Claims (17)
- 1.一種又は数種の官能基を付与した担体から始めて、与えられたアミノ酸配列 に応じて逐次的に所要のペプチドを適当な合成方法によって合成する、担体に結 合した又は遊離のペプチド類を迅速に合成する方法に於いて、−担体として偏平 材料を使用し、 −カップリングのために、活性化した誘導体の形の所要のアミノ酸又は所要のジ ペプチド又はオリゴペプチドの溶液の少量のアリコートを平面状担体の上に点状 又は線状に塗布し、 −濡れることにより生成した反応面の中でカップリング反応を実施し、−必要に 応じて生成したペプチドを適当な分離剤を用いて平面状担体から分離することを 特徴とする前記方法。
- 2.数種乃至非常に多種の異なるペプチドを一つの偏平材料の上で同時に並べて 、相互に接触しない点状又は線状の反応平面上で合成することを特徴とする請求 項1の方法。
- 3.偏平材料としてアミノ酸又はペプチドのカップリングに適した化学反応性官 能基を有する多孔性又は非多孔性フィルム、メンブラン、又はそのような紙を使 用することを特徴とする請求項1又は2の方法。
- 4.前記偏平材料がセルロース、ガラス又は有機のポリマーから成り、その表面 が適した化学的官能基を備えていることを特徴とする請求項3の方法。
- 5.前記表面への官能基の付与をアミノプロピル基、グリシン又はジグリシンの アンカー基の導入によって実施することを特徴とする請求項3又は4の方法。
- 6.合成しようとするペプチドを、(a)C末端を直接前記偏平材料と結合する か或いは(b)前記偏平材料と一種のリンカーを介して結合して構成することを 特徴とする請求項4又は5の方法。
- 7.反応性官能基の面積当たりの付着量が1pmol〜1μmol/cm2であ ることを特徴とする前記請求項の何れか1項の方法。
- 8.一種又は数種の官能基を付与した担体から始めて、与えられたヌクレオチド 配列に応じて逐次的に所要のオリゴヌクレオチドを適当な合成方法によって合成 する、担体に結合した又は遊離のオリゴヌクレオチド類を迅速に合成する方法に 於いて、 −担体として偏平材料を使用し、 −カップリングのために、活性化した誘導体の形の所要のヌクレオチド又は所要 のジヌクレオチド又はオリゴヌクレオチドの溶液の少量のアリコートを平面状担 体の上に点状又は線状に塗布し、 −濡れることにより生成した反応面の中でカップリング反応を実施し、−必要に 応じて生成したオリゴヌクレオチドを適当な分離剤を用いて平面状担体から分離 する ことを特徴とする前記方法。
- 9.数種乃至非常に多種の異なるオリゴヌクレオチドを一つの偏平材料の上で同 時に並べて、相互に接触しない点状又は線状の反応平面上で合成することを特徴 とする請求項8の方法。
- 10.偏平材料としてヌクレオチドのカップリングに適した化学反応性官能基を 有する多孔性又は非多孔性フィルム、メンブラン、又はそのような紙を使用する ことを特徴とする請求項8又は9の方法。
- 11.請求項4、5又は7の対策を使用することを特徴とする請求項8、9又は 10の方法。
- 12.合成しようとするオリゴヌクレオチドを、(a)3′末端を直接前記偏平 材料と結合するか或いは(b)前記偏平材料と一種のリンカーを介して結合して 構成することを特徴とする請求項11の方法。
- 13.平面状担体を備えた1個の自動ピペット操作装置を特徴とする請求項1又 は8の方法を実施するための装置。
- 14.請求項1乃至7の何れか1項により調製されたペプチド類を結合した偏平 材料。
- 15.請求項8乃至12の何れか1項により調製されたオリゴヌクレオチド類を 結合した偏平材料。
- 16.微生物学的目的、特に人間医学又は獣医学に於いて細菌又はウイルスの感 染を特異的に検出するための診断の目的への請求項14の偏平材料の使用。
- 17.雑種形成の目的への請求項15の偏平材料の使用。
