Description
a)式(II)の2置換アニリンから式(III)のジアゾニウム塩をin situにて生成した後、未反応の式(II)の2置換アニリンを添加して、式(IV)のトリアゼン中間体を形成する工程と、

還元は少なくとも1つの金属触媒を伴って少なくとも1つの還元剤の存在下で行うことができる。還元剤は二水素、ギ酸アンモニウム、ヒドラジンから選択することができる。還元剤は亜鉛/酢酸、鉄/塩酸等の金属元素と酸との組合せ、及び他の既知の還元剤であってもよい。還元剤は有利には二水素であり得る。水素圧は大気圧〜200psi、代替的には50psi〜60psiの範囲であり得る。
2−ニトロベンジルアルコール(100g、0.653mol)を、50%NaOH(136g、1.7mol)と石油エーテル(500mL)との間で分配した。ベンジルトリエチルアンモニウムクロリド(2g)を添加し、続いて添加漏斗を介して硫酸ジメチル(106.4g、0.84mol)を導入した(適度な速度)。混合物を完全に(機械的)撹拌した。硫酸ジメチル(10mL)を添加し、反応物を1時間撹拌した。過剰な硫酸ジメチルをギ酸アンモニウム(100mL)の添加によってクエンチし、混合物を分液漏斗に移した。石油エーテル相を回収した。水相を500mLの石油エーテルで更に1回抽出し、合わせた抽出物を蒸発させ、残留溶媒を高真空下で除去した。2−メトキシメチル−ニトロベンゼンが薄黄色の油として得られた(104g、95%)。TLC分析(50%EtOAc−石油エーテル)により、非常に高い純度の単一成分が示された。H1−NMR(600MHz,CDCl3)δ3.55(s,3H)、4.85(s,2H)、7.43(dd,1H)、7.68(d,1H)、7.79(d,1H)、8.09(d,1H)。C13 NMR(600MHz,CDCl3)135.4、132.8、128.7、128.15、124.8、71.3、59.1。IS−MS m/z 168[MH+]。
2−ブロモメチル−ニトロベンゼン(25g、116mmol)をメタノール(400mL)に溶解した。25%ナトリウムメトキシド(25g、116mmol)を室温で添加した。反応物を50℃に加熱し、反応をTLC分析によってモニタリングした。更に2gの25%ナトリウムメトキシドを添加することで、TLCにより出発物質の完全な消費が示された。メタノールを蒸発させ、残渣をジクロロメタン(700mL)に懸濁し、水(125mLで2回)及び飽和塩化ナトリウムで洗浄した。蒸発により2−メトキシメチル−ニトロベンゼン(19.5g、100%)が薄黄色の油として得られた。TLC及びNMR分析により高い純度が示された。H1−NMR(600MHz,CDCl3)δ3.55(s,3H)、4.85(s,2H)、7.43(dd,1H)、7.68(d,1H)、7.79(d,1H)、8.09(d,1H)。C13 NMR(600MHz,CDCl3)135.4、132.8、128.7、128.15、124.8、71.3、59.1。IS−MS m/z 168[MH+]。
2−メトキシメチル−ニトロベンゼン(100g、0.598mol)を、エタノール(600mL)及び5%Pd/C(5g)と共に2L容Parrボトルに添加した。混合物を50psiで2時間水素化した。反応混合物をセライト(商標)ベッド、続いてガラスマイクロファイバーフィルターに通して濾過し、触媒を残らず除去した。溶媒の蒸発により、プロトンNMR及びTLC分析によって判断されるように非常に高い純度で2−メトキシメチルアニリン(40g、97.5%)が得られた。H1−NMR(600MHz,CDCl3)3.55(s,3H)、4.15(s,2H,NH2)、4.5(s,2H)、6.68(m,2H)、7.08(d,1H)、7.15(d,1H)。C13−NMR(600MHz,CDCl3)146.5、130.8、129.6、122.2、118.8、115.3、73.9、57.8。IS−MS 105.4(MH+,100)
2−メトキシメチルアニリン(5.0g、37.5mmol)を水(25mL)に懸濁した。氷及び濃HClを添加して温度を10℃にした。亜硝酸ナトリウム(1.26g、18.3mmol)を4mLの水に溶解し、上記の冷溶液に温度を10℃未満に維持しながら添加した。酢酸ナトリウム(5.1g、62.2mmol)を10mLの水に溶解し、反応混合物に添加した。反応混合物を氷水浴内に入れ、0℃〜5℃で2時間撹拌した。反応混合物をジクロロメタン(125mL及び50mL)で2回抽出した。合わせた抽出物を蒸発させ、不純な生成物を得た。シリカゲルクロマトグラフィーにより、純粋な1,3−ビス(2−(メトキシメチル)フェニル)トリアザ−1−エン(3.7g、71%)が得られた。H1−NMR(600MHz,CDCl3)3.45(s,6H)、4.7(s,4H)、7.2(m,2H)、7.4(m,4H)、7.7(d,2H)。C13−NMR(600MHz,CDCl3)172、129、125、116、71.8、57.9。IS−MS m/z 286[MH+](5),222[M−2X−OCH3]+,(20)。
1,3−ビス(2−(メトキシメチル)フェニル)トリアザ−1−エン(1.0g、3.5mmol)を、2−(メトキシメチル)アニリン(3.0g、21.87mmol)及び2−(メトキシメチル)アニリン塩酸塩(0.500g、5.8mmol)と共にジクロロメタンに溶解した。混合物を4時間還流させた後、125mLのジクロロメタンに希釈し、1N HCl(5×25mL)で洗浄した。シリカゲルクロマトグラフィー(Combiflash)により、2−(メトキシメチル)−4−((2−(メトキシメチル)フェニル)ジアゼニル)アニリン(0.560g、56%)が赤色の油として得られた。H1−NMR(600MHz,CDCl3)3.4(s,3H)、3.51(s,3H)、4.58(s,2H)、4.65(s,2H,NH2)、5.05(s,2H)、6.78(d,1H)、6.38(d,1H)、6.4(d,1H)、7.61(dd,1H)、7.65(dd,1H)、7.75(s,1H)、7.8(d,1H).C13−NMR 150.25、149.97、145.4、136.6、130.1、128.6、128.2、128.17、125.15、121.85、115.6、73.9、70.15、58.68、57.75
2−(メトキシメチル)−4−((2−(メトキシメチル)フェニル)ジアゼニル)アニリン(0.75g、2.63mmol)をメタノール(10mL)に溶解した。ギ酸アンモニウム(2.0g)及び活性亜鉛(2g)を添加し、混合物を室温で撹拌した。1時間後に、TLC分析により出発物質の完全な消費が示された。シリカゲルクロマトグラフィー(combiflash)により、2−メトキシメチル−1,4−ベンゼンジアミン(0.180g、45%)が白色の固体として得られた。H1−NMR(500MHz,CDCl3)δ3.2〜3.9(m,7H)、4.39(s,2H)、6.5(s,1H)、6.56(s,2H)。
Claims (14)
- 2置換1,4−ベンゼンジアミン(I)、化粧品として許容可能なその塩又はそれらの混合物を調製する方法であって、
a)式(II)の2置換アニリンから式(III)のジアゾニウム塩をin situにて生成した後、未反応の式(II)の2置換アニリンを添加して、式(IV)のトリアゼン中間体を形成する工程と、
X−は無機又は有機アニオンであり、
m=0.5、1又は2である)を含む、方法。 - 工程a)を少なくとも1つの鉱酸又は有機酸と組み合わせた少なくとも1つのニトロソ化剤の存在下で行い、該ニトロソ化剤が亜硝酸ナトリウム、亜硝酸カリウム、N2O5、ニトロシル硫酸及びそれらの混合物からなる群から選択され、該鉱酸又は有機酸が塩化水素、硫酸、酢酸及びそれらの混合物からなる群から選択される、請求項1に記載の方法。
- 工程a)を、アクリロニトリル、メタクリレート、尿素及びそれらの混合物からなる群から選択される少なくとも1つのラジカル捕捉剤の存在下で行う、請求項1又は2に記載の方法。
- 工程a)及び/又はb)をペンタン、シクロペンタン、ヘキサン、シクロヘキサン、ベンゼン、トルエン、1,4−ジオキサン、クロロホルム、ジエチルエーテル、ジクロロメタン、テトラヒドロフラン、メチル−テトラヒドロフラン、1,2−ジメトキシエタン、酢酸エチル、アセトン、ジメチルホルムアミド、アセトニトリル、ジメチルスルホキシド、グリコール、n−ブタノール、イソプロパノール、n−プロパノール、エタノール、メタノール、水及びそれらの混合物からなる群から選択される少なくとも1つの溶媒の存在下で行う、請求項1〜3のいずれか一項に記載の方法。
- 工程c)を少なくとも1つの金属触媒を伴って少なくとも1つの還元剤の存在下で行い、該触媒がラネーニッケル、ニッケル、パラジウム、リンドラー触媒、コバルト、銅クロマイト、白金、酸化白金、レニウム、塩化スズ(II)、塩化チタン(III)、亜鉛、サマリウム、鉄及びそれらの混合物からなる群から選択される、請求項1〜4のいずれか一項に記載の方法。
- 工程c)を水、石油エーテル、ペンタン、シクロペンタン、ヘキサン、シクロヘキサン、ベンゼン、トルエン、1,4−ジオキサン、クロロホルム、ジエチルエーテル、ジクロロメタン、テトラヒドロフラン、メチル−テトラヒドロフラン、酢酸エチル、アセトン、ジメチルホルムアミド、アセトニトリル、ジメチルスルホキシド、n−ブタノール、イソプロパノール、n−プロパノール、エタノール、メタノール及びそれらの混合物からなる群から選択される少なくとも1つの溶媒の存在下で行う、請求項1〜5のいずれか一項に記載の方法。
- 工程d)において使用される前記酸がD,L−リンゴ酸、L−リンゴ酸、D−リンゴ酸、塩酸、臭化水素酸、クエン酸、酢酸、乳酸、コハク酸、酒石酸、リン酸又は硫酸、及びそれらの混合物からなる群から選択される、請求項1〜6のいずれか一項に記載の方法。
- 工程d)をペンタン、シクロペンタン、ヘキサン、シクロヘキサン、ベンゼン、トルエン、1,4−ジオキサン、クロロホルム、ジエチルエーテル、ジクロロメタン、テトラヒドロフラン、メチル−テトラヒドロフラン、酢酸エチル、アセトン、ジメチルホルムアミド、アセトニトリル、ジメチルスルホキシド、n−ブタノール、イソプロパノール、n−プロパノール、エタノール、メタノール及びそれらの混合物からなる群から選択される少なくとも1つの溶媒の存在下で行う、請求項1〜7のいずれか一項に記載の方法。
- 前記エーテル化をアルキル化反応、縮合反応又は求核置換によって行う、請求項9に記載の方法。
- 前記アルコキシ化に使用されるアルコキシ化剤がアルコール、ナトリウムアルコキシド、カリウムアルコキシド、亜鉛アルコキシド、カルシウムアルコキシド、マグネシウムアルコキシド、タンタルアルコキシド、トリブチルスズアルコキシド、それらの塩及びそれらの混合物からなる群から選択される、請求項11に記載の方法。
- 前記アルコキシ化をペンタン、シクロペンタン、ヘキサン、シクロヘキサン、ベンゼン、トルエン、1,4−ジオキサン、クロロホルム、ジエチルエーテル、テトラヒドロフラン、メチル−テトラヒドロフラン、アセトン、ジメチルホルムアミド、アセトニトリル、ジメチルスルホキシド、n−ブタノール、イソプロパノール、n−プロパノール、エタノール、メタノール及びそれらの混合物からなる群から選択される溶媒を用いて行う、請求項11又は12に記載の方法。
- 前記還元を少なくとも1つの金属触媒を伴って少なくとも1つの還元剤の存在下で行い、該触媒がラネーニッケル、ニッケル、パラジウム、リンドラー触媒、コバルト、銅クロマイト、白金、酸化白金、レニウム、塩化スズ(II)、塩化チタン(III)、亜鉛、サマリウム、鉄及びそれらの混合物からなる群から選択される、請求項9〜13のいずれか一項に記載の方法。
