Description
発育鶏卵にて増幅し、限外ろ過、しょ糖密度勾配遠心分離法及びベータプロピオラクトンによって精製及び不活化したA/Brisbane/59/2007株の不活化全粒子ウイルスに、終濃度が1.0%となるように、TritonX−100(商品名、シグマ アルドリッチ ジャパン社製)、NP−40(商品名、ナカライテスク社製)、Tween80(商品名、和光純薬工業社製)、Brij35(商品名、和光純薬工業社製)、CHAPS(商品名、同仁化学研究所社製)、Zwittergent 3−14(商品名、Calbiochem社製)及びCTAB(和光純薬工業社製)を添加し、終濃度が0.3%となるようにSDSを、終濃度が4.0M及び3.0MとなるようにUrea(エムピーバイオジャパン社製)及び塩酸グアニジン(エムピーバイオジャパン社製)を添加して、37℃にて60分間静置して反応させた。反応溶液に終濃度20%となるようにしょ糖を添加して、これを分画密度20〜50w/w%のしょ糖密度勾配遠心分離法にて17フラクションに分画した。各フラクション溶液とSDS−PAGE用サンプルバッファー(8%SDS,40%グリセロール/250mM Tris−HCl Buffer pH6.8)とを等量混合して100℃にて5分間静置にて反応させた。反応溶液を12.5% ポリアクリルアミドゲル(アトー社製 ePAGEL)にて電気泳動し、セミドライ式転写装置(アトー社製)にてPVDF膜への転写反応を行った。転写後のPVDF膜を75mLのブロッキングバッファー(10%スキムミルク含有TBS)に浸し、室温にて4時間のマスキング反応を行った。反応後、適当量のTBSにてPVDF膜を3回洗浄し、その後PVDF膜を抗HA抗体溶液(SRD(Single radial immunodiffusion)用抗血清)に浸して4℃にて約16時間反応した(一次抗体反応)。一次抗体反応後、Tween20(商品名)含有TBSにてPVDF膜を5回洗浄し、HRP標識抗ヒツジ抗体(Bethyl社製)溶液を添加して室温にて60分間反応させた(二次抗体反応)。二次抗体反応後、Tween20(商品名)含有TBSにてPVDF膜を5回洗浄し、Super Signal West Pico Chemiluminescent Substrate(商品名、サーモサイエンチフィック社製)にてヘムアグルチニンの検出を行った。検出には、LAS-3000(商品名、GEヘルスケア社製)を使用した。
MDCK細胞及び発育鶏卵にて、A/California/7/2009(H1N1)、A/Brisbane/59/2007(H1N1)、A/Victoria/210/2009(H3N2)、A/Uruguay/716/2007(H3N2)、B/Brisbane/60/2008(B型 Victoria lineage)及びB/Florida/4/2006(B型 Yamagata lineage)の6株のウイルス溶液を調製した。調製したウイルスとSDS−PAGE用サンプルバッファー(8%SDS,40%グリセロール/250mM Tris−HCl Buffer pH6.8)とを等量混合して100℃にて5分間静置にて反応させた。反応溶液を12.5% ポリアクリルアミドゲル(アトー社製 ePAGEL)にて電気泳動し、セミドライ式転写装置(アトー社製)にてPVDF膜への転写反応を行った。転写後のPVDF膜を75mLのブロッキングバッファー(10%スキムミルク含有TBS)に浸し、室温にて4時間のマスキング反応を行った。反応後、適当量のTBSにてPVDF膜を3回洗浄し、各種ビオチン化レクチン:VECTOR LABORATORIES社製)との反応を室温にて4時間行った。レクチンとの反応後、Tween20(商品名)含有TBSにてPVDF膜を5回洗浄し、HRP標識ストレプトアビジン(サーモサイエンティフィック社製)溶液を添加して室温にて60分間反応させた。反応後、Tween20(商品名)含有TBSにてPVDF膜を5回洗浄し、Super Signal West Pico Chemiluminescent Substrate(商品名、サーモサイエンチフィック社製)にてヘムアグルチニンとレクチンとの複合体を検出した。検出には、LAS-3000(商品名、GEヘルスケア社製)を使用した。
ストレプトアビジンコートしたマイクロプレート(Nunc社製)に0.05%Tween20(商品名)/トリス塩酸緩衝液生理食塩水(TBST)で終濃度が30μg/mLとなるように希釈した各ビオチン化レクチンを100μL/ウェルで添加して25℃で2時間反応させた。レクチン反応後、各ウェルをWash buffer(TBST)300μLで5回洗浄した。次に、2.5%スキムミルク/TBSTを各ウェルに300μL添加して、25℃で1時間のブロッキングを行い、その後、各ウェルをWash buffer 300μLで5回洗浄し、0.5%スキムミルク/TBSTもしくは0.5%BSA/TBSTで希釈したHRP標識化IgG抗体(マウス抗体:BioSS社製 Mouse Anti−Rabbit IgG Secondary Antibody(H+L),HRP Conjugated、ウサギ抗体:Bethyl Laboratories社製 Sheep IgG−heavy and light chain antibody、ヒツジ抗体:Bethyl Laboratories社製 Rabbit IgG−heavy and light chain antibody)を各ウェルに100μL添加して25℃で1時間反応させた。抗体反応後、各ウェルをWash buffer300μLで5回洗浄し、TMB溶液(和光純薬工業社製)を各ウェルに200μL添加して25℃で20分間反応させた後、1mol/L硫酸(和光純薬工業社製)を各ウェルに50μL添加して反応を停止した。反応停止後、450nmにおける吸光度を測定した。
NIBSC(The National Institute for Biological Standards and Control)より購入したSRD試験用標準抗原のバイアルに1mLの水を加え、5分間静置後に十分に攪拌した(50μg HA/mL)。50μLのNIBSC標準品(50μg HA/mL)と50μLの1.0%CTABを混ぜて十分に攪拌した後(25μg HA/mL)、37℃で2時間静置した。次いで、10倍希釈して2.5μgHA/mLのヘムアグルチニン溶液を調製した。この標準液を希釈し、3.13、6.25、12.5、25、50、100及び250ngHA/mLのヘムアグルチニンン溶液を調製した。
A/California/07/2009(X−179A)、A/Victoria/361/2011(IVR−165)及びB/Wisconsin/01/2010(BX−41A)の標準インフルエンザHA抗原(一元放射免疫拡散試験用 国立感染症研究所)の各バイアルに1mLの水を加え、5分間静置後に十分に攪拌した。50μLの標準インフルエンザHA抗原溶液と50μLの1.0%CTABを混ぜて十分に攪拌した後、37℃で2時間静置した。次いで、10倍希釈してヘムアグルチニン溶液を調製した。この標準液を希釈し、1.95、3.91、7.81、31.3、62.5及び125ngHA/mLのヘムアグルチニンン溶液を調製してこれを検量線用標準溶液とした。また、一元放射免疫拡散試験(SRD試験)によりHA濃度を値付けた製造工程液(A/California/07/2009(X−179A)株、A/Victoria/361/2011(IVR−165)株及びB/Wisconsin/01/2010(BX−41A)株)も上記の検量線用標準溶液と同様にCTAB処理及び希釈操作を行い測定検体とした。
Claims (5)
- インフルエンザウイルスのヘムアグルチニンと結合するが、該ヘムアグルチニンと抗原抗体反応する抗ヘムアグルチニン抗体と結合しないレクチンと、該抗ヘムアグルチニン抗体とで該ヘムアグルチニンをサンドイッチすることを含む、サンドイッチ免疫測定法によるインフルエンザウイルスのヘムアグルチニンの測定方法であって、
前記レクチンが、ヒママメレクチン、チョウセンアサガオレクチン及びデイゴレクチンから成る群より選ばれる少なくとも1種である、測定方法。 - 前記レクチンが固相に固定化され、前記抗ヘムアグルチニン抗体が標識されており、前記レクチンと前記ヘムアグルチニンを介して固相に結合した該標識抗体を測定することを含む請求項1記載の方法。
- 前記ヘムアグルチニンが、インフルエンザウイルスを陽イオン性又は陰イオン性界面活性剤で処理することにより抽出されたものである請求項1又は2記載の方法。
- 前記陽イオン性又は陰イオン性界面活性剤が、ドデシル硫酸ナトリウム、ドデシル硫酸リチウム、臭化ヘキサデシルトリメチルアンモニウム、塩化ヘキサデシルトリメチルアンモニウム及び塩化ヘキサデシルピリジニウムから成る群より選ばれる少なくとも1種である請求項3記載の方法。
- 前記界面活性剤が、臭化ヘキサデシルトリメチルアンモニウムである請求項4記載の方法。
