Abstract
PURPOSE: Using a new substrate of anilide derivative is measured the activity of leucine aminopeptidase in samples from living bodies, which is required for diangoses of acute hepatitis, hepatic cancer, metastatic hepatic cancer, hepato- cirrhosis, cholangia, etc.
CONSTITUTION: A derivative of L-leucyl-4-hydroxyanilide of the formula is used as the substrate and the 4-hydroxyaniline derivative formed by the action of LAP is condensed through oxidation to form a colored compound, which is colorimetrically determined, whereby the activity of LAP in samples from living bodies can be measured. The L-leucy 1-4-hydroxyanilide derivative, the substrate, is obtained by reacting usually activated L-leucine having protected amino group with p-hydroxyaniline derivative followed by removing the protecting group from the condensation product. As shown in the table, the substrate in the present invention is greatly excellent in substrate reactivity compared to conventional substrates.
