また、本発明は、核酸類と強固な複合体を形成する機能ととともに、プロトンスポンジ効果を発現する機能を併せ持つ薬物運搬体として、カチオン性の高分子化合物を提供することを目的とする。
本発明における高分子化合物は、ヘッド部、テイル部ともに各種のものが本発明に包含される。
(I)
以下、実施例により本発明をさらに具体的に説明する。但し、本発明は実施例に限定されるものではない。
ポリアミドアミンデンドロンとポリリシンからなるヘッド−テイル型高分子化合物の酸塩基滴定
ポリアミドアミンデンドロンとポリリシンからなるヘッド−テイル型高分子化合物(ポリアミドアミンデンドロン世代数3.5、ヘッド−テイル型高分子化合物1本鎖当たり重合度70のポリリシン)を0.05M塩酸に溶解した後、0.01M水酸化ナトリウム水溶液を滴下することによって滴定した。
ポリアミドアミンデンドロンとポリリシンからなるヘッド−テイル型高分子化合物とプラスミドDNAからの複合体調製
ポリアミドアミンデンドロンとポリリシンからなるヘッド−テイル型高分子化合物(ポリアミドアミンデンドロン世代数3.5、ヘッド−テイル型高分子化合物1本鎖当たり重合度70のポリリシン)をトリス緩衝液(塩濃度10mM、pH7.4)に、プラスミドDNAをトリス緩衝液に溶解した後、この各々の溶液を種々混合比で混合した。ポリリシンとプラスミドDNA、ポリアミドアミンデンドロンとプラスミドDNAについても同様に混合した。
得られた混合溶液について、DNA可視化色素としてエチジウムブロミドを含有したアガロースゲル電気泳動を行った。また、ルミノイメージアナライザーUV-LAS1000mini(富士写真フィルム社)により画像解析を行い、複合体形成していないDNA量を定量した。
以上のことから、ヘッド−テイル型高分子化合物は、テイル部でDNAと複合体形成し、ヘッド部はプロトンスポンジ効果を発現できる複合体であることが確認された。
