Description
材料および方法DNA塩基配列決定: DNA塩基配列決定は、Sangerら(1977, P.N.A.S. USA, 74:6543-5647)に記載されているとおりに実施した。
発現カセットおよびベクターの構築: GUSレポーター遺伝子を用いて発現分析用カセットを以下のとおりに構築した。プロモーターの無いGUSカセットは、pBI101をHindIIIおよびEcoRIで消化し、pUC19のポリクローニング部位のHindIIIおよびEcoRI部位に挿入した。得られたプラスミドをpBI201と命名し、次のコンストラクト構築のために使用した。pGPlas3-2の種々の欠失断片を、pBI201のGUSレポーター遺伝子の5'端に転写的および移行的に融合し、DNAボンバードメントを用いた一時的発現試験に使用した。適切な欠失断片を同定し、pBI101(Clonetech社)などのバイナリー植物発現ベクターにサブクローニングした。これらの組換えプラスミドを、下記のA.ツメファシエンス(A. tumefaciens)感染により安定な組み込みのために用いた。
Claims (4)
- 配列番号2を有する単離されたプロモーターであって、該プロモーターは外来性遺伝子の発現が操作可能であるように該遺伝子に位置付けられている、前記プロモーター。
- 請求項1記載のプロモーターを含む発現ベクター。
- 請求項2記載の発現ベクターで遺伝子形質転換された、植物細胞又は植物。
- 遺伝子形質転換された、双子葉植物細胞又は双子葉植物である、請求項3記載の遺伝子形質転換された植物細胞又は植物。
