本明細書に記載のオリゴヌクレオチドアレイは、種々の増幅技術において使用され得る。使用することができる例示的な技術は、ポリメラーゼ連鎖反応(PCR)、ローリングサークル増幅(RCA)、多重置換増幅(MDA)、又はランダムプライム増幅(RPA)、又はこれらの組み合わせを含有するが、これらに限定されない。特定の例において、増幅に使用される1つ以上のプライマーは、基材上のコポリマー被覆に付着される。2種の付着プライマーを利用するフォーマットは、二本鎖アンプリコンが、コピーされた鋳型配列にそって並ぶ2つの付着プライマーの間のブリッジ様構造を形成するので、ブリッジ増幅を可能にする。増幅は、コポリマー被覆に付着した一つの増幅プライマー及び溶液中の第二のプライマー(例えば、エマルジョンPCR)、又は溶液中の両方のプライマーを用いて行うこともできる。

Claims (27)
- 前記窒素含有複素環が、
(i)6員環を含み、任意に、A及びEが6員環の2位と5位において結合し、若しくは、5員環を含み;及び/又は
(ii)芳香族もしくは飽和である、
請求項1又は2に記載の化合物。 - 請求項6又は7に記載のコポリマーと基材との反応から形成される基材-コポリマー生成物であって、任意に、前記コポリマーのアジド基と、前記基材の表面上のアルケニル-又はアルキニル-含有基との反応によって形成される、基材-コポリマー生成物。
- 基材-結合コポリマーが、基材に共有結合している繰り返しモノマー単位を含み、繰り返し、共有-結合モノマー単位が式(IV)の化合物である、基材に共有結合したコポリマーを含む構造。
R1はH又はC1-4アルキルであり;
R2はH又はC1-4アルキルであり;
Lは、炭素、酸素、及び窒素からなる群から選択される2から20個の原子から成る直鎖、及び前記鎖において炭素及びいずれかの窒素原子上に任意の置換基を含むリンカーであり;
Eは、任意に置換されたC1-4アルキレンであり;
Aは以下の式を有するN置換アミドであり、
前記トリアゾール又はトリアゾリンは、一方又は両方のトリアゾール/トリアゾリン環の炭素原子を介して直接又はリンカーを介して間接的に、基材に共有結合する。) - 請求項5から7のいずれか一項に記載のコポリマーと1つ以上のオリゴヌクレオチドとの間の反応から形成されるコポリマー-オリゴヌクレオチド生成物であって、任意に、前記コポリマー-オリゴヌクレオチドが前記コポリマーのアジド基と、アルケン-又はアルキン-官能基化オリゴヌクレオチドの反応から形成される、コポリマー-オリゴヌクレオチド生成物。
- 式(IV)のオリゴヌクレオチド-結合繰り返しモノマー単位を含む、オリゴヌクレオチド結合コポリマー。
R1はH又はC1-4アルキルであり;
R2はH又はC1-4アルキルであり;
Lは、炭素、酸素、及び窒素からなる群から選択される2から20個の原子から成る直鎖、及び前記鎖において炭素及びいずれかの窒素原子上に任意の置換基を含むリンカーであり;
Eは、任意に置換されたC1-4アルキレンであり:
Aは以下の式を有するN置換アミドであり、
Zは窒素含有複素環であり;
- 前記オリゴヌクレオチド結合繰り返しモノマー単位が式(IVa)である、請求項13に記載のオリゴヌクレオチド結合コポリマー。
任意に、前記オリゴヌクレオチド-結合繰り返しモノマー単位が式(VI)であり、
さらに任意に、前記オリゴヌクレオチド-結合繰り返しモノマー単位が式(VIa)であり、
前記オリゴヌクレオチド-結合繰り返しモノマー単位が式(IVb)、式(IVc)、式(IVd)、又は式(IVe)である、請求項13に記載のオリゴヌクレオチド結合コポリマー。
- 請求項5から7のいずれか一項に記載のコポリマーと、基材及び一つ以上のオリゴヌクレオチドとの反応によって形成されるコポリマー-基材-オリゴヌクレオチド生成物。
- 前記コポリマーのアジド基を、前記基材の表面上の複数のアルケニル-又はアルキニル-含有官能基と反応させて、前記コポリマーと前記表面との間に共有結合を形成することを含む、請求項5から7のいずれか一項に記載のコポリマーを基材に固定化する方法。
- 前記コポリマーのアジド基を前記複数のオリゴヌクレオチドと反応させることを含み、
任意に、前記複数のオリゴヌクレオチドがアルケニル又はアルキニル基を含み、前記反応が前記コポリマーの前記アジド基と前記オリゴヌクレオチドの前記アルケニル又は前記アルキニル基との間で起こる、請求項5から7のいずれか一項に記載のコポリマーに複数のオリゴヌクレオチドをグラフト化する方法。 - 式(IIb)、式(IIc)、式(IId)、又は式(IIe)の繰り返しモノマー単位及び少なくとも1つの他の繰り返しモノマー単位を含み、
R1はH又はC1-4アルキルであり;
R2はH又はC1-4アルキルであり;
R3はH又はC1-4アルキルであり;
XはCH又はNであり;
pは1、2、3又は4であり;
式(IIb)におけるLは、炭素、酸素、及び窒素からなる群から選択される2から20個の原子からなる直鎖、及び前記鎖において炭素及び窒素原子上に任意の置換基をもつリンカーであり;及び
式(IIc)におけるnは2から20までの整数である。);
任意に、式(IIb)、(IIc)、(IId)又は(IIe)の繰り返しモノマー単位及び式(III)の繰り返しモノマー単位を含み、

任意に、
(i)共有付着が、請求項6に規定される式(IIb)、式(IIc)、式(IId)又は式(IIe)のモノマー単位のアジド基と、基材の表面上のアルケニル-又はアルキニル-含有基との反応によって作られ;及び/又は
(ii)前記基材-結合コポリマーが、基材に共有結合している繰り返しモノマー単位をさらに含み、ここで、前記繰り返し、共有-結合モノマー単位が、式(IVb)、式(IVc)、式(IVd)、又は式(IVe)によって表され、

さらに任意に、前記オリゴヌクレオチド-結合コポリマーが、式(IVb)、(IVc)、(IVd)又は(IVe)の繰り返しモノマー単位を含み、式中、R 1 はH又はC 1-4 アルキルであり;R 2 はH又はC 1-4 アルキルであり;R 3 はH又はC 1-4 アルキルであり;Lは、炭素、酸素、及び窒素からなる群から選択される2から20個の原子を有する直鎖、及び前記鎖において炭素及びいずれかの窒素原子上に任意の置換基を含むリンカーであり;XはCH又はNであり;Zは窒素含有複素環であり;pは1、2、3又は4であり;nは2から20までの整数であり、

任意に、前記コポリマーが、式(Vb)、式(VIc)、式(VId)、及び式(VIe)の1つ以上の繰り返しモノマー単位を含む、請求項23に記載のコポリマー。 - 請求項6に規定される式(IIb)、(IIc)、(IId)又は(IIe)の繰り返しモノマー単位を含むコポリマーのアジド基を、基材の表面上の複数のアルケニル-又はアルキニル-含有官能基と反応させて、前記コポリマーと前記表面との間に共有結合を形成することを含む、請求項6に規定される式(IIb)、(IIc)、(IId)又は(IIe)の繰り返しモノマー単位を含むコポリマーを基材に固定化する方法。
- コポリマーのアジド基をオリゴヌクレオチドと反応させることを含む、請求項6に規定される式(IIb)、(IIc)、(IId)又は(IIe)の繰り返しモノマー単位を含み、
前記オリゴヌクレオチドがアルケニル又はアルキニル基を含み、反応が前記コポリマーの前記アジド基と前記オリゴヌクレオチドとの間で起こる、
