Abstract

PURPOSE: To obtain the title compound, which is an antibacterial, by treatment of the reaction product between cephamycin C and a tetrazole with an oxalyl halide and an alcohol, and by reduction, halogenation, and introduction of a cyanomethylthio group to exchange the acyl group at the 7β-position.
CONSTITUTION: A compound of formula I: (R1 is amino-protecting group) is reacted with 1-methyl-1H-5-mercapto-tetrazole and then an oxalyl halide, e.g. oxalyl chloride, in the presence of a base, e.g. Na2CO3, and treated with water to give a compound of formula II, which is reacted with a compound of the formula R2OH (R2 is lower alkyl group) in the presence of a mineral or Lewis acid, and reduced with a metal hydride to form a compound of formula III. The resulting compound is reacted with a halogenating or sulfonylating agent, then with a compound of the formula NCCH2S-M (M is alkali metal) to afford the objective cyanomethylthio derivative of formula IV.
