Claims (1)
- 【特許請求の範囲】 1.液体試薬中の固体担体に、ポリマー構成単位を連続的に添加することにより ポリマー鎖を形成するための、ポリマー合成装置であって、 反応ウエルを含み、かつ該ウエル内に延びた少なくとも一つのオリフィスを 有するベースアセンブリーと、 該ウエル内に配置された少なくとも一つの固体担体と、ここで該固体担体上 でポリマー鎖を成長させかつこれに固定する、 該ウエル中で、該固体担体および該固体担体に固定された該ポリマー鎖の少 なくとも1種のポリマー構成単位と接触状態にある、試薬溶液と、 該ウエル内に配置され、かつ該固体担体の該オリフィスの通過を実質的に防 止するように形成され、かつそのような寸法とされた保持手段と、 該オリフィスが該ウエルへの入口とそこからの出口とを有し、かつ該ウエル 中に該試薬溶液を維持するための毛管液体封止を形成するサイズと寸法とをもつ ものであって、該反応ウエル上にかかる第一のガス圧と、該オリフィス出口にか かる第二のガス圧との間の差圧が予め定められた値未満である場合に、該ウエル 内でのポリマー鎖の成長を可能とし、および 該差圧が該予め定められた値を越えた場合に、該試薬溶液が該オリフィスを 通して該ウエルから追い出されるように、該差圧を制御するための圧力調節デバ イスと、 を含むことを特徴とする上記ポリマー合成装置。 2.該保持手段が、該ウエル内の、該一つのオリフィスと該支持構造との間に設 けられたフィルタ部材を含む、請求の範囲第1項に記載のポリマー合成装置。 3.該フィルタ部材がガラス線維フリットで構成される、請求の範囲第2項に記 載のポリマー合成装置。 4.該フィルタ部材と該オリフィスとが、該毛管液体封止を形成するように協働 する、請求の範囲第2項に記載のポリマー合成装置。 5.更に、該ベースアセンブリーと協働して、内部に該反応ウエルを収容する上 部チャンバーを形成する、上部チャンバー機構を含み、該調節デバイスが該上部 チャンバーと連通して、該差圧を調節する、請求の範囲第1項に記載のポリ マー合成装置。 ーを含んでいて、該それぞれのノズルと該ウエルとの運動性を高め、かつ該選択 された試薬の投入用の該それぞれのノズルと該ウエルとを整合させる、請求の範 囲第54項に記載のポリマー合成装置。 56.液状試薬中に連続的にポリマー構成単位を添加することにより、ポリマー鎖 を形成するためのポリマー合成装置であって、 フレームアセンブリーと、 該フレームアセンブリーに運動可能に搭載され、複数の隔置されたウエル行 および列として配列された反応ウエルのアレイを含むベースアセンブリーと、こ こで該各ウエルは、その中に延びた少なくとも一つの各オリフィスを有する、 該フレームアセンブリーに搭載され、かつ整合のために、該ウエルの行と列 との間の間隔に実質的に等しい距離だけ隔置された、複数のノズル行および列状 に配列されたノズルのアレイを有するヘッドアセンブリーと、ここで各ノズルは 、制御された放出のために、液状試薬のタンクと結合されている、 該ベースアセンブリーに結合された輸送機構と、ここで該機構は、該ノズル 列に実質的に平行な通路に沿って、該ヘッドアセンブリーに相対的に支持体を移 動させ、反応ウエルの少なくとも一つの行を、ノズルの選択された行との投入配 列に位置させて、ポリマー鎖の合成のために、液状試薬を選択された反応ウエル に同時に投入する、 該ヘッドアセンブリーと該ベースアセンブリーとの間に配置され、かつ内部 に該反応ウエル全ておよび該ノズル全てを収容して、共通のチャンバーを形成す る滑動封止と、 該ウエルオリフィス各々は、該選択されたウエル中に該選択された液状試薬 を維持するための毛管液体封止を形成するサイズと寸法とをもつものであって、 該チャンバー内の第一のガス圧と、各オリフィス出口にかかる第二のガス圧との 間の差圧が予め定められた値未満である場合に、該ウエル内でのポリマー鎖の成 長を可能とし、 該差圧が該予め定められた値を越えた場合に、該液状試薬が各オリフィスを 通して各ウエルから追い出されるように、該差圧を制御するための圧力調節デバ イスと、 を含むことを特徴とする上記ポリマー合成装置。 57.該滑動封止が、該ヘッドアセンブリーの底部表面に固定された一端と、該ベ ースアセンブリーの上部表面と滑動接触状態にある対向端部とを有する、気球封 止により与えられる、請求の範囲第56項に記載のポリマー合成装置。 58.該輸送機構が該ベースアセンブリーに動作可能に接続されていて、該通路に 沿った該反応ウエルの直線状の運動性を増大する、請求の範囲第57項に記載のポ リマー合成装置。 59.各ノズル行のノズル各々が、該複数の独立した試薬タンクの一つの試薬タン クに接続されている、請求の範囲第58項に記載のポリマー合成装置。 60.各ノズルが、通路を形成し、かつ該ノズルに結合した一端と、該試薬タンク 内で終端している対向端部とを有するチューブを介して、該各試薬タンクと連通 しており、かつ 各チューブが、該各通路と直列状のバルブアセンブリーを含んでいて、該チ ューブを通る該試薬溶液の流動を制御する、請求の範囲第59項に記載のポリマー 合成装置。 61.該圧力調節デバイスが、該ノズルアレイの上流側に配置された、該共通のチ ャンバーへの入口と、該ノズルアレイの下流側に配置された該共通チャンバーか らの出口と、該入口に連結された加圧ガス源とを含み、該加圧ガス源が、該ガス 源からのガスを、該チャンバーの上流側から該チャンバーの下流側までの該共通 チャンバーを通って、該出口から出ていくように連続的に流動させ、該試薬によ 63.少なくとも一つの固体担体に、該担体上でポリマー鎖を成長させかつこれを 固定化するために、連続的にポリマー構成単位を添加することによりポリマー鎖 を形成するための合成法であって、 A) 少なくとも一つの固体担体および該固体担体に固定された該ポリマー鎖の 少なくとも1種のポリマー構成単位と接触状態で、反応ウエル中に液状試薬を投 入する工程と、ここで該ウエルは該ウエル内に延びた少なくとも一つのオリフィ スを有し、かつ該ウエル中に該試薬溶液を維持するための毛管液体封止を形成す るサイズおよび寸法を有していて、該固体担体上でのポリマー鎖の成長を可能と する、 B) 該反応ウエルに第一のガス圧を印加する工程と、ここで該第一のガス圧と 該オリフィスの出口に及ぼされる第二のガス圧との間の差圧は、該毛管液体封止 を凌駕し、かつ該オリフィスを通して該ウエルから該試薬溶液を追い出すのに必 要とされる予め定められた値を越える、 を含むことを特徴とする、上記合成方法。 64.更に、圧力調節デバイスを通して該差圧を調節して、該差圧を該予め定めら れた値以上に上げるか、あるいは該差圧を該予め定められた値以下に下げる工程 をも含む、請求の範囲第63項に記載のポリマー鎖を形成するための合成方法。 65.更に該工程AおよびBを繰り返して、ポリマー構成単位の少なくとも一つの 連続した連鎖を形成する、請求の範囲第63項に記載のポリマー鎖を形成するため の合成方法。 66.該予め定められた値が約1/100psi〜約1/10psiの範囲にある、請求の範囲第6 3項に記載のポリマー鎖を形成するための合成方法。 67.ポリマー構成単位を合成装置に連続的に添加することにより、該装置内でポ リマー鎖を合成する方法であって、該合成装置が、各々制御された放出を行うた めに液状試薬のタンクに連結され、一般的に隔置された関係で搭載された複数の ノズルをもつヘッドアセンブリーと、少なくとも一つの反応ウエルをもつベース アセンブリーと、該ヘッドアセンブリーと該ベースアセンブリーとの間に配置さ れて、これらの間の相対的な運動を可能とし、かつ該反応ウエルおよび該ノズル 両者を内部に収容して共通のチャンバーを形成する滑動封止とを含み、かつ該方 法が以下の諸工程: A) 該反応ウエルと選択された一つのノズルとを、輸送機構を介して整合させ る工程と、ここで該輸送機構は該ヘッドアセンブリーおよび該ベースアセン ブリーの少なくとも一方と連結されて、該ウエルと該ノズルとの間の相対的な運 動を生じる、 B) 該試薬タンクから、該一つのノズルを介して、該ウエルに液状試薬を投入 して、ポリマー鎖の合成を可能とする工程と、 C) 該共通のチャンバーから、該共通チャンバーへの入口に結合され、かつ該 ノズルの上流側に位置する加圧ガス源からのガス通路を介して、該試薬から放出 される有害な煙霧を掃気し、かつ該ノズルの下流側に位置する該共通のチャンバ ーからの出口から追い出す工程、 を含むことを特徴とする上記ポリマー鎖の合成法。 68.更に、以下の工程: D) 該共通のチャンバーに第一のガス圧を印加することにより、該ウエル内に 延びたオリフィスを通して、該ウエルから、該投入された液状試薬を追い出す工 程、 をも含み、ここで該第一のガス圧と、該オリフィスの出口に及ぼされる第二の ガス圧との間の差圧は、該ウエル内に該液状試薬を保持するために、該液状試薬 とオリフィスとの間に形成された、毛管液体封止を凌駕するのに必要とされる予 め定められた値を越える、請求の範囲第67項に記載のポリマー鎖の合成法。 69.更に、上記工程A〜Dを繰り返して、ポリマー構成単位の少なくとも一つの 連続する連鎖を形成する工程を含む、請求の範囲第68項に記載のポリマー鎖の合 成法。 70.該追い出し工程が、第一のガス圧を該予め定められた値を越える程度まで高 めることによって、該チャンバーの出口を介する該ガスの流出を制御することに より達成される、請求の範囲第68項に記載のポリマー鎖の合成法。 71.該制御工程を、該チャンバーの出口に連結されていて、該ガスの通過を制御 するチャンバーバルブによって達成する、請求の範囲第70項に記載のポリマー鎖 の合成法。
