Description

【発明の詳細な説明】 〔産業上の利用分野〕 本発明は、Mucor pusiLLusが産生するの
凝乳酵素であるムコールレンニン(以下、MPRという
)遺伝子とプラスミノーゲンアクチヘータ−(以下、P
Aという)遺伝子との融合遺伝子、該融合遺伝子が挿入
されたプラスミド、該プラスミドにより形質転換された
酵母、該酵母によるPAの製造に関する。
さらに詳しくは、遺伝子レヘルにおいてMPRのプレま
たはプレプロ配列と様々なタイプのPAとを連結させた
融合遺伝子を、組み換えDNAの技術を応用して酵母に
て発現させることからなる様々なタイプのPAの効率良
い製造方法を提供する一連の技術に関する。
(従来の技術・発明が解決しようとする課題〕線維素溶
解に係わるPAは血栓溶解治療薬として広く用いられて
おり、その内ヒト由来のPAとしては、ウロキナーゼ(
以下、UKという)と血(aloa+  p/、mp 
 1ap)〔θ〕 (Jou+ p/lu+p 2ap) 〔θ〕 巳 一見区生 継 迅 蛎 匹 く ″  S ε −目     a 寸 犠 郷 − ♂

Claims (6)

    【特許請求の範囲】
  1. (1)ムコールレンニン遺伝子のプレ配列とプラスミノ
    ーゲンアクチベーター遺伝子とが連結されてなる融合遺
    伝子。
  2. (2)前記ムコールレンニン遺伝子のプレプロ領域の内
    部または下流に、前記プラスミノーゲンアクチベーター
    遺伝子が連結されてなる請求項(1)記載の融合遺伝子
  3. (3)請求項(1)または(2)記載の融合遺伝子が挿
    入されてなるプラスミド。
  4. (4)プロモーターとして酵母GAL7プロモーターを
    有する請求項(3)記載のプラスミド。
  5. (5)請求項(3)または(4)記載のプラスミドによ
    り形質転換された酵母。
  6. (6)請求項(5)記載の酵母を発現させることを特徴
    とするプラスミノーゲンアクチベーターの製造方法。
