皮膚バリア機能は、外界からの刺激から生体を防御するとともに、皮膚を含めた生体組織が恒常的な水分を保持するために不可欠である。そのため、皮膚バリア機能が乾燥や紫外線などの外的要因、各種の疾患や加齢などの内的要因等により低下すると、外界からの刺激に対して炎症を起こしやすくなるとともに、皮膚に水分を結合できなくなり柔軟性が低下する。皮膚バリア機能が低下すると、アトピー性皮膚炎、肌荒れや皮膚炎等を生じやすくなる。また、正常な皮膚では水分を吸収する吸収機能と、吸収した水分を結合し保持する保持機能が働いているが、皮膚バリア機能が低下した皮膚では吸収機能も保持機能も低下するため、ドライスキン(乾皮症)、鱗屑、キメが粗くなる、シワ等の外観的変化となって現れる。

化粧料には保湿効果を高めるために、保湿剤が配合されることが多い。保湿剤は、通常水溶性の保湿成分と、通常油性の被膜形成成分に大別され、これらの成分を単独で、あるいは組み合わせて用いることによって、皮膚表面に被膜を形成しながら皮膚の角質水分量を増加することができる。しかしながら、皮膚バリア機能の改善効果が十分ではないために、期待した保湿効果を得ることができなかった。

【発明が解決しようとする課題】
本発明は、皮膚バリア機能の改善に優れ、ひいては高い保湿効果を有する化粧料を提供することを目的する。

【課題を解決するための手段】
本発明者らは、上記課題を解決するために鋭意研究を行なった結果、ある特定の成分とサイトカイニンをともに含有することによって、皮膚バリア機能が向上することを見出した。サイトカイニンは、植物に対しては各器官の生長調節作用という共通の作用を持っている。そして、ヒトに対する生理作用としては、サイトカイニン単独の効果としては、カイネチンにより、乾燥肌や老化した肌、いわゆるかさかさした肌を正常なしっとりとした潤いのある肌にする皮膚正常化効果のあること(特開昭4839646号公報)、カイネチンに育毛効果があること(特開平6−211632)、皮膚の成長速度と全増殖能を有意に変化させない量の6−(置換アミノ)プリンサイトカイニンが皮膚の美的外観を改善すること(特表平10−510240号公報)が知られている。また、サイトカイニンと他の成分とを組み合わせることによる効果としては、カイネチンとシコン(紫根)を化粧品製造の際に配合することにより、皮膚細胞の分裂を促進すること(特開昭60−19709号公報)等が知られていた。しかし、サイトカイニンと皮膚バリア機能との関係は知られていなかった。

本発明はかかる知見に基づいて開発されたものである。すなわち本発明は、下記(1)〜(2)に掲げる化粧料である:
(1)アラントイン、アラントイン誘導体、アロエ抽出物、グルコサミン、グルコサミン誘導体及びそれらの薬理学的に許容される塩からなる群から選ばれる少なくとも1種と、サイトカイニンを含有する化粧料、
(2)さらに、保湿成分、抗老化成分、抗酸化成分、抗炎症成分及び被膜形成成分からなる群から選択される少なくとも1種を含有する(1)に記載の化粧料、
さらに本発明は、下記(3)に掲げる美容方法である:
(3)アラントイン、アラントイン誘導体、アロエ抽出物、グルコサミン、グルコサミン誘導体及びそれらの薬理学的に許容される塩からなる群から選ばれる少なくとも1種と、サイトカイニンとを併用してなる美容方法。

【発明の実施の形態】
本発明で用いられるサイトカイニンは、カイネチンに代表される植物ホルモンまたはその誘導体のうち、カイネチン同様に植物又は植物の組織培養において、細胞分裂促進作用又は再分化促進作用の少なくともいずれかの作用を示す物質をいう。サイトカイニンは、植物組織培養の細胞増殖を促進する物質としてDNAの分解物から単離された生理活性物質であるカイネチン(6−フルフリルアミノプリン)と同様の作用を持つ物質の総称であり、プリン誘導体(カイネチン等)や尿素誘導体など多数のサイトカイニンが知られている。

本発明で用いられるサイトカイニンは、具体的には、6−フルフリルアミノプリン(別名カイネチン)、6−ベンジルアミノプリン、6−(4−ヒドロキシ−3−メチル−トランス−2−ブチニルアミノ)プリン(別名ゼアチン)、6−(3−メチル−2−ブテニルアミノ)プリン、6−(4−ヒドロキシ−3−メチルブチルアミノ)プリン(別名ジヒドロゼアチン)、6−(4−ヒドロキシ−3−メチル−トランス−2−ブテニルアミノ)−9−β−D−リボフラノシルプリン(別名リボシルトランスゼアチン)、6−メチルアミノプリン、6-n-プロピルアミノプリン、ベンゾイルアミノ−1−デアザプリン等のプリン系サイトカイニン;N,N’ジフェニル尿素,N−フェニル−N’−(4−ピリジル)尿素等の尿素系サイトカイニン;ベンジルカイネチン等のカイネチンの誘導体等が挙げられる。

なかでも好ましくは、プリン系サイトカイニンであり、特に好ましくは6−フルフリルアミノプリン(別名カイネチン)である。なお、上記のサイトカイニンは、1種単独で使用しても、また2種以上を任意に組み合わせて使用することもできる。

本発明の化粧料中に配合するサイトカイニンの含有量は、化合物の種類、剤型等によっても異なるが、通常0.001〜3.0重量%、好ましくは0.001〜1.0重量%、より好ましくは0.001〜0.5重量%の範囲を挙げることができる。

本発明の化粧料に用いられるアラントイン、アラントイン誘導体またはそれらの塩は抗炎症作用が知られている成分である。本発明の化粧料中に配合するアラントイン誘導体としては、例えば、アラントイングリチルレチン、アラントインクロルヒドロキシアルミニウムなどが挙げられる。本発明の化粧料中に配合するアラントイン又はアラントイン誘導体の含有量は、化合物の種類、剤型等によっても異なるが、通常0.001〜5.0重量%、好ましくは0.001〜3.0重量%、より好ましくは0.01〜0.5重量%の範囲を挙げることができる。

本発明におけるグルコサミン、グルコサミン誘導体またはそれらの薬学的に許容される塩は、動物、植物、微生物菌体、培養液などから精製された天然品であることが好ましいが、公知の方法により化学合成により得た合成品を利用してもよく、市販品を利用してもよい。本発明において、グルコサミン誘導体としては、生体内に適用された後にグルコサミンに変化しうる化合物が好ましく、例えば、N−アセチルグルコサミンのような誘導体が例示できる。なお、グルコサミン誘導体は本発明の効果を奏するものである限り、特にこれらに限るものではない。

本発明のグルコサミン、またはグルコサミン誘導体の塩としては、薬理学的に(製薬上)又は生理学的に許容される塩が使用できる。薬理学的又は生理学的に許容できる塩としては、例えば、有機酸塩(例えば、乳酸塩、酢酸塩、酪酸、トリフルオロ酢酸塩、フマル酸塩、マレイン酸塩、酒石酸塩、クエン酸塩、コハク酸塩、マロン酸塩、メタンスルホン酸塩、トルエンスルホン酸塩、トシル酸塩、パルミチン酸、ステアリン酸など)、無機酸塩(例えば、塩酸塩、硫酸塩、硝酸塩、臭化水素酸塩、リン酸塩など)などが例示できる。好ましくは、塩酸グルコサミン、硫酸グルコサミンである。特に好ましくは塩酸グルコサミンである。
なお、これらのグルコサミン、その誘導体またはそれらの塩は、単独で又は二種以上組み合わせて使用してもよい。

本発明の化粧料において、グルコサミン、グルコサミン誘導体またはその塩の含有量は、グルコサミンの種類、剤型等によっても異なるが、通常0.00001〜70.0重量%、好ましくは0.001〜50重量%、より好ましくは0.001〜20重量%、特に好ましくは0.01〜10重量%の範囲を挙げることができる。

本発明の化粧料において用いられるアロエ抽出物は、ユリ科アロエ族に属する植物(Aloe
ferox、Aloe spicata、Aloe barbadensis、Aloe arborescens、Aloe
africana等)の抽出物であり、原料であるアロエの葉等の抽出物は、抗炎症作用、保湿作用、美白作用が知られており、古くから医薬品や化粧料に用いられている安全性の高い抽出物である。アロエはその種類や産地により含有する成分が異なるが、本発明に用いる抽出物においては、特に限定することなく用いることができる。
アロエ抽出物の抽出方法は、特に限定されるものではないが、主として葉を原料とし、葉を乾燥、細切、圧搾等、適宜処理を施すか、または葉そのままに対して、低温もしくは加温下で溶媒により抽出する方法が例示できる。得られた抽出液は濾過またはイオン交換樹脂精製、吸着等を用いて精製し、ペースト状、ゲル状、粉末状、液状として利用することもできる。また、市販のアロエエキス等のアロエ抽出物を利用することもできる。

アロエの抽出に用いることのできる抽出溶媒としては、例えば水、低級アルコール(メチルアルコール、エチルアルコール、プロパノール、ブタノール、ブタノール等)、多価アルコール(グリセリン、1,3−ブチレングリコール等)等が挙げられる。

本発明の化粧料において、アロエ抽出物の含有量は、アロエ抽出物の乾燥固形分として、好ましくは0.00001〜10質量%であり、より好ましくは0.0001〜5質量%、特に好ましくは0.0001〜1質量%である。また、アロエエキスなどの抽出物を使用する場合は、アロエエキス等の抽出物として、好ましくは、0.001〜20質量%であり、より好ましくは0.001〜10重量%、特に好ましくは0.01〜10重量%である。

本発明の化粧料は、上記した成分にさらに界面活性成分から選択される少なくとも1種類以上を配合することができる。

界面活性成分を使用する場合、皮膚や粘膜に刺激などの影響を与えず、且つ本発明の効果を奏するサイトカイニンが製剤中により安定に保持され、その結果、本発明の化粧料がより高い効果を発揮することができることから、化粧料への含有量としては、化粧料中に0.01〜50重量%で含まれるような範囲で適宜選択して使用することができる。好ましくは0.1〜40重量%、より好ましくは0.5〜20重量%の範囲を挙げることができる。

本発明の化粧料には、外観安定性や粘度等の品質を損なわず、また本発明の効果を損なわない量的及び質的範囲内で、上記した成分以外に必要に応じて医薬品、医薬部外品または化粧品分野において外用剤の成分として一般的に用いられる各種の成分、例えば基剤、乳化剤、安定化剤、刺激軽減剤、増粘剤、防腐剤、着色剤、分散剤、pH調整剤、香料等を配合することができる。なお、これらの各成分としては、医薬品、医薬部外品、または化粧品分野において従来より使用され、また将来使用される成分を用いることができ、これらの成分は1種単独で、または2種以上を任意に組み合わせて配合することができる。

本発明の化粧料は、溶媒や通常使用される外用剤の基剤等を配合することによって、ペースト状、ムース状、ジェル状、液状、乳液状、クリーム状、シート状、エアゾール状、スプレー状などの各種所望の形態に調製することができる。また、本発明に用いられる成分は、そのまま配合してもよく、また、リポソームやマイクロスフェアなど皮膚への浸透・吸収を促進する機能体に封入した形態としたうえで製剤中に配合してもよい。そして、これらは当業界の通常の方法にて製造することができる。

本発明の化粧料は、通常pH2〜8の液性を備えていればよいが、皮膚や粘膜に対する低刺激性、及び皮膚使用感のよさという観点から、好ましくはpH3〜7、より好ましくはpH3.5〜6の弱酸性〜中性であることが望ましい。
