Description
まず、SYBR(登録商標)Green Iを用いるインターカレーター法等が挙げられる。インターカレーター法では、二本鎖DNAに結合することで蛍光を発する性質を有するインターカレーターを用いる。このインターカレーターをPCR反応の過程で生成する二本鎖DNAに結合させ、これに対して励起光を照射することで蛍光が発せられる。この蛍光強度を検出することで、PCR増幅産物の生成量をモニターする。このインターカレーター法では、ターゲットに特異的な蛍光標識プローブを設計・合成する必要がなく、種々のターゲットの測定に簡便に利用できる。
dsDNAからRNAを増幅産物として得るIVT法(In Vitro Transcription);
逆転写酵素のRNaseH活性を用いてRNAをトリミングし、RNAを増幅産物として得るTRC法(Transcription Reverse transcription Concerted amplification);
逆転写酵素等を用いてRNAプロモーターが組み込まれたdsDNAとし、次にRNAポリメラーゼを用いてdsNDAからRNAを増幅産物として得るNASBA法(Nucleic Acid Sequence-Based Amplification);
RNAとDNAのキメラ構造からなるプライマーを利用し、RNAからssDNAを増幅産物として得るSPIA法;
DNAのループ形成を利用し、一定温度でDNAやRNAからdsDNAを増幅産物として得るLAMP法(Loop-Mediated Isothermal Amplification);
複数の酵素を組み合わせて用い、一塩基の違い(SNP)を正確に識別しながらdsDNAからdsDNAを増幅産物として得るSMAP法(SMart Amplification Process);
DNAとRNAが結合した合成キメラ・プライマーを用いて、dsDNAからdsDNAを増幅産物として得るICAN法(Isothermal and Chimeric primer-initiated Amplification of Nucleic acids)等が挙げられる。

Claims (11)
- 核酸増幅反応を行なう核酸増幅装置であって、
前記核酸増幅反応を行なうウェルを複数備え、
前記ウェルごとに設けられた個別に制御可能な加熱部と、
前記複数のウェル全てに特定波長の励起光を照射可能な光学手段と、
前記ウェルごとに設けられた蛍光検出部と、
を少なくとも備えた核酸増幅装置。 - 前記核酸増幅反応は、少なくともPCR(polymerase chain reaction)法により行なわれる特徴とする請求項1記載の核酸増幅装置。
- 前記核酸増幅反応は、等温で行われる請求項1または2記載の核酸増幅装置。
- 前記ウェルの加熱温度と加熱時間は、前記ウェルに対応する前記加熱部により個別に制御される請求項1から3のいずれか一項に記載の核酸増幅装置。
- 前記加熱部は、薄膜トランジスタにより形成され、スイッチング制御される請求項1から4のいずれか一項に記載の核酸増幅装置。
- 前記加熱部は、発熱抵抗体により形成され、薄膜トランジスタによりスイッチング制御される請求項1から5のいずれか一項に記載の核酸増幅装置。
- 定温制御するペルチェ素子を備える請求項1から6のいずれか一項に記載の核酸増幅装置。
- 前記光学手段は、
特定波長の励起光を発する光源と、
前記励起光を各ウェルに導入する導光板と、
を少なくとも備える請求項1から7のいずれか一項に記載の核酸増幅装置。 - 前記ウェルと前記蛍光検出部との間に、特定の波長の光を透過するフィルター膜を備える請求項1から8のいずれか一項に記載の核酸増幅装置。
- 前記ウェルと前記導光板との間に、特定の波長の光を透過するフィルター膜を備える請求項8または9記載の核酸増幅装置。
- 前記光源が、発光ダイオードであることを特徴とする請求項8から10のいずれか一項に記載の核酸増幅装置。
