・ケイ素溶出型炭酸カルシウム(Si-CaCO3):消石灰(ミクロスターT 純度96%以上 矢橋工業株式会社)、メタノール(特級試薬 純度99.8%以上 キシダ化学株式会社)、γ‐アミノプロピルトリエトキシシラン(TSL8331 純度98%以上 GE東芝シリコーン株式会社)、炭酸ガス(高純度液化炭酸ガス 純度99.9% 大洋化学工業株式会社)を用いて調製されたSi含量:2.9wt%のバテライト
・ポリ乳酸(PLA):PURASORB PL Poly(L-lactide) 分子量20-30万 PURAC biochem
・クロロホルム(CHCl3):特級試薬 純度99.0%以上 キシダ化学株式会社
・N,N-ジメチルホルムアミド(DMF):特級試薬 純度99.5%以上 キシダ化学株式会社
PLA:10g、CHCl3:110g、DMF:27.5gを混合した紡糸溶液(PLA:6.8wt%)、Si-CaCO3:15g、PLA:10g、CHCl3:140g、DMF:35gを混合した紡糸溶液(Si-CaCO3:7.5wt%、PLA:5.0wt%)を準備した。これらの紡糸溶液を用いて、エレクトロスピング法により不織布2層構造を有するGBR膜を作製した。
紡糸溶液供給速度:約0.1ml/min、印加電圧:15kV、ノズルとコレクターの距離:10cm、ノズル:15cm/minで左右(3-4cm幅)に移動、コンベア式コレクター(コンベア速度:5-6m/min)、紡糸時間:約170分
紡糸溶液供給速度:約0.16ml/min、印加電圧:20kV、ノズルとコレクターの距離:10cm、ノズル:15cm/minで左右(3-4cm幅)に移動、コンベア式コレクター(コンベア速度:5-6m/min)、紡糸時間:約130分
PLA:9g、CHCl3:91gを混合した紡糸溶液(PLA:9.0wt%)を用いてエレクトロスピニング法によりPLA層を作製した。
紡糸溶液供給速度:0.05ml/min、印加電圧:20kV、ノズルとコレクターの距離:15cm、ノズル:固定、プレートコレクター:固定、紡糸時間:60分
PLAとSi-CaCO3を加熱ニーダーで200℃、15分間混練してSi-CaCO3を60wt%含有するSi-CaCO3/PLA複合体を調製した。Si-CaCO3/PLA複合体:25g、CHCl3:90gを混合した紡糸溶液(Si-CaCO3:13.0wt%、PLA:8.7wt%)を用いて、エレクトロスピング法によりSi-CaCO3/PLA層を作製した。
紡糸溶液供給速度:0.05ml/min、印加電圧:20kV、ノズルとコレクターの距離:15cm、ノズル:固定、プレートコレクター:固定、紡糸時間:30分

24wellプレート使用
細胞種;マウス骨芽細胞様細胞(MC3T3-E1細胞:理研)
細胞播種数;1X104cell/well
培地;α-MEM(10%ウシ胎仔血清含有)
培地交換;播種翌日、その後は1日おき
・細胞カウント方法
Cell Counting Kit-8(細胞増殖/細胞毒性測定試薬、株式会社同仁化学研究所)を使用。試薬添付のプロトコルに沿って処理。
Claims (10)
- バテライト相であるケイ素溶出型炭酸カルシウムと生分解性樹脂とを主成分として含む繊維からなることを特徴とする生体吸収性骨組織誘導再生用材料。
- 前記繊維が、ケイ素溶出型炭酸カルシウムと生分解性樹脂との複合体を溶媒に溶かして得られた紡糸溶液をエレクトロスピニング法に供することによって得られたものである、請求項1に記載の生体吸収性骨組織誘導再生用材料。
- 前記生分解性樹脂がポリ乳酸である、請求項1又は2に記載の生体吸収性骨組織誘導再生用材料。
- 前記ケイ素溶出型炭酸カルシウムがケイ素原子とバテライト相炭酸カルシウムとを含む複合体である、請求項1乃至3のいずれか一項に記載の生体吸収性骨組織誘導再生用材料。
- 前記生体吸収性骨組織誘導再生用材料が不織布である、請求項1乃至4のいずれか一項に記載の生体吸収性骨組織誘導再生用材料。
- 前記生体吸収性骨組織誘導再生用材料がさらにアパタイトを含み、前記繊維がアパタイトでコーティングされている、請求項1乃至5のいずれか一項に記載の生体吸収性骨組織誘導再生用材料。
- 請求項1乃至6のいずれか一項に記載の生体吸収性骨組織誘導再生用材料と生分解性樹脂を主成分とする不織布層とを有する骨再生誘導膜から構成されていることを特徴とする生体吸収性骨組織誘導再生用材料。
- 請求項1乃至6のいずれか一項に記載の生体吸収性骨組織誘導再生用材料も不織布層となっており、前記骨再生誘導膜が不織布層の2層構造を有する、請求項7に記載の生体吸収性骨組織誘導再生用材料。
- バテライト相であるケイ素溶出型炭酸カルシウムと生分解性樹脂との複合体を調製する工程(i)、
前記複合体を溶媒に溶かして紡糸溶液とする工程(ii)、
前記紡糸溶液をエレクトロスピニング法に供して繊維とする工程(iii)、及び
前記工程(iii)で得られた繊維をコレクター上で薄い繊維の層とする工程(iv)
を含むことを特徴とする生体吸収性骨組織誘導再生用材料の製造方法。 - さらに、前記繊維の表面をアパタイトでコーティングする工程(v)を含む、請求項9に記載の生体吸収性骨組織誘導再生用材料の製造方法。
