioxide gas film on which a microorganism film having a microorganism film fixed.
CONSTITUTION: A microorganism electrode 7 in which a microorganism germ 2 having a L-decarbonization enzyme activity is sealed between a diaphragm 1 of carbon dioxide gas electrode and a thin film 4 having a finely small opening covering thereon or a microorganism electrode 7 in which a microorganism germ fixing microorganism film is attached onto a diaphragm of dissolved carbon dioxide gas electrode is employed. This microorganism electrode 7 is immersed into L-lysine containing liquid to be tested or contacted therewith under an air repellent condition, and electromotive force is measured to determine L-lysine. For example, dissolved carbon dioxide gas electrode 1, a microorganism layer 2, a carrier 3 a dialysis film (cellophane film), pH electrode 6, an internal liquid 6 (NaHCO3-NaCl) constitute the microorganism electrode 7.
