Description
すなわち、本発明は、ポインセチア抽出物を含有することを特徴とするラジカル消去性または活性酸素消去性抗酸化剤およびそれを用いた化粧料組成物を提供する。
参考例1
ポインセチア苞由来抽出物
赤く色づいたポインセチア(ミレニアム)の苞を切り取り、その10グラムをエタノール100ミリリットルに一晩浸漬した。その後、浸漬液を定性濾紙(No.2)で濾過し、抽出液とした。抽出残渣はさらに100ミリリットルのエタノールで同様に抽出及び濾過を繰り返し、200ミリリットルの抽出液を得た。抽出液はロータリーエバポレーターでエタノールを留去し、乾固後、1.0gの赤紫色のポインセチア苞由来抽出物を得た。
ポインセチア苞由来抽出物のラジカル消去能
DPPH(1,1-diphenyl-2-pycrylhydrazyl)ラジカルに対する消去能を測定し、試料の抗酸化性を分光学的に分析した。
参考例1のポインセチア苞由来抽出物1.0gを水5ミリリットルに溶解し、ポインセチア苞由来抽出物溶液原液とした。96穴マイクロプレートを用い、50%エタノール水溶液で適宜に希釈したポインセチア苞由来抽出物溶液20マイクロリットルに125マイクロリットルの0.2mM DPPH(エタノール溶液)と125マイクロリットルの0.2M MES(2−モルホノリノエタンスルホン酸水溶液 pH6.0)を加え室温におき、20分後に510ナノメートルの吸光度を測定した。コントロールには試料溶液の代わりに50%エタノール水溶液、試料が着色しているためブランクとして試料溶液に50%エタノール水溶液250マイクロリットルを加えたものを用いて同時に測定した。なお、ラジカル消去活性は各吸光度を用いて次式により算出した。
ラジカル消去活性(%)=(1−(試料−ブランク)/コントロール)×100
この結果を表1に示す。この結果よりポインセチア苞由来抽出物はラジカル消去能を有していることが明らかになった。
ポインセチア苞由来抽出物のSOD活性
参考例2のポインセチア苞由来抽出物溶液原液を50%エタノール水溶液にて適宜希釈して調製したポインセチア苞由来抽出物溶液を用い、該ポインセチア苞由来抽出物のSOD活性を測定した。測定には、キサンチン−キサンチンオキシダーゼをスーパーオキシドの発生系とし、発生したスーパーオキシドが水溶性ホルマザンを生成することを検出原理としたSOD活性測定キットSOD Assay Kit(dojindo製)を用いた。なお、コントロールには試料溶液の代わりに純水を用いた。また、試料が着色しているためブランクとしては、キサンチンオキシダーゼの代わりに希釈用緩衝液(キットに付属)を用いて同時に測定した。SOD活性(阻害率)は各吸光度を用いて次式により算出した。
SOD活性(阻害率)(%)=(1−(試料−ブランク)/コントロール)×100
この結果を表2に示す。この結果よりポインセチア苞由来抽出物はSOD活性を有していることが明らかになった。
ポインセチア根由来抽出物
ポインセチア(ミレニアム)の根を切り取り、その10グラムをエタノール200ミリリットルに3日間浸漬した。その後、浸漬液を定性濾紙(No.2)で濾過し、抽出液とした。抽出残渣はさらに200ミリリットルのエタノールで同様に抽出濾過を繰り返し、400ミリリットルの抽出液を得た。抽出液はロータリーエバポレーターでエタノールを留去し、乾固後、53mgの淡黄色のポインセチア根由来抽出物を得た。
ポインセチア根由来抽出物のラジカル消去能
DPPH(1,1-diphenyl-2-pycrylhydrazyl)ラジカルに対する消去能を測定し、試料の抗酸化性を分光学的に分析した。実施例4のポインセチア根由来抽出物50mgを水10ミリリットルに溶解し、ポインセチア根由来抽出物溶液原液とした。96穴マイクロプレートを用い、50%エタノール水溶液で適宜に希釈したポインセチア根由来抽出物溶液20マイクロリットルに125マイクロリットルの0.2mM DPPH(エタノール溶液)と125マイクロリットルの0.2M MES(2−モルホノリノエタンスルホン酸水溶液 pH6.0)を加え室温におき、20分後に510ナノメートルの吸光度を測定した。コントロールには試料溶液の代わりに50%エタノール水溶液、試料が着色しているためブランクとして試料溶液に50%エタノール水溶液250マイクロリットルを加えたものを用いて同時に測定した。なお、ラジカル消去活性は各吸光度を用いて次式により算出した。
ラジカル消去活性(%)=(1−(試料−ブランク)/コントロール)×100
この結果を表3に示す。この結果よりポインセチア根由来抽出物はラジカル消去能を有していることが明らかになった。

・プロピレングリコール 4.0質量部
・1%ポインセチア苞由来抽出物(参考例1)
1,3ブチレングリコール溶液 2.0質量部
・ポリオキシエチレンラウリルエーテル
(20E.O.) 1.0質量部
・エタノール 10.0質量部
・酸化防止剤 適量
・防腐剤 適量
・香料 適量
・精製水 残部
合計 100質量部
・グリセリン 5.0質量部
・流動パラフィン 5.0質量部
・ミリスチン酸イソプロピル 5.0質量部
・1%ポインセチア苞由来抽出物(参考例1)
1,3ブチレングリコール溶液 2.0質量部
・ポリオキシエチレン硬化ヒマシ油 5.0質量部
・エタノール 5.0質量部
・酸化防止剤 適量
・防腐剤 適量
・香料 適量
・精製水 残部
合計 100質量部

- ポインセチアの根のエタノール抽出物を含有することを特徴とするラジカル消去性抗酸化剤。
- ポインセチアの根のエタノール抽出物を含有することを特徴とする活性酸素消去性抗酸化剤。
- 請求項1または2に記載の抗酸化剤を含有することを特徴とする化粧料組成物。
