Description
・形態学的特徴−偏性嫌気性条件下、固体MRS寒天増殖培地で増殖する細菌コロニーは、楕円形でわずかに凸状、乳白色であり、縁は規則的で、大きさは約1mmである;未熟なコロニーはクリーム色であり、成長したものはさびた色合いを呈する。顕微鏡下では、それらの細菌は、単一または群のねじれた短桿菌の形を呈する。それらは内生胞子を形成しない。それらの細菌はグラム+、カタラーゼ陰性である。
・生理学的および生化学的特徴−株増殖の最適温度は30℃〜37℃である。25℃および45℃の限界温度では増殖は起こらない。
試験の範囲に、2%フルクトースを含む流動MRS培地中での25℃、30℃、37℃および45℃で24時間にわたるラクトバチルス・デルブリッキーKKP 1645 p生菌の増殖を含めた[表1]。この試験では、乳酸発酵菌増殖の増大が観察されなかったのは25℃および45℃のみであった。
試験の範囲に、NaClの種々の添加物(0.5〜12%)が入った2%フルクトース含有流動MRS培地中での37℃の温度で24時間にわたるラクトバチルス・デルブリッキーKKP 1645 p生菌の増殖を含めた[表2]。この試験は、乳酸発酵菌の増殖に対するNaClの有意な効果を示さなかった。
試験の範囲に、pH4.0、5.5、6.5、7.5、8.0の2%フルクトース含有流動MRS培地中での37℃の温度で24時間にわたるラクトバチルス・デルブリッキーKKP 1645 p生菌の増殖を含めた[表3]。この試験では、乳酸発酵菌増殖の増大が観察されなかったのはpH4.0のみであった。
2%フルクトースを含むMRSブリオン(MRS bullion)中でのラクトバチルス・デルブリッキーKKP 1645 p生菌の増殖、37℃の温度で24時間置いた。
OD(光学密度)システムで細菌の増殖におけるワートの効果を判定した。細菌の繁殖は、流動増殖培地1cm3を移植した後に行った、24時間のラクトバチルス・デルブリッキーKKP 1645 p菌増殖。7、15、30、40、45および50°Blgのハニーワートに関する細菌細胞の増殖を判定するために、光学密度の定量を実施した[表5]。OD測定は、分光光度計(spectrophometer)を波長λ=550nmで使用して実施した。ハニーワートに関するラクトバチルス・デルブリッキーKKP 1645 p株のバイオマス増殖は、既知乳酸桿菌属株と比べて、著しく高くなっている。新規ラクトバチルス・デルブリッキーKKP 1645 p株は、高い糖濃度に対して耐性があり、すなわち、他の乳酸発酵菌と比べて高浸透圧耐性を有する。ハニーワートに関するラクトバチルス・デルブリッキーKKP 1645 p菌増殖を図1に示す。
アンタゴニストラクトバチルス・デルブリッキーKKP 1645 p株の特性の決定。
株間拮抗作用試験を滴注法(Klewicka E. et al. 1999. Antagonistic activity of Lactobacillus acidophilus lactic acid fermentation bacteria. Food. Science. Technology. Quality)によって実施した。その試験は対照および供試株の増殖も含む。対照微生物の群は、セレウス菌(Bacillus cereus)KKP 358、リステリア菌(Listeria monocytogenes)KKP 1170、サルモネラ菌属種(Salmonella sp.)KKP 996、緑膿菌(Pseudomonas aeruginosa)KKP 161、大腸菌(Escherichia coli)KKP 364、セラチア・リクファシエンス(Serratia liquefaciens)KKP 1665、黄色ブドウ球菌(Staphylococcus aureus)KKP 1497、腐性ブドウ球菌(Staphylococcus saprophyticus)KKP 1493とした。これらの細菌は、IAFBのカルチャー・コレクション・オブ・インダストリアル・マイクロオーガニズムズ(Culture Collection of Industrial Micro-organisms)由来のものであった。
選択した乳酸ラクトバチルス・デルブリッキーKKP 1645 p菌を特性評価するために、MRS寒天増殖培地およびCaCO3含有Blickfeldt溶液を用いたKB試験により抗生物質耐性分析を実施した[表6]。
ビーポーレンを生産するための本発明による細菌株の使用の可能性を試験するために、それを用いた発酵を実施した。48時間増殖物として10%または50%の株接種材料を、花粉を含むワートに加えた。加えた接種材料中の細菌の数は106〜107jtk/mlの範囲であった。同時に、対照サンプルでの発酵を、天然花粉微生物叢のみ含め、ワート密度7、14、30°Blgを用いて実施した。

- ビーポーレンを生産するための、請求項1に記載の株の使用。
