ここで、Dは直径、Sは面積を表す。
図17及び図18に示すように、半透膜110(旭化成株式会社又はSPECTRUM)を、培養側プレート21及び培地側プレート22で挟み、さらに、半透膜110、培養側プレート21及び培地側プレート22を、培養槽30及び培地保持槽40で挟み込んだ。
実施例1と同様にゲル培地を作製した。ゲル培地にシングルセルにされたiPS細胞を添加した。15mLファルコンチューブ(Falcon)にiPS細胞を含むゲル培地を入れた。その後、ファルコンチューブのキャップを固く締めた。
実施例1と同様に細胞含有培地を調製した。また、図2に示した細胞培養器と同様の細胞培養器を用意した。培養槽30内の空気層が完全になくなるまで、培養槽30内に細胞含有培地を注入した。その後、培養槽30の供給口及び排出口を遮蔽した。また、培地保持槽40、培地流路200、及び培地タンク60をゲル培地で充填した。その後、培地タンク60の導入口及び排出口を遮蔽した。これにより、培養槽30及び培地保持槽40内部を密閉し、培養槽30及び培地保持槽40の内部と外部とで、ガス交換が完全に生じないようにした。
増殖因子を培地(StemSpan H3000、登録商標、STEMCELL Technologies Inc.)に添加し、さらに培地に脱アシル化ゲランガムを添加して、ゲル培地を用意した。
Claims (34)
- 培養成分が透過可能な培養成分透過部材と、
前記培養成分透過部材の一方の面を覆う、細胞含有培地を保持し、細胞を培養するための培養槽と、
前記培養成分透過部材の他方の面を覆う、培地を保持するための培地保持槽と、
前記培地保持槽に接続された培地流路と、
を備える細胞培養器であって、
前記培養槽に細胞を供給するための培養槽供給口と、細胞を排出するための培養槽排出口と、が設けられており、
前記培地流路が、密閉可能な培地流路であって、
前記培地流路内に流体を供給するための流路供給口と、前記培地流路内の流体を排出するための流路排出口と、が、前記培地流路に設けられており、
前記流路供給口及び前記流路排出口が密閉可能であり、
前記培地流路に設けられた、前記流体を移動させるための流体機械と、
前記培地流路及び前記流体機械が配置される培地流路用外気遮断部材と、
を備え、
前記培地流路及び前記流体機械のポンプヘッドが、前記培地流路用外気遮断部材内に配置され、
前記流体機械の前記ポンプヘッドに接続される駆動部が、前記培地流路用外気遮断部材外に配置される、
細胞培養器。 - 前記流路供給口に流体を供給するための供給器が着脱可能である、請求項1に記載の細胞培養器。
- 前記供給器が供給用ニードルレスコネクターを介して前記流路供給口に接続される、請求項2に記載の細胞培養器。
- 前記流路排出口に流体を排出するための排出器が着脱可能である、請求項1から3のいずれか1項に記載の細胞培養器。
- 前記排出器が排出用ニードルレスコネクターを介して前記流路排出口に接続される、請求項4に記載の細胞培養器。
- 前記流路供給口に流体を供給するための供給器が着脱可能であり、
前記流路排出口に流体を排出するための排出器が着脱可能であり、
前記供給器から前記培地流路内に流体を供給すると、前記培地流路内の流体が前記排出器内に移動する、
請求項1に記載の細胞培養器。 - 前記流路供給口に流体を供給するための供給器が着脱可能であり、
前記流路排出口に流体を排出するための排出器が着脱可能であり、
前記流体機械が駆動されると、前記供給器から前記培地流路に流体を供給され、前記培地流路内の流体が前記排出器内に移動する、

- 前記培地流路が密閉された状態で、前記培地流路内に、前記培地流路外の細胞、微生物、ウイルス、及び塵が進入しない、請求項1から11のいずれか1項に記載の細胞培養器。
- 前記培地流路が密閉された状態で、前記培地流路内の物質が、前記培地流路外に流出しない、請求項1から12のいずれか1項に記載の細胞培養器。
- 前記培地流路が密閉された状態で、前記培地流路の内外で気体の交換が生じない、請求項1から13のいずれか1項に記載の細胞培養器。
- 前記培地流路が密閉された状態で、前記培地流路内に、二酸化炭素ガス、窒素ガス、及び酸素ガスの少なくともいずれかが供給されない、請求項1から14のいずれか1項に記載の細胞培養器。
