Description

【発明の詳細な説明】[Detailed description of the invention]

本発明はミツバチによつて植物から収集し、生
産されるヤニ状物質のプロポリス(蜂ヤニ:
Propolis)の親水性溶媒または水との混液抽出物
を固型分として0.01〜1重量%含有する抗アクネ
または抗フケ剤に関する。 プロポリスは古くから伝承的民間薬として利用
されており、近年東欧諸国を中心にその機能、作
用が解明されつつある。抗菌作用、止血作用、瘢
痕化作用、抗炎症作用、抗酸化作用、局所麻酔作
用、腫瘍増殖抑制作用等について多くの報告があ
る。プロポリスの成分はフラボン類、フラバノン
類、芳香族アルコール、芳香族アルデヒド、クマ
リン等が主なもので、これらの組成、含有量等は
プロポリスの産地によつて異なり、作用も差異が
あるといわれている。 プロポリスの抗菌作用についてはブドウ球菌、
枯草菌、カビ、酵母等に対する抗菌作用が報告さ
れているが、フケ、カユミ及びアクネの各々の原
因菌に対する報告は見られない。そこで本発明者
らはプロポリスの抗菌作用に着目し幅広い実験を
組んで鋭意研究を重ねた結果、プロポリスが、頭
皮のフケ、カユミの原因菌とされる頭皮常在酵母
pityrosprum ovale及びアクネ(尋常性座瘡:
Acnes Valganis)の原因菌とされる
propionibacterium acnesに対し有効な抗菌力を
示すことを見き出した。 すなわち、本発明は、プロポリスの親水性溶媒
または水との混液抽出物を固型分として0.01〜1
重量%含有することを特徴とする抗アクネまたは
抗フケ剤である。 以下、本発明について詳述する。 本発明で用いられるプロポリス(propolis)は
前述の通り、ミツバチによつて植物から収集され
るヤニ状の物質であり、蜂ヤニとも呼ばれてい
る。 一般的には養蜂の際に巣箱の巣枠やすき間に多
量に付着するのでこれをかき取つて採集される。 上記プロポリスからの抽出液又は抽出物は次の
様にして製造される。 プロポリスの親水性溶媒または水との混液に浸
漬して抽出し(一般的にはプロポリスの10重量倍
程度の抽出液を用いる)、プロポリス抽出液を得
る。親水性溶媒としては、例えばメタノール、エ
タノール等のアルコール及びアセトンが挙げられ
る。 プロポリス抽出液から蒸留その他の方法で溶媒
を留去することにより、プロポリス抽出物を得
る。 抗アクネまたは抗フケ剤の剤型はどのようなも
のでもよく、例示すれば可溶化系、乳化系、透明
溶解系、粉末分散系、粉末系、水相〜油相混合系
等をあげることができる。抗アクネまたは抗フケ
剤の用途もどのようなものでもよく、好ましくは
化粧水、乳液、クリーム、パツク、化粧用油、ヘ
アトニツク、ヘアクリーム、ヘアリキツド、シヤ
ンプー、リンス等の皮膚化粧料及び頭髪化粧料で
あるが、当然のことながら、口紅、フアンデーシ
ヨン、アイシヤドウの如きメイクアツプ化粧料、
ボデイパウダー、オーデコロン等のボデイ化粧
料、芳香化粧料等の他の用途の化粧料も全て対象
となる。 又、本発明の上記プロポリスの抽出物に併せて
既存のフケ.カユミ防止剤、抗アクネ剤、美白
剤、育毛剤を本発明の効果を損なわない範囲で併
用することができる。 上記プロポリスの親水性溶媒または水との混液
抽出物の抗アクネまたは抗フケ剤への配合量は任
意であるが通常、抽出液の場合は0.1〜10重量%
(抗アクネまたは抗フケ剤全量中の)、抽出物の場
合は、固型分で0.01〜1重量%(上記に同じ)の
範囲である。 次に実施例によつて本発明をさらに詳細に説明
する。 〔実施例 1〕フケ・カユミ防止作用 (1) 頭皮常在酵母Pityrosporum ovale及び皮膚
常在菌に対する抗菌力測定 頭皮に常在するPityrosporum ovaleは頭皮の
脂質を分解し脂肪酸を生成する。この脂肪酸は頭
皮に紅斑やカユミを与える。さらにこれに対する
生体の防御反応として皮膚のターンオーバーの加
速がはじまり、この結果、表皮角化層の剥離堆積
をきたしフケを発生する。これらのことから
Pityrosporum ovaleの活性を阻止又は弱力化す
れば、フケ、カユミを防止することができる。 そこでPityrosporum ovaleに対するプロポリ
スの抗菌力を検討した。 培地(ポテトエキス2%、酵母エキス1%、ペ
プトン1%、オリーブ油3%)を100ml培養フラ
スコに入れ、更に最終濃度で設定濃度となるよう
に調整したプロポリス抽出物のエタノール溶液を
1ml加え加熱処理をした。これに、人頭皮より分
離したPityrosprum ovaleを接種し、37℃で20日
間振盪培養を行い培養終了時の乾燥菌体量を測定
し生育の有無を判定した。 この結果、Pityrosporum ovaleに対するプロ
ポリス抽出物の抗菌作用は最少発育阻止濃度
(MIC)が32ppmであることが認められた。 さらに、本発明者等は一般皮膚常在菌2種
(Staphyrococcus aureus,
Staphyrococcusepidermidis)に対する抗菌力を
測定したところ、何れに対してもMIC=16ppm
であることが認められた。 (2) ウサギの耳翼を用いたフケ防止効果測定 (1)の結果をもとに、動物を用いたフケ防止効果
テストを行なつた。 ウサギの左側の耳翼にプロポリス抽出物の0.1
%エタノール溶液を、右耳翼にはコントロールと
してエタノールを塗布し、約10分後に両耳翼にオ
レイン酸を塗布した。この操作を1日1回、のべ
4〜6日間行ない、両耳翼の紅斑と落屑を観察し
た。陽性コントロールにはジンクピリチオン
(ZPT)1%を用い同時に行なつた。 結果を表−1に示す。

せたGAM寒天培地にこれを接着させ、37℃、7
日間嫌気培養を行ない、培養終了時、発育阻止帯
