Description
いくつかの実施形態では、誘導体化キトサンは、湿気にさらされると組織内のシステイン基と反応する。例えば、誘導体化キトサンは、多官能性アルデヒドで誘導体化されたキトサンであり得、誘導体化キトサンは、多官能性エポキシドで誘導体化されたキトサンであり得、誘導体化キトサンは、多官能性アクリレートで誘導体化されたキトサンであり得るか、又は誘導体化キトサンは、ゲニピンで誘導体化されたキトサンであり得る。
上記の態様及び実施形態と併せて使用され得るいくつかの実施形態では、誘導体化キトサンは、とりわけ、不飽和基で誘導体化されたキトサンであり得るか、又は誘導体化キトサンは、チオール基で誘導体化され得る。
いくつかの実施形態では、キトサン塩は、組織又は血液中の負に帯電した種とイオン的に架橋する。
本発明で使用するためのキトサン粉末は、任意の適切な粒径のものであってよい。様々な実施形態において、粒子サイズは、例えば、その他の可能性の中でもとりわけ、1μm未満から1000μmまでの範囲(例えば、1μm~2.5μm、10μmまで、25μmまで、50μmまで、100μmまで、250μmまでの範囲であり得る。これに関して、50μm~425μmの間のサイズの粒子を有するキトサン粉末は、8フレンチのカテーテルを通して分配されたときに良好に機能する。
キトサンは、ランダムに分布した、β-(1-4)-結合のD-グルコサミンとN-アセチル-D-グルコサミンモノマー単位を含む修飾された多糖である。キトサンはキチンのアルカリ性N-脱アセチル化によって商業的に生産され、それは主に修飾グルコース、特にN-アセチル-D-グルコサミンの非分岐鎖からなるセルロース様ポリマーである。
いくつかの実施形態では、キトサン、キトサン塩、修飾キトサン、又はそれらの組み合わせが、組織部位への適用前、又は組織部位への適用時に、非共有結合架橋又は共有結合架橋されているキトサン粉末が使用され得る。
例えば、アミンと反応する多官能性(例えば、二官能性、三官能性など)反応性分子を含む第1の粒子、例えば、ゲニピン、多官能性アルデヒド分子、又は上記のような多官能性エポキシド分子(例えば、PEGジエポキシド、PEGジアルデヒド又は固体である任意の小分子ジアルデヒド又は小分子ジエポキシド)は、キトサン又はキトサン塩粉末と混合され、乾燥形態で組織に適用されてもよい。特定の実施形態において、多官能性反応性分子は、例えば、上記のアルデヒド修飾キトサン(ペンダントアルデヒド基を有するキトサン)、エポキシ修飾キトサン(すなわち、ペンダントを有するキトサン)及びゲニピン修飾キトサン(すなわち、ペンダントゲニピン基を有するキトサン)から選択され得る、上記のものなどの修飾キトサンである。混合された粉末が濡れると(例えば、体液及び/又は液体の適用により)、粉末成分が溶解し、多官能性反応性分子がキトサン又はキトサン塩にあるアミンと架橋し、さらに組織のアミンと反応する。
実施形態A1 組織部位での出血を治療又は予防する方法であって、キトサン粉末を組織部位に適用することを含み、キトサン粉末は、キトサン塩、架橋キトサン、誘導体化キトサン、又はそれらの組み合わせを含むことを要旨とする。
実施形態A3 体腔が胃腸管である、実施形態A2の方法を要旨とする。
実施形態A4 実施形態A1~A3のいずれかの方法であって、キトサン粉末はカテーテルを介して適用されることを要旨とする。
実施形態A6 流動化ガスがCO2である、実施形態A6の方法を要旨とする。
実施形態B1 予め充填されたカテーテルアセンブリであって、内部を貫通する管腔と、近位端と、出口オリフィスを有する遠位端とを有するカテーテルと、キトサン粉末を含むリザーバーとを有し、カテーテルアセンブリは、キトサン粉末を、リザーバーからルーメンを通って、そして出口オリフィスから送達することを要旨とする。
実施形態C3 架橋剤がポリアニオン性架橋剤である、実施形態C2の組成物を要旨とする。
実施形態C5 共有結合架橋剤が多官能性エポキシ、多官能性アルデヒド、及びゲニピンから選択される、実施形態C4の組成物を要旨とする。
実施形態C7 誘導体化ポリマーが、アルデヒド誘導体化ポリマー、エポキシ誘導体化ポリマー、及びゲニピン誘導体化ポリマーから選択される、実施形態C6の組成物を要旨とする。
実施形態C9 誘導体化キトサンが、アルデヒド誘導体化キトサン、エポキシ誘導体化キトサン、及びゲニピン誘導体化キトサンから選択される、実施形態C8の組成物を要旨とする。
実施形態C11 第2の粒子が共有架橋剤を含む、実施形態C10の組成物を要旨とする。
実施形態C13 第1の粒子がチオール修飾キトサンを含み、第2の粒子が複数の不飽和基を有する分子を含む、実施形態C10の組成物を要旨とする。
実施形態D1 組織部位に適用するための粉末組成物であって、多官能性カルボキシル化ポリマーで架橋されたキトサンを含む粉末組成物を要旨とする。
実施形態D3 実施形態D1又はD2の組成物において、キトサンは、ジイミドカップリングを使用して多官能性カルボキシル化ポリマーと架橋されることを要旨とする。
実施形態E2 誘導体化されたキトサンが水分に曝されると組織と反応する、実施形態E1の粉末を要旨とする。
実施形態E4 誘導体化キトサンが、多官能性アルデヒドで誘導体化されたキトサンである、実施形態E2の粉末を要旨とする。
実施形態E6 誘導体化キトサンがゲニピンで誘導体化されたキトサンである、実施形態E2の粉末を要旨とする。
実施形態E8 誘導体化キトサンが不飽和基で誘導体化されたキトサンである、実施形態E7の粉末を要旨とする。
実施形態E10 キトサンが、カルボン酸基及びチオール基を含む化合物で誘導体化されている、実施形態E9の粉末を要旨とする。
(実施例)
シグマ・アルドリッチ社から入手したキトサンは、キトサン2重量%と水98重量%の濃度で水に懸濁される。混合物は、機械ミキサーを使用して室温で撹拌される。次に、撹拌中に酢酸を添加して、5時間撹拌した後、pHが5.0近くになるようにする。(最初に使用したキトサンの重量に対して)2重量%のクエン酸を容器に加え、さらに5時間混合する。このプロセスでゲルが形成され、その後乾燥される。乾燥したゲルは、使用のために細かい粉末に粉砕される。
Claims (4)
- 組織部位の止血を達成するための粉末組成物において、粉末組成物は第1の粒子を含み、前記第1の粒子には第2の粒子が混合され、それにより、組織部位に適用するための粉末組成物を形成し、前記第2の粒子は湿気にさらされると前記第1の粒子と共有結合又は非共有結合で相互作用する架橋剤を含み、前記第1の粒子はキトサン塩を含み、前記架橋剤は、エポキシ誘導体化ポリマー、アクリレート誘導体化ポリマー及びゲニピン誘導体化ポリマーからなる群から選択される誘導体化ポリマーであり、前記エポキシ誘導体化ポリマーはエポキシ誘導体化キトサンであり、前記アクリレート誘導体化ポリマーはアクリレートで誘導体化されたキトサンであり、前記ゲニピン誘導体化ポリマーはゲニピンで誘導体化されたキトサンである、前記粉末組成物。
- 前記第1の粒子は、有機一酸のキトサン塩、または、有機二酸のキトサン塩酸を含む、請求項1に記載の組成物。
- 前記第1の粒子の前記キトサン塩は、酢酸塩である、請求項1に記載の組成物。
- 前記第1の粒子は、キトサンハロゲン化物を含む、請求項1に記載の組成物。
