Description
前記反応または検査チャンバの概ね上方に配置された、1以上の液体試薬を収容する1以上のそれぞれの貯蔵チャンバと、
貯蔵チャンバの口からの重力による液体の溢流により、選択された試薬を、貯蔵チャンバから、貯蔵チャンバの概ね下にある反応または検査チャンバ内に押出すべく、選択された貯蔵チャンバの口内に挿入できるように配置されかつ操作可能なプランジャ要素とを有することを特徴とする、反応または検査チャンバ内に1以上の液体試薬を選択的に配給する配給装置を提供する。
本発明のこの第二態様の好ましい実用的実施形態では、反応または検査チャンバおよび1以上の貯蔵チャンバは、適当な成形プラスチック材料(例えばポリプロピレン)で形成でき、好ましくはこのプラスチック材料の一体ハウジング内に収容される。ハウジングは、好ましくは上方周壁を有し、該周壁は、貯蔵チャンバの口を集合的に包囲しかつ該口を通って上方に延びており、貯蔵チャンバから溢流する液体試薬を、貯蔵チャンバの下の反応または検査チャンバ内に導くバッフルとして機能する。
各チャンバを閉じるシーリング手段は、プランジャ要素がそれぞれのチャンバの口内に移動するときに、プランジャ要素の先端部により破断または穿孔可能であることが好ましい。従って、プランジャ要素の先端部は、この目的のための適当な形状になっている。例えば、鋭い先端部をもつ円錐状にするか、その外表面にバットレス(buttresses)またはリブを設けることができる。或る実施形態では、プランジャの先端部の外表面には、プランジャがいずれかのチャンバから抜去されるときに液体試薬が排出される通路としての1以上のチャネルを設けることができる。
反応または検査チャンバの概ね上方に配置された、1以上のそれぞれの貯蔵チャンバ内に1以上の液体試薬を用意する段階と、
選択された貯蔵チャンバの口内にプランジャ要素を挿入して、貯蔵チャンバの口からの重力による液体の溢流により、貯蔵チャンバ内の選択された試薬を、該貯蔵チャンバから、貯蔵チャンバの概ね下にある反応または検査チャンバ内に押出す段階とを有することを特徴とする方法が提供される。
最初に図1を参照すると、ハウジング1は射出成形されたポリプロピレンで形成されている(ポリプロピレンは、例えばナイロンまたはポリエチレンと比較して、容易にオートクレーブできること、および水性液体が容易に付着しないことから好ましい材料である)。ハウジング1は、この下方かつ中央のウェルすなわち反応チャンバ2を収容しており、該反応チャンバ2は、使用前は空にしておくか、或いは予分配された固体または液体試薬3を入れておくこともできる。このチャンバ2内には、反応または分析を行なうための試料が添加される。鋭い円錐状先端部4aを備えた中実ロッドまたは中空杖の形態をなすプランジャ4が、制御ハードウェアおよびソフトウェア(図示せず)の制御により水平および垂直に移動できる。
ウェル36、38、39、43、44は、金属箔積層シート45によりシールされる。
次に箔シートが取除かれ、円筒状磁石30(図5)がプランジャ4内に配置され、次に該プランジャ4はウェル36から磁気ビーズを収集するのに使用される。磁石30が取外されるとビーズが解放され、ビーズは、例えば攪拌棒としてプランジャ4を使用することにより試料と完全に混合される。この間、細胞がビーズに結合する。
Claims (26)
- 1以上の液体試薬を選択的に反応または検査チャンバに配給する試薬配給システムにおいて、
反応または検査チャンバおよび1以上の液体試薬で予め満たされた1以上の貯蔵チャンバを備えた使い捨てユニットを有し、1以上の貯蔵チャンバは、閉じた底および開けることができ、反応または検査チャンバの上方に配置された口を備えており、
貯蔵チャンバの口からの重力による液体の溢流により、選択された試薬を、貯蔵チャンバから、全体として貯蔵チャンバの下にある反応または検査チャンバ内に押出すべく、選択された貯蔵チャンバの口内に挿入できるように配置されかつ操作可能なプランジャ要素を有することを特徴とするシステム。 - 1以上の貯蔵チャンバは、所定量の1以上の液体試薬で予め満たされ、各貯蔵チャンバの口は、それぞれシーリング手段によりシールされていることを特徴とする請求項1記載の試薬配給システム。
- 前記各チャンバを閉じるシーリング手段は、プランジャ要素がそれぞれのチャンバの口内に移動するときに、プランジャ要素の先端部により破断または穿孔可能であることを特徴とする請求項2記載の試薬配給システム。
- 請求項1〜3の何れか1項記載の試薬配給システムと、プランジャ要素の物理的移動を制御する制御手段とを有することを特徴とする反応、検査または検定装置。
- 食品のバクテリア汚染を検出または検定する装置、またはDNAを抽出しかつ精製する装置であることを特徴とする請求項4記載の反応、検査または検定装置。
- 前記反応または検査チャンバおよび/または貯蔵チャンバ(単一または複数)の加熱手段を更に有することを特徴とする請求項4または5記載の反応、検査または検定装置。
- 前記加熱手段は、制御される電流により加熱できる導電性ポリマーからなることを特徴とする請求項6記載の反応、検査または検定装置。
- 前記プランジャ要素は、配給すべき試薬を収容する各貯蔵チャンバ内に挿入されかつ該貯蔵チャンバから抜去できるように配置されかつ制御されることを特徴とする請求項4〜7のいずれか1項記載の装置。
- 前記プランジャ要素は中実または中空円筒状のロッドまたは杖の形態をなしており、1以上の試薬貯蔵チャンバは支持された円筒状チューブの形態をなしていることを特徴とする請求項4〜8のいずれか1項記載の装置。
- 前記プランジャ要素は、実質的に垂直な軸線に沿う方向におよび実質的に水平な平面内で移動できることを特徴とする請求項4〜9のいずれか1項記載の装置。
- 前記プランジャ要素は、抜去可能な磁石を収容できる中空ロッドの形態をなしていることを特徴とする請求項4〜10のいずれか1項記載の装置。
- 前記プランジャ要素は、使用前に配給装置をシールする上方閉じ蓋に取付けられた下方に延びるロッドとして形成され、蓋には、該蓋が装置から取外されたときにプランジャ制御手段が物理的に係合する手段が組込まれていることを特徴とする請求項4〜11のいずれか1項記載の装置。
- 前記プランジャは蓋の中心から外れた位置で蓋に取付けられ、蓋は、プランジャ制御手段が係合できるカルーセル型要素を形成しており、対称的に配置された試薬チャンバの間でのプランジャの水平移動は蓋の回転により行なわれることを特徴とする請求項12記載の装置。
- 請求項1記載の試薬配給システムに使用するための使い捨て反応または検査ユニットであって、反応または検査チャンバと、1以上の液体試薬を収容した1以上の貯蔵チャンバを有し、1以上の貯蔵チャンバは、閉じた底、および反応または検査チャンバの上方に配置された開いた口を備えていることを特徴とする使い捨て反応または検査ユニット。
- 前記反応または検査チャンバおよび1以上の貯蔵チャンバは、成形プラスチックハウジング内に収容されていることを特徴とする請求項14記載の反応または検査ユニット。
- 前記反応または検査チャンバおよび1以上の貯蔵チャンバは成形ポリプロピレンで形成されていることを特徴とする請求項14または15記載の反応または検査ユニット。
- 前記ハウジングは上方周壁を有し、該周壁は、貯蔵チャンバ(単一または複数)の口を集合的に包囲しかつ該口を通って上方に延びており、貯蔵チャンバから溢流する液体試薬(単一または複数)を、貯蔵チャンバの下の反応または検査チャンバ内に導くバッフルとして機能することを特徴とする請求項15記載の反応または検査ユニット。
- 前記反応または検査チャンバの下には、予め分配された量の液体または固体の補助試薬を収容する補助試薬チャンバが更に設けられており、該補助試薬チャンバには、反応または検査手順の所望段階で反応または検査チャンバの内容物が配給され、補助試薬チャンバの口は、使用前に反応または検査チャンバの底を形成する口自体の保護シーリング手段によりシールされることを特徴とする請求項14〜17のいずれか1項記載の反応または検査ユニット。
- それぞれの複数の試薬の配給を行なうための複数の試薬貯蔵チャンバを有し、該試薬貯蔵チャンバは、反応または検査チャンバの回りに対称的に配置され、かつ反応または検査チャンバの周囲の概ね上方に配置されていることを特徴とする請求項14〜18のいずれか1項記載の反応または検査ユニット。
- 前記検査チャンバおよび/または貯蔵チャンバ(単一または複数)は、加熱手段を更に有することを特徴とする請求項6〜13のいずれか1項記載の反応または検査ユニット。
- 前記加熱手段は導電性ポリマーからなることを特徴とする請求項20記載の反応または検査ユニット。
- 前記導電性ポリマーは検査チャンバおよび/または貯蔵チャンバ(単一または複数)に密着していることを特徴とする請求項21記載の反応または検査ユニット。
- 前記導電性ポリマーは検査チャンバおよび/または貯蔵チャンバと一体であることを特徴とする請求項21記載の反応または検査ユニット。
- 前記導電性ポリマーは検査チャンバおよび/または貯蔵チャンバの一部にオーバーモールディングされていることを特徴とする請求項23記載の反応または検査ユニット。
- 1以上の液体試薬を、検査または検定装置の反応または検査チャンバ内に選択的に配給することにより化学反応を発生させる方法において、
反応または検査チャンバの概ね上方に配置された、1以上の貯蔵チャンバ内に1以上の液体試薬をそれぞれ用意する段階を有し、該1以上の貯蔵チャンバは、閉じた底と、開けることができ、または開いた口を備えており、
選択された貯蔵チャンバの口内にプランジャ要素を挿入して、貯蔵チャンバの口からの重力による液体の溢流により、貯蔵チャンバ内の選択された試薬を、該貯蔵チャンバから、全体として貯蔵チャンバの下にある反応または検査チャンバ内に押出す段階とを有することを特徴とする方法。 - 反応、検査または検定装置にプランジャを使用する方法において、プランジャを、使い捨てユニットの、液体試薬を収容する貯蔵チャンバ内に挿入し、試薬を押出して該試薬を貯蔵チャンバの口から溢流させ、かつ重力の作用により、貯蔵チャンバの概ね下に配置された装置の反応または検査チャンバ内に流入させ、該貯蔵チャンバは、閉じた底と、開けることができ、または開いた口を備えていることを特徴とする方法。
