Description
の化合物または薬学的に許容されるその塩から選択されるCDK阻害剤と、BRAF阻害剤またはMEK阻害剤から選択される少なくとも1種の抗がん剤とを含む組合せ医薬が提供される。
(a)ルイス酸触媒の存在下で、式VIA
(b)塩基および溶媒の存在下で、分割された式VIIAのアセチル化化合物を、式ArCOOHの酸または式ArCOClの酸塩化物または式(ArCO)2Oの酸無水物または式ArCOOCH3のエステル(式中、Arは、式Iの化合物に関連して本明細書で上記に定義された通りである)と反応させることによって、式VIIIA
(c)適切な溶媒中で式VIIIAの分割化合物を塩基で処理することによって、式IXA
の対応する分割されたβ−ジケトン化合物を得るステップと、
(d)式IXAの分割されたβ−ジケトン化合物を、塩酸などの酸で処理することによって、式XA
(e)120〜180℃の範囲の温度で脱アルキル化剤と共にこれを加熱することにより、式XAの化合物を脱アルキル化の対象とすることによって、式Iの化合物の(+)−transエナンチオマーを得、および場合によって、対象化合物をその薬学的に許容される塩に変換するステップと
を含む。

化合物A(ボルシクリブ)および化合物B(リビシクリブ)の調製、式1の代表的な化合物を本明細書で参照例として提示する:
(a)(+)−trans−2−(2−クロロ−4−トリフルオロメチルフェニル)−8−(2−ヒドロキシメチル−1−メチルピロリジン−3−イル)−5,7−ジメトキシ−クロメン−4−オンの調製
窒素雰囲気下、0℃で維持されたn−BuLi(ヘキサン中15%溶液、2.2mL、5mmol)のTHF(10mL)中溶液、に、ヘキサメチルジシラザン(1.08mL、5.1mmol)を滴加し、15min撹拌した。温度を0℃で維持しながら、これに、(+)−trans−2−クロロ−4−トリフルオロメチル安息香酸2−(2−アセトキシメチル−1−メチル−ピロリジン−3−イル)−6−アセチル−3,5−ジメトキシフェニルエステル(1.44g、2.5mmol)のTHF(10mL)中溶液を滴加した。添加後、室温まで温めて反応させ、2.5h撹拌した。反応混合物を希釈HClで酸性化し、10%炭酸水素ナトリウムでpH8〜9に塩基性化した。水層をクロロホルム(3×25mL)で抽出した。有機層を水(25mL)、ブライン(25mL)で洗浄し、無水Na2SO4で脱水した。有機層を減圧下で濃縮し、真空下で乾燥させることによって、油として酢酸3−{3−[3−(2−クロロ−4−トリフルオロメチル−フェニル)−3−オキソ−プロピオニル]−2−ヒドロキシ−4,6−ジメトキシ−フェニル}−1−メチル−ピロリジン−2−イルメチルエステルを生成した(1.3g、90.2%)。このエステルを濃縮HCl(10mL)に溶解し、3h撹拌して環化を生じさせた。3hの終わりに、反応混合物を固体NaHCO3でpH8〜9に塩基性化した。水層をクロロホルム(25×3mL)で抽出し、水(25mL)およびブライン(25mL)で洗浄した。有機層を無水Na2SO4で脱水し、減圧下で濃縮し、真空で脱水した。クロロホルム中3%メタノールおよび0.1%アンモニアを溶離液として有するカラムクロマトグラフィーで残渣を精製することによって、黄色の固体として、化合物、(+)−trans−2−(2−クロロ−4−トリフルオロメチルフェニル)−8−(2−ヒドロキシメチル−1−メチルピロリジン−3−イル)−5,7−ジメトキシ−クロメン−4−オンを生成した。
収量: 0.56 g (48.2 %); 1H NMR (CDCl3, 300MHz): δ 7.95 (d, 1H), 7.78 (s, 1H), 7.69 (d, 1H), 6.61 (s, 1H), 6.46 (s, 1H), 4.21 (m, 1H), 4.01 (s, 3H), 3.93 (s, 3H), 3.71 (dd, 1H), 3.41 (d, 1H), 3.26 (m, 1H), 2.84 (m, 1H), 2.70 (m, 1H), 2.44 (s, 3H), 2.10 (m, 2H); MS (ES+): m/z 497 (M+1).
パート(a)で得た化合物(0.25g、0.5mmol)、ピリジン塩酸塩(0.25g、2.16mmol)および触媒量のキノリンの混合物を180℃で2.5hの間加熱した。反応混合物をメタノール(25mL)で希釈し、固体Na2CO3でpH10に塩基性化した。反応混合物を濾過し、メタノールで洗浄した。有機層を濃縮し、0.1%アンモニアおよびクロロホルム中4.5%メタノールを溶離液として使用するカラムクロマトグラフィーで残渣を精製することによって、黄色の固体として、化合物、(+)−trans−2−(2−クロロ−4−トリフルオロメチルフェニル)−5,7−ジヒドロキシ−8−(2−ヒドロキシ−メチル−1−メチルピロリジン−3−イル)−クロメン−4−オンを生成した。
収量: 0.15 g (63.7 %); 1 H NMR (CDCl3, 300MHz): δ 7.99 (m, 2H), 7.83 (d, 1H), 6.65 (s, 1H), 6.41 (s, 1H), 4.24 (m, 1H), 3.90 (m, 2H), 3.70 (m, 1H), 3.60 (m, 1H), 3.41 (m, 1H), 2.99 (s, 3H), 2.54 (m, 1H), 2.28 (m, 1H); MS (ES+): m/z 470 (M+1).
(b)で得た化合物(0.1g、0.2mmol)をメタノール(2mL)中に懸濁させ、エーテルのHClで処理し、有機溶媒を蒸発させることによって、化合物、(+)−trans−2−(2−クロロ−4−トリフルオロメチル−フェニル)−5,7−ジヒドロキシ−8−(2−ヒドロキシメチル−1−メチル−ピロリジン−3−イル)−クロメン−4−オン塩酸塩を生成した。
収量: 0.1 g (92.8 %); 1H NMR (CDCl3, 300MHz): δ 8.02 (d, 2H), 7.83 (d, 1H), 6.64 (s, 1H), 6.41 (s, 1H), 4.23 (m, 1H), 3.73 (m, 2H), 3.68 (m, 1H), 3.51 (m, 1H), 3.39 (m, 1H), 2.99 (s, 3H), 2.54 (m, 1H), 2.31 (m, 1H).
(a)(+)−trans−2−(2−クロロフェニル)−8−(2−ヒドロキシメチル−1−メチルピロリジン−3−イル)−5,7−ジメトキシ−クロメン−4−オンの調製
窒素雰囲気下で撹拌しながら、水素化ナトリウム(50%、0.54g、11.25mmol)を、(−)−trans−1−[2−ヒドロキシ−3−(2−ヒドロキシメチル−1−メチルピロリジン−3−イル)−4,6−ジメトキシフェニル)−エタノン(0.7g、2.2mmol)の乾燥DMF(15mL)中溶液に、0℃で少しずつ加えた。10min後、メチル2−クロロベンゾエート(1.15g、6.75mmol)を加えた。反応混合物を25℃で2h撹拌した。メタノールを20℃未満で慎重に加えた。反応混合物をクラッシュアイス(300g)上に注入し、1:1HCl(pH2)で酸性化し、EtOAc(2×100mL)を使用して抽出した。飽和したNa2CO3(pH10)を使用して水層を塩基性化し、CHCl3(3×200mL)を使用して抽出した。有機層を脱水し(無水Na2SO4)、濃縮した。残渣に、濃縮HCl(25mL)を加え、室温で2h撹拌した。反応混合物をクラッシュアイス(300g)上に注入し、Na2CO3飽和水溶液を使用して塩基性にした。CHCl3(3×200mL)を使用して混合物を抽出した。有機抽出物を水で洗浄し、脱水し(無水Na2SO4)、濃縮することによって、化合物、(+)−trans−2−(2−クロロ−フェニル)−8−(2−ヒドロキシメチル−1−メチル−ピロリジン−3−イル)−5,7−ジメトキシ−クロメン−4−オンを得た。

溶融したピリジン塩酸塩(4.1g、35.6mmol)をパート(a)で得た化合物(0.4g、0.9mmol)に加え、180℃で1.5h加熱した。反応混合物を25℃に冷却し,MeOH(10mL)で希釈し、Na2CO3を使用してpH10に塩基性化した。混合物を濾過し、有機層を濃縮した。残渣を水(5mL)中に懸濁させ、30min撹拌し、濾過し、乾燥させることによって、化合物、(+)−trans−2−(2−クロロ−フェニル)−5,7−ジヒドロキシ−8−(2−ヒドロキシメチル−1−メチル−ピロリジン−3−イル)−クロメン−4−オンを得た。
収量: 0.25 g (70 %); IR (KBr): 3422, 3135, 1664, 1623, 1559 cm-1 ; 1H NMR (CDCl3, 300MHz): δ 7.56 (d, 1H), 7.36 (m, 3H), 6.36 (s, 1H), 6.20 (s, 1H), 4.02 (m, 1H), 3.70 (m, 2H), 3.15 (m, 2H), 2.88 (m, 1H), 2.58 (s, 3H), 2.35 (m, 1H), 1 .88 (m, 1H); MS (ES+): m/z 402 (M+1); 分析: C21H20ClNO5 C, 62.24 (62.71); H, 5.07 (4.97); N, 3.60 (3.48); Cl, 9.01 (8.83).
パート(b)で得た化合物(0.2g、0.48mmol)をIPA(5mL)中に懸濁させ、3.5%HCl(25mL)を加えた。懸濁液を加熱して、透明な溶液を得た。溶液を冷却し、固体を濾過することによって、化合物、(+)−trans−2−(2−クロロフェニル)−5,7−ジヒドロキシ−8−(2−ヒドロキシメチル−1−メチル−ピロリジン−3−イル)−クロメン−4−オン塩酸塩を得た。

ATCC(アメリカ培養細胞系統保存機関)、USAから得たG361およびSK−MEL3黒色腫細胞株をこの研究に使用した。G361は、ベムラフェニブ感受性細胞株であり、SK−MEL3はベムラフェニブ耐性細胞株である。両方の細胞株はBRAF V600E変異している。
G361/SK−MEL3黒色腫細胞を25mm3組織培養物フラスコ内に播種した。24h後、G361細胞を以下で5日間処置した:i)化合物A(1μΜ);ii)ベムラフェニブ(1μΜ);およびiii)化合物A(1μΜ)とベムラフェニブ(1μΜ)とを一緒に。
AnnexinV−FITCは、アポトーシス細胞を同定するための感受性プローブである。初期アポトーシスの間、膜リン脂質ホスホチジルセリン(PS)は、原形質膜の内側から外側のリーフレットに転位し、これによってPSが外部細胞環境に曝露される。Annexin Vは、PSに対して高親和性を有し、曝露されたPSを有する細胞に結合する35〜36kDaカルシウム依存性リン脂質結合タンパク質である。
これらの試験の結果は図1〜3に示されている。
図1は、ベムラフェニブで処置したG361細胞は、対照細胞(72.36%)と対比して、91.94%の有意なG1の停止を示していることを指摘している。サブG1期から認められるように、化合物Aおよびベムラフェニブ単独で処置した細胞は、それぞれ9.48%および2.3%アポトーシスしか示さなかったのに対して、化合物Aとベムラフェニブの組合せで処置した細胞は、53.67%の細胞にアポトーシスが起こっていることを示し、これは、組合せが相乗的であることを指摘している。
目的:この研究目的は、BRAF変異した黒色腫細胞における化合物A(CDK阻害剤)とベムラフェニブ(BRAF V600E阻害剤)の組合せの効力を評価することであった。
試験化合物:化合物A(PEL’S Labで調製);ベムラフェニブ(Nanjing Chemlin Chemical Industry Co.、Ltd、中国)
ビヒクル:DMSO(Sigma−Aldrich−Chemie Gmbh、ドイツ)
投薬の準備:化合物Aおよびベムラフェニブは、秤量し、必要量のDMSO中に溶解して、必要とされるストック溶液を得た。
試験システム:試験システムは、G361、A375およびMDAMB−435S(BRAF V600E変異した)細胞株を含んでいた。これらは、ATCC(American Type Culture Collection)、USAから得た。
化合物Aとベムラフェニブの様々な組合せを使用する細胞毒性試験を、CCK8生細胞デヒドロゲナーゼアッセイを使用して行った。
ヒトBRAF V600E変異した黒色腫がん細胞株、G361、A375およびMDAMB−435Sを、96−ウェルプレート中の199μlLのRPMI 1640培地に、3000個の細胞/ウェルの密度で播種し、一晩インキュベートして細胞を付着させた。次いで細胞をそれぞれの試験化合物で処理した。全部で10の群があった;
i)1μΜのベムラフェニブを単独で48h;
ii)0.3μΜ/0.1μΜの化合物Aを単独で48h;
iii)1μΜ/0.3μΜの化合物Aを単独で48h;
iv)3μΜ/1μΜの化合物Aを単独で48h;
v)10μΜ/3μΜの化合物Aを単独で48h;
vi)0.3μΜ/0.1μΜの化合物Aと1μΜのベムラフェニブの組合せを48h;
vii)1μΜ/0.3μΜの化合物Aと1μΜのベムラフェニブの組合せを48h;
viii)3μΜ/1μΜの化合物Aと1μΜのベムラフェニブの組合せを48h;
ix)10μΜ/3μΜの化合物Aと1μΜのベムラフェニブの組合せを48h;
x)対照ウェルをDMSOビヒクルで48h処理した。
スチューデントt検定を使用して統計分析を実施し、p値<0.05は有意とみなされた。データは、平均値±標準誤差(SEM)として表現される。平均値は、それぞれ三重反復で実施される、少なくとも2つの独立した実験から得られる。
対数的に増大する細胞を、3x103細胞/ウェルの密度でプレーティングし、16〜18hの間回復させた。化合物Aとベムラフェニブの両方の化合物の濃度を48h変化させて、細胞を検証した。48h後、細胞毒性をCCK−8試薬(Dojindo Molecular Technologies、Inc、Maryland、およびJapan)により求めた。製造業者の指示に従い、5μL/ウェルのCCK−8試薬を加え、プレートを2hインキュベートした。450nmから650nmの波長に補正し、対照に対して正規化し、Tecan Sapphire マルチ−蛍光 ミクロ−プレートリーダー上の吸光度を測定することによって毒性を求めた。すべての実験は三重反復で実施した。
表1および2は、A375細胞およびG361およびMDA−MB435S細胞における化合物Aおよびベムラフェニブのそれぞれの処置スケジュールを示している。
異なる細胞株における化合物Aおよびベムラフェニブおよびこれらの組合せに関する細胞毒性のIC50値が表3に提示されている。図4〜6および表4〜6は、様々な黒色腫細胞株における化合物Aとベムラフェニブの組合せの効果を表している。
ベムラフェニブと化合物Aの組合せは、BRAF変異した黒色腫細胞において顕著な相乗効果を示した。

本研究の目的は、BRAF変異した黒色腫細胞における、CDK阻害剤(ボルシクリブ)と、BRAF阻害剤(ベムラフェニブまたはダブラフェニブ)またはMEK阻害剤(トラメチニブ)の組合せの効力を評価することであった。
試験化合物:化合物A(PEL’S Lab.で調製);ベムラフェニブ(Selleckchem USA、S1267);ダブラフェニブ(Selleckchem USA、S2807)およびトラメチニブ(Selleckchem USA、S2673)
ビヒクル:DMSO(Sigma−Aldrich−Chemie Gmbh、ドイツ)
投薬の準備:試験化合物を秤量し、必要量のDMSO中に溶解して、必要とされるストック溶液を得た。
試験系:試験系はA375(BRAF V600E変異)細胞株を含んだ。これは、ATCC(American Type Culture Collection(アメリカ培養細胞系統保存機関))、USAおよびA375R細胞株(ベムラフェニブ耐性−PEL’S Lab.で開発)から得た。
CCK8生細胞デヒドロゲナーゼアッセイを使用して、すべての試験化合物を単剤としておよび組み合わせて使用する細胞毒性試験を行った。
対数的に増大するヒトBRAF V600E変異した黒色腫細胞を、1500細胞/ウェルの密度で、384−ウェルプレート(Corning、USA)中の30μLのダルベッコ改変イーグル培地培地(DMEM)に、Tecan自動化プラットフォーム(FreedomEvo Liquid 取扱いシステム)を使用して播種し、約12〜16hインキュベートして、細胞を付着させた。次いで、細胞を異なる用量の化合物Aおよび抗がん剤(ベムラフェニブまたはダブラフェニブまたはトラメチニブ)で、単剤療法として(個々に使用)および組み合わせて、48h処置した。異なる投薬量を使用する試験を、各組合せ(例えば化合物Aとベムラフェニブの組合せ)に対して行った。A375およびA375R細胞において個々におよび組み合わせて使用した場合の試験化合物の処置の比率を表7A〜7Cに示す(すなわち、化合物Aとベムラフェニブの組合せ;化合物Aとダブラフェニブの組合せ;および化合物Aとトラメチニブの組合せ)。化合物A(μΜ)の以下の用量を使用した:1、0.5、0.25、0.125、0.06、0.03、0.015;ベムラフェニブ(μΜ)30、15、7.5、3.75、1.875、0.9、0.4、0.2、0.1、0.05;ダブラフェニブ(μΜ)3、1.5、0.75、0.37、0.18、0.09、0.04、0.02、0.01、0.005;トラメチニブ(μΜ)1、0.5、0.25、0.125、0.0625、0.031、0.015、0.007、0.003、0.001。表7A〜7Cに示されているように、上記用量の抗がん剤(ベムラフェニブまたはダブラフェニブまたはトラメチニブ)を化合物Aと複数の比率で混合した。使用されている対照は、細胞のみまたは細胞とビヒクル(DMSO)である。
これらの試験の結果が図7〜13に示されている。
1.図7aおよび7bから、A375細胞株はベムラフェニブに対して極めて感受性があるのに対して、A375Rは耐性があると結論づけることができる。さらに、A375とA375Rの両方が化合物A(ボルシクリブ)に対して同等に感受性がある。
2.図8aおよび8bから、化合物A(ボルシクリブ)とベムラフェニブの組合せはA375細胞株において強い相乗効果(CI<0.5)を示すと結論づけることができる。
3.図9aおよび9bから、化合物A(ボルシクリブ)とベムラフェニブの組合せは、A375R細胞株において強い相乗効果(CI<0.5)を示すと結論づけることができる。
4.図10aおよび10bから、化合物A(ボルシクリブ)とダブラフェニブの組合せはA375細胞株において強い相乗効果(CI<0.5)を示すと結論づけることができる。
5.図11aおよび11bから、化合物A(ボルシクリブ)とダブラフェニブの組合せはA375R細胞株において強い相乗効果(CI<0.5)を示すと結論づけることができる。
6.図12aおよび12bから、化合物A(ボルシクリブ)とトラメチニブの組合せはA375細胞株において強い相乗効果(CI<0.5)を示すと結論づけることができる。
7.図13aおよび13bから、化合物A(ボルシクリブ)とトラメチニブの組合せはA375R細胞株において強い相乗効果(CI<0.5)を示すと結論づけることができる。
Claims (40)
- 黒色腫を治療するための医薬組成物であって、式I
の化合物または薬学的に許容されるその塩であるCDK(サイクリン依存性キナーゼ)阻害剤の治療有効量と、ベムラフェニブ、ダブラフェニブ、及びトラメチニブから選択される少なくとも1種の抗がん剤の治療有効量を含む、医薬組成物。 - 黒色腫を治療するための医薬組成物であって、式I
- CDK阻害剤が(+)−trans−2−(2−クロロ−4−トリフルオロメチルフェニル)−5,7−ジヒドロキシ−8−(2−ヒドロキシメチル−1−メチル−ピロリジン−3−イル)−クロメン−4−オン塩酸塩である、請求項1または2に記載の医薬組成物。
- 抗がん剤がベムラフェニブである、請求項1から3のいずれか一項に記載の医薬組成物。
- 抗がん剤がダブラフェニブである、請求項1から3のいずれか一項に記載の医薬組成物。
- 抗がん剤がトラメチニブである、請求項1から3のいずれか一項に記載の医薬組成物。
- 前記CDK阻害剤と、前記少なくとも1種の抗がん剤とが、それを必要とする対象に同時に投与される、請求項1から6のいずれか一項に記載の医薬組成物。
- 前記CDK阻害剤と、前記少なくとも1種の抗がん剤とが、それを必要とする対象に逐次的に投与される、請求項1から6のいずれか一項に記載の医薬組成物。
- 黒色腫が非難治性黒色腫である、請求項1から8のいずれか一項に記載の医薬組成物。
- 黒色腫が非難治性BRAF変異型黒色腫である、請求項9に記載の医薬組成物。
