Claims (10)
- 免疫療法生産物を発生させるための方法であって、該方法は、
第1の細胞培養チャンバーの中で末梢血単核細胞および樹状細胞を培養して、T細胞を含む上清を作り出すステップと、
前記T細胞を含む前記上清を前記第1の細胞培養チャンバーから流体コネクターを通して第2の細胞培養チャンバーの中へ移送するステップと、
を含み、
前記第1の細胞培養チャンバーは、
細胞が付着する第1の材料から構成された底部表面と、
ガス透過性の第2の材料から構成されている、少なくとも1つの追加的な表面と、
前記底部表面に対して実質的に平行で対称のインフローとして前記第1の細胞培養チャンバー内に1つまたは複数の流体を入れるように方向付けるように配置された少なくとも2つの入口部と、
前記底部表面に対して実質的に垂直のアウトフローとして、前記第1の細胞培養チャンバーから1つまたは複数の流体を方向付けるように配置されている、1つまたは複数の出口部と、
を含む、方法。 - 前記第2の細胞培養チャンバーは新鮮な樹状細胞を含み、前記方法は、前記T細胞をさらに培養するステップをさらに含む、請求項1に記載の方法。
- 前記第1の細胞培養チャンバーの中での培養は、第1のセットの刺激抗原の存在下で起こり、前記第2の細胞培養チャンバーの中での培養は、第2のセットの刺激抗原の存在下で起こる、請求項1に記載の方法。
- 前記第1のセットの刺激抗原と前記第2のセットの刺激抗原とが同じである、請求項3に記載の方法。
- 前記第1のセットの刺激抗原と前記第2のセットの刺激抗原とが異なっている、請求項3に記載の方法。
- 1つまたは複数のセンサーを介して、少なくとも前記第1のまたは第2の細胞培養チャンバーの中の1つまたは複数のパラメーターを測定するステップをさらに含む、請求項1に記載の方法。
- 前記1つまたは複数のパラメーターは、pH、溶存酸素、合計バイオマス、細胞直径、グルコース濃度、乳酸濃度、および細胞代謝産物濃度からなる群から選択される、請求項6に記載の方法。
- 垂直方向の流路に沿った前記垂直のアウトフローの移動は、流体の流量が前記第1の細胞培養チャンバーの中の細胞の沈殿レートに打ち勝つには不十分とされている、請求項1に記載の方法。
- 前記第2の細胞培養チャンバーの中の前記T細胞をさらに培養し、増殖させるステップをさらに含む、請求項1に記載の方法。
- さらに培養され、増殖された前記T細胞を、前記流体コネクターを通して第3の細胞培養チャンバーに移送するステップをさらに含む、請求項9に記載の方法。
