Claims (1)
- 【特許請求の範囲】 1.メトプロロール(metoprolol)またはその塩の持続放出性経口固体剤形であっ て、 ヒト患者に治療効果を付与するのに必要な量のメトプロロールまたは製剤学 的に許容しうるその塩、および 次の成分を含む持続放出性マトリックス: ヘテロ多糖ガム約8〜約35%; 該製剤が環境中の液体にさらされるときにゲル強度を高めるための、該 ヘテロ多糖ガムと架橋することができる陽イオン架橋剤約0.5〜20%;およ び 不活性の製剤学的希釈剤; を含んでなり、メトプロロール対ヘテロ多糖ガムの比が約1:1から約1:5 であり、該製剤が環境中の液体にさらされるときに少なくとも約24時間のメト プロロールの持続放出をもたらす、上記の剤形。 2.薬物対ガムの比が約1:1.5から約1:4である、請求項1に記載の経口 固体剤形。 3.陽イオン架橋剤がアルカリ金属またはアルカリ土類金属の硫酸塩、塩化物、 ホウ酸塩、臭化物、クエン酸塩、酢酸塩または乳酸塩である、請求項1または2 に記載の経口固体剤形。 4.陽イオン架橋剤が硫酸カルシウムを含む、請求項1〜3のいずれか1つに記 載の経口固体剤形。 5.不活性の製剤学的希釈剤が単糖、二糖、多価アルコール、微結晶質セルロー ス、デンプンおよびその混合物から成る群から選ば れる、請求項1に記載の経口固体剤形。 6.不活性希釈剤が前記剤形に配合される前に直接圧縮可能である、請求項1に 記載の経口固体剤形。 7.不活性希釈剤がスクロース、デキストロース、ラクトース、微結晶質セルロ ース、フルクトース、キシリトール、ソルビトール、デンプンおよびその混合物 から成る群から選ばれる、請求項5に記載の経口固体剤形。 8.不活性希釈剤が圧縮可能な微結晶質セルロース、デキストレート、直接圧縮 可能な糖、無水直接打錠用ラクトース、粉末セルロース、噴霧乾燥ラクトース、 凝集マルトデキストリン、直接圧縮ソルビトール、クロスポビドン、デンプング リコール酸ナトリウム、カルボキシメチルデンプン、セルロースフロック(cellu losefloc)、予備糊化デンプンおよびその組合せから成る群から選ばれる、請求 項6に記載の経口固体剤形。 9.陽イオン架橋剤が前記製剤の約10重量%を占める、請求項1〜8のいずれ か1つに記載の経口固体剤形。 10.アルキルセルロース、アクリル酸およびメタクリル酸のエステルのコポリマ ーおよび前記のものの混合物から成る群から選ばれる疎水性ポリマーをさらに含 み、前記薬剤を配合する前に、該疎水性ポリマーが環境中の液体にさらされると きに前記ガムの水和を遅らせるのに有効な量で前記剤形中に配合される、請求項 1〜8のいずれか1つに記載の経口固体剤形。 11.疎水性ポリマーがエチルセルロースを含む、請求項10に記載の経口固体剤形 。 12.疎水性物質が前記剤形の約1〜20重量%を占める、請求項10 または11に記載の経口固体剤形。 13.疎水性ポリマーが前記剤形の約5〜10重量%を占める、請求項10〜12のい ずれか1つに記載の経口固体剤形。 14.錠剤である、請求項1〜13のいずれか1つに記載の経口固体剤形。 15.50mg、100mgまたは200mgのメトプロロールを含む、請求項1〜14のいずれか 一つに記載の経口固体剤形。 16.持続放出性マトリックスが凝集粒子を含有する、請求項1〜15のいずれか1 つに記載の経口固体剤形。 17.メトプロロールの24時間製剤を製造する方法であって、 約8〜約35%のヘテロ多糖ガム;該ガムが環境中の液体にさらされるとき にゲル強度を高めるための、該ヘテロ多糖ガムと架橋することができる陽イオン 架橋剤約1〜20%;および不活性の製剤学的希釈剤;を含む持続放出性マトリ ックスを調製し、 該持続放出性マトリックスとメトプロロールまたは製剤学的に許容しうる塩 とを薬剤対ガム比が約1:1から約1:5となるように組み合わせ、そして 得られた混合物を打錠して、各錠剤が少なくとも約24時間の治療効果を付 与するのに十分な用量のメトプロロールを含むようにする、 ことを含んでなる方法。 18.メトプロロールの24時間製剤を製造する方法であって、 約8〜約35%のヘテロ多糖ガム;該ガムが環境中の液体にさらされるとき にゲル強度を高めるための、該ヘテロ多糖ガムと架橋することができる陽イオン 架橋剤約1〜20%;および不活性 の製剤学的希釈剤;を含む、湿式顆粒化によって粒状化した持続放出性マトリッ クスを調製し、 該持続放出性マトリックスとメトプロロールまたは製剤学的に許容しうる塩 とを薬剤対ガム比が約1:1から約1:5となるように組み合わせ、そして 得られた混合物を打錠して、各錠剤が少なくとも約24時間の治療効果を付 与するのに十分な用量のメトプロロールを含むようにする、 ことを含んでなる方法。 19.メトプロロールを用いて患者を治療する方法であって、 約8〜約35%のヘテロ多糖ガム;該ガムが環境中の液体にさらされるとき にゲル強度を高めるための、該ヘテロ多糖ガムと架橋することができる陽イオン 架橋剤約1〜20%;および不活性の製剤学的希釈剤;を含む持続放出性マトリ ックスを調製し、 該持続放出性賦形剤とメトプロロールまたは製剤学的に許容しうる塩とを薬 剤対ガム比が約1:1から約1:5となるように組み合わせ、 得られた混合物を打錠して、各錠剤が少なくとも約24時間の治療効果を付 与するのに十分な用量のメトプロロールを含むようにし、そして 該錠剤を適当な投与間隔で患者に投与する、 ことを含んでなる方法。 20.打錠するに先立って、持続放出性マトリックスとメトプロロールの前記混合 物に、疎水性物質を、環境中の液体にさらされるときに前記ガムの水和を遅らせ るのに有効な量で添加することをさ らに含む、請求項17〜19のいずれか1つに記載の方法。 21.約1:1.5から1:4の薬剤対ガム比を有する錠剤を提供することをさら に含む、請求項17〜19のいずれか1つに記載の方法。 22.陽イオン架橋剤がアルカリ金属またはアルカリ土類金属の硫酸塩、塩化物、 ホウ酸塩、臭化物、クエン酸塩、酢酸塩または乳酸塩を含み、疎水性ポリマーが アルキルセルロース、アクリル酸およびメタクリル酸のエステルのコポリマーお よび前記のものの混合物から成る群から選ばれる、請求項17〜19のいずれか1つ に記載の方法。 23.疎水性ポリマーがエチルセルロースを含み、そして陽イオン架橋剤が硫酸カ ルシウムを含む、請求項20に記載の方法。 24.疎水性物質が前記製剤の約1〜10重量%を占める、請求項23に記載の方法 。 25.粒状化した持続放出性マトリックスをメトプロロールとともに湿式顆粒化す る工程をさらに含む、請求項17〜19のいずれか1つに記載の方法。 26.ヘテロ多糖ガムと陽イオン架橋剤を湿式顆粒化して凝集塊を形成し、そして 不活性の製剤学的希釈剤を添加する、請求項17〜19のいずれか1つに記載の方法 。 27.ヘテロ多糖ガム、陽イオン架橋剤および不活性の製剤学的希釈剤を湿式顆粒 化して、前記の粒状化持続放出性マトリックスを形成する、請求項17〜19のいず れか1つに記載の方法。 28.メトプロロールまたはその塩の持続放出性経口固体剤形であって、 ヒト患者に治療効果を付与するのに必要な量のメトプロロール または製剤学的に許容しうるその塩; 約25〜35%のヘテロ多糖ガム;および 不活性の製剤学的希釈剤; を含んでなり、メトプロロール対ヘテロ多糖ガムの比が約1:1から約1:5 であり、該剤形が環境中の液体にさらされるときに少なくとも約24時間のメト プロロールの持続放出をもたらす、上記の剤形。 29.不活性の製剤学的希釈剤が単糖、二糖、多価アルコール、デンプン、微結晶 質セルロースおよびその混合物から成る群から選ばれる、請求項28に記載の持続 放出性経口固体剤形。
