Description
本発明を説明および特許請求するにおいて、以下の用語が下記の定義に従い用いられる。
眼内薬物送達デバイスは、移植可能で、以降の手術が多くの場合不必要なような生物分解する嚢内リザーバーを提供することで、頻回注射または複雑な点眼薬治療法の必要性を軽減することにより、改善された目の薬物の送達を提供することが可能である。さらに、デバイスは異なる薬を寄せ集めまたは組合せ送達することが可能である。
グループII、水晶体超音波乳化吸引およびIOL挿入、DXM点眼剤(漸減を伴う最大4週間)および抗生物質点眼薬剤(最大2日間)、陽性コントロール群、n=6
グループIII、水晶体超音波乳化吸引およびIOL挿入、手術後2日間(1日2回)までの低用量のBDI移植(眼球につき1つの移植片)および抗生物質点眼薬剤、n=8
グループIV、水晶体超音波乳化吸引およびIOL挿入、手術後2日間(1日2回)までの高用量のBDI移植(眼球につき2つの移植片)および抗生物質点眼薬剤、n=8
試験管内の放出動力学の結果を図8に示す。すべてのバッチは第1日目における最初のバースト放出により二相性放出パターンを示し、その後減速し、放出を継続した。バーストの影響は、HPMCを含む移植片により若干より高かった。
Claims (6)
- 眼内活性薬剤送達デバイスであって、
生物分解性の活性薬剤マトリックス内に分散する活性薬剤を有し、前記活性薬剤はデキサメタゾンを含み、前記送達デバイスは固体であり、かつ水晶体嚢内の眼内レンズの視線を遮ることなく、眼球の前記水晶体嚢または毛様溝の中に嵌まるように構成され、前記送達デバイスは前記活性薬剤および前記生物分解性の活性薬剤マトリックスから形成される均質なデバイスであり、前記活性薬剤は約100mcg〜約150mcgの低用量で存在し、前記活性薬剤は約2週間〜約6週間の送達持続時間を有する、眼内活性薬剤送達デバイス。 - 請求項1記載のデバイスにおいて、
前記生物分解性の活性薬剤マトリックスは、ポリ(乳酸−コ−グリコリド)、ポリ乳酸−ポリグリコール酸ブロック共重合体(PLGA)、ヒドロキシプロピルメチルセルロース、ヒドロキシメチルセルロース、ポリグリコリド−ポリビニルアルコール、クロスカルメロースナトリウム、ヒドロキシプロピルセルロース、カルボキシメチルセルロースナトリウム、ポリグリコール酸−ポリビニルアルコールブロック共重合体(PGA/PVA)、ヒドロキシプロピルメチルセルロース(HPMC)およびポリカプロラクトン−ポリエチレングリコールブロック共重合体のうちの少なくとも1つを有するものであるデバイス。 - 請求項1記載のデバイスにおいて、
前記生物分解性の活性薬剤マトリックスは、52/48〜90/10の共重合体比率を有するポリ(乳酸−コ−グリコリド)を有するものであるデバイス。 - 請求項1記載のデバイスにおいて、
前記送達デバイスはディスクまたはペレット形状であるデバイス。 - 請求項1記載のデバイスにおいて、このデバイスは、さらに
前記生物分解性の活性薬剤マトリックスの中に配置される少なくとも1つの第2の活性薬剤リザーバーを有するものであるデバイス。 - 請求項1記載のデバイスにおいて、前記送達デバイスは0.2mg〜4mgの総質量を有するものであるデバイス。
