【発明の属する技術分野】
本発明は、真皮層の線維芽細胞によるコラーゲンの産生を活発化させる作用を有するコラーゲン産生促進剤、女性ホルモンの一種であるエストロゲンと同様の作用を有するエストロゲン様作用剤、ならびにコラーゲン産生促進作用またはエストロゲン様作用を有する皮膚化粧料および美容食品に関するものである。

【従来の技術】
人の皮膚の表皮および真皮は、表皮細胞、線維芽細胞、およびこれらの細胞の外にあって皮膚構造を支持するエラスチン、コラーゲン等の細胞外マトリックスにより構成されている。若い皮膚においては線維芽細胞の増殖は活発であり、またエラスチン、コラーゲン等の皮膚組織の相互作用が恒常性を保つことにより水分保持、柔軟性、弾力性等が確保され、肌は外見的にも張りや艶があってみずみずしい状態に維持される。

ところが、紫外線、空気の著しい乾燥、過度の皮膚洗浄等の外的因子の影響や加齢の進行により、細胞外マトリックスの主要構成成分であるエラスチンは分解・変質を起こし、またコラーゲンは産生量が減少するとともに架橋による弾力低下を起こす。これにより、皮膚の保湿機能や弾力性は低下し、角質は異常剥離を始め、その結果、肌は張りや艶を失い、荒れ、シワ、くすみ等の老化症状を呈するようになる。

上記機構による皮膚の老化を防止・改善するために最も一般的に行われているのは、天然保湿因子(NMF)である糖、アミノ酸、有機酸、ピリドンカルボン酸塩、コラーゲン、ヒアルロン酸等のムコ多糖類、グリセリン、1,3−ブチレングリコール等の保湿作用を有する物質を塗布して、皮膚の保湿性を高めることである。

しかしながら、このような保湿剤は表皮の角質の状態を改善するだけのものであって、真皮内の張力保持機構まで改善することは期待できない。また、保湿剤は皮膚からの水分蒸発を遅くするものであるから、概して使用感が悪く、長期間使用すると皮膚障害を起こすことさえある。

一方、加齢に伴う皮膚老化の一因は、女性ホルモンの一種であるエストロゲンの分泌が減退することにある。エストロゲンは成人女性の健康維持に深く関わっており、その分泌不足は種々の内科的疾患を招くほか、肌の過敏症、弾力性低下、潤いの減少等、好ましくない肌の変化の原因となることが知られている。

そこで、エストロゲンの分泌が衰える更年期以後の女性に対して、エストロゲンと同様の作用をする物質を経皮的または経口的に投与することが行われている。このようなエストロゲン様作用剤としては、従来より、ステロイド系エストロゲン、非ステロイド系エストロゲン、フラボン系化合物等が使われている。

【発明が解決しようとする課題】
本発明の第1の目的は、真皮層線維芽細胞におけるコラーゲンの産生を促進するのに有効な物質を見出し、それを有効成分としたコラーゲン産生促進剤を提供することにある。
本発明の第2の目的は、皮膚の老化防止または改善作用を有するエストロゲンと同様の作用を有する物質を見出し、それを有効成分としたエストロゲン様作用剤を提供することにある。
本発明の第3の目的は、コラーゲン産生促進作用および/またはエストロゲン様作用を有する皮膚化粧料を提供することにある。
本発明の第4の目的は、エストロゲン様作用を有する美容食品を提供することにある。

【課題を解決するための手段】
上記目的を達成するために、本発明のコラーゲン産生促進剤およびエストロゲン様作用剤は、サウスレア・インボルクラタ(Saussurea involucrata (Kar.et Kir.) Sch. Blp.)からの抽出物を有効成分として含有することを特徴とし、本発明の皮膚化粧料は、サウスレア・インボルクラタ(Saussurea involucrata (Kar.et Kir.) Sch. Blp.)から抽出されるコラーゲン産生促進作用および/またはエストロゲン様作用を有する抽出物を含有することを特徴とし、本発明の美容食品は、サウスレア・インボルクラタ(Saussurea involucrata (Kar.et Kir.) Sch. Blp.)から抽出されるエストロゲン様作用を有する抽出物を含有することを特徴とする。

なお、本明細書における「抽出物」とは、抽出処理によって得られる抽出液、抽出液の希釈液もしくは濃縮液、抽出液を乾燥して得られる乾燥物、またはこれらの粗精製物もしくは精製物のいずれをも含むものとする。

【発明の実施の形態】
以下、本発明について詳細に説明する。

〔コラーゲン産生促進剤・エストロゲン様作用剤〕
本発明のコラーゲン産生促進剤およびエストロゲン様作用剤は、それぞれキク科植物の一種であるサウスレア(Saussurea)属に属する植物の抽出物を有効成分とする。

サウスレア(Saussurea)属に属する植物としては、例えば、サウスレア・インボルクラタ(Saussurea involucrata (Kar.et Kir.) Sch.Blp.)(漢方名:雪蓮)、サウスレア・グナパロイデス(Saussurea gnaphaloides (Royle) Sch.-Bip.)(漢方名:鼠▲麹▼雪兎子)、サウスレア・ステラ(Saussurea stella Maxim.)(漢方名:星状風毛菊)、サウスレア・メデュサ(Saussurea medusa Maxim.)、サウスレア・ラニセプス(Saussurea laniceps Hand.-Mazz.)、サウスレア・トリダクタイラ(Saussurea tridactyla Sch.-Bip.ex Hook.f.)、サウスレア・ナミカワエ(Saussurea namikawae Kitam.)、サウスレア・ゴッシピポラ(Saussurea gossypiphora D.Don)、サウスレア・ニシオカエ(Saussurea nishiokae Kitam.)、サウスレア・レウコマ(Saussurea leucoma Diels)、サウスレア・クゥエルキフォリア(Saussurea quercifolia W.W.Smith)、サウスレア・エリオケパラ(Saussurea eriocephala Franch.)、サウスレア・キンギイ(Saussurea kingii J.R.Drumm.ex C.E.C.Fisch.)、サウスレア・シムプソニアナ(Saussurea simpsoniana (Field.et Gardn.) Edgew.)、サウスレア・ウェットステイニアナ(Saussurea wettsteiniana Hand.-Mazz.)、サウスレア・グロボサ(Saussurea globosa Chen)、サウスレア・ロンギフォリア(Saussurea longifolia Franch.)等が挙げられる。これらサウスレア(Saussurea)属に属する植物は、それぞれ単独で用いることもできるし、異なる種のものを混合して用いることもできる。これらサウスレア(Saussurea)属に属する植物の中でも、サウスレア・インボルクラタ(Saussurea involucrata (Kar.et Kir.) Sch.Blp.)がコラーゲン産生促進作用またはエストロゲン様作用の点、あるいは入手容易性の点で好ましい。

上記サウスレア(Saussurea)属に属する植物の部位としては、地上部分を用いるのが好ましく、特に葉および/または茎を用いるのが好ましい。また、抽出原料とするサウスレア(Saussurea)属に属する植物は、採取後ただちに乾燥し粉砕したものが適当である。乾燥は、天日で行ってもよいし、通常用いられる乾燥機を使用して行ってもよい。

抽出方法は特に限定されるものではなく、常法に従って行えばよい。
抽出溶媒としては、極性溶媒、例えば、水、メタノール、エタノール、プロパノール、1,3−ブチレングリコール、グリセリン、プロピレングリコール等の1種または2種以上の混合物を使用するのが好ましく、特に水を使用するのが好ましい。
上記極性溶媒による抽出を行う前に、ヘキサン、ベンゼン等の非極性溶媒を用いた脱脂処理を施してもよく、かかる前処理を行うことにより、極性溶媒による抽出処理を効率良く行うことができる。

抽出処理は、室温ないし還流加熱下において任意の装置を用いて行うことができる。一般的には、抽出溶媒を満たした処理槽に抽出原料の乾燥・粉砕物を投入し、適宜攪拌しながら可溶性成分を抽出溶媒に溶出させた後、濾過して抽出残差を除き、抽出液を得る。このとき、抽出温度は80〜95℃とするのが好ましく、抽出時間は1〜4時間とするのが好ましい。

得られた抽出液は、そのままの状態でもコラーゲン産生促進剤またはエストロゲン様作用剤に使用することができるが、濃縮液またはその乾燥物としたもののほうが利用しやすい。
また、サウスレア(Saussurea)属に属する植物の抽出物は特有の匂いと味を有するため、生理活性上の低下を招かない範囲で脱色、脱臭等を目的とする精製を行ってもよいが、皮膚化粧料や美容食品に使用する場合には、一般的に大量に配合するものではないから、精製を行うことなく使用しても実用上支障はない。

以上のようにして得られる抽出物(以下「サウスレア(Saussurea)抽出物」という。)は、コラーゲン産生促進作用およびエストロゲン様作用を有する。すなわち、サウスレア(Saussurea)抽出物は、経皮的に吸収されて線維芽細胞に達し、コラーゲンの産生を活発化して真皮層に十分なコラーゲンが補給されるようにする作用を有するとともに、エストロゲン分泌の衰えによる肌の老化を防止し改善する作用を有する。

上記サウスレア(Saussurea)抽出物は、本発明のコラーゲン産生促進剤またはエストロゲン様作用剤としてそのまま用いることもできるし、製剤化することもできる。製剤化する場合、保存や取扱いを容易にするために、デキストリン、シクロデキストリン等のキャリアー、その他任意の助剤を上記サウスレア(Saussurea)抽出物に加えてもよい。また、製剤化する場合の剤形に制限はなく、粉末状、顆粒状、錠剤状等、任意の剤形のものとすることができる。

本発明のコラーゲン産生促進剤は、経皮的に吸収されて真皮層の線維芽細胞
に達し、真皮層線維芽細胞におけるコラーゲンの産生を促進し、老化した皮膚を真に若返らせることができる。また、本発明のコラーゲン産生促進剤はエストロゲン様作用をも併有するため、経口的に吸収されることにより、皮膚の老化防止と改善に多面的に作用して、皮膚を内部から活性化して若々しい状態に保つことができる。

一方、本発明のエストロゲン様作用剤においても、経口的に吸収されることにより、エストロゲン分泌の衰えによる皮膚の老化を防止し、改善させることができる。

〔皮膚化粧料〕
上述したサウスレア(Saussurea)抽出物は、皮膚に適用した場合の使用感と安全性に優れているため、皮膚化粧料に配合するのに好適である。本発明の皮膚化粧料は、上記サウスレア(Saussurea)抽出物を含有し、コラーゲン産生促進作用および/またはエストロゲン様作用を有する。もちろん、製剤化した本発明のコラーゲン産生促進剤および/またはエストロゲン様作用剤を含有するものも本発明の皮膚化粧料の範囲に含まれる。

皮膚化粧料の種類としては、例えば、軟膏、クリーム、乳液、ローション、パック、入浴剤等が挙げられ、その皮膚化粧料におけるサウスレア(Saussurea)抽出物の好適な配合率は、約0.01〜10重量%である。

本発明の皮膚化粧料には、コラーゲン産生促進作用および/またはエストロゲン様作用の妨げにならない限り、皮膚化粧料の製造に通常使用される各種主剤、助剤等を配合することができる。なお、本発明の皮膚化粧料は、皮膚の老化防止・改善に関しサウスレア(Saussurea)抽出物のみが主剤となるものに限られるわけではない。

本発明の皮膚化粧料において、サウスレア(Saussurea)抽出物とともに構成成分として利用可能なものとしては、例えば、グリセリン、コラーゲン、ヒアルロン酸およびその塩、コンドロイチン硫酸およびその塩、キチン、キトサン等の保湿剤;パラジメチルアミノ安息香酸アミル等の紫外線吸収剤;グリセロリン脂質、スフィンゴリン脂質等の複合脂質;β−カロチン、油溶性甘草エキス、リコカルコンA、バイカリン、バイカレインその他の活性酸素消去作用を有する物質;アズレン、グリチルリチン酸およびその塩類、グリチルレチン酸およびその誘導体、酸化亜鉛等の抗炎症作用物質;リボフラビン、トコフェロール、アスコルビン酸、葉酸等のビタミンおよびその誘導体類;ホホバ油、ラノリン、流動パラフィン、スクワラン、イソステアリルアルコール等の油性成分;ステアリル硫酸ナトリウム、セチル硫酸ジエタノールアミン、ステアリン酸グリセリン等の界面活性剤;エリソルビン酸ナトリウム等の酸化防止剤;エチルパラベン等の防腐剤;オウバク抽出物、カミツレ抽出物、カンゾウ根抽出物、ローズマリー抽出物、マロニエ抽出物等のコレステロール類;植物ステロール類;リポプロテイン類;ビフィズス菌培養物、乳酸菌培養物、酵母抽出物、ブクリョウ抽出物等の微生物由来成分;褐藻抽出物、紅藻抽出物等の藻類抽出物;γ−オリザノール等の血行促進剤;硫黄等の抗脂漏剤;香料;アルコール;カルボキシポリマー等の増粘剤;チタンイエロー、ベニバナその他着色料等が挙げられる。

〔美容食品〕
上述したサウスレア(Saussurea)抽出物は、消化管で消化されるようなものではないことが確認されたため、食品として経口摂取するのに好適である。本発明の美容食品は、上記サウスレア(Saussurea)抽出物を含有し、エストロゲン様作用を有する。もちろん、製剤化した本発明のエストロゲン様作用剤を含有するものも本発明の美容食品の範囲に含まれる。
ここで、美容食品とは、皮膚の老化防止または改善を図ることを目的とした飲食物をいうものとする。

本発明の美容食品は、上記サウスレア(Saussurea)抽出物を、その活性を妨げないような任意の食品に配合したものであってもよいし、上記サウスレア(Saussurea)抽出物を主成分とする食品(栄養補助食品)であってもよい。
ここにいう任意の食品には、例えば、麺類、パン、米飯、餅等の穀物加工品、マーガリン、マヨネーズ等の油脂加工品、ハム、ソーセージ等の食肉加工品、かまぼこ、ちくわ等の水産加工品、ヨーグルト、バター、チーズ、アイスクリーム等の乳製品、ジャム等の果実加工品、漬物等の野菜加工品、チョコレート、クッキー、ケーキ、キャンディー、チューインガム、ゼリー等の菓子類、ジュース、コーヒー、紅茶、緑茶、ウーロン茶、炭酸飲料、牛乳等の各種飲料、醤油、ソース、みりん等の調味料など、あらゆる形態の食品が含まれる。

本発明の美容食品におけるサウスレア(Saussurea)抽出物の配合量は、当該美容食品の一般的な摂取量を考慮して、標準的なサウスレア(Saussurea)抽出物の成人1日当たりの摂取量が約1〜1000mgになるように設定するのが好ましい。

以上説明した本発明のコラーゲン産生促進剤、エストロゲン様作用剤、皮膚化粧料および美容食品は、ヒトに対して好適に適用されるものであるが、本発明の作用効果が奏される限り、ヒト以外の動物に対して適用されてもよい。

【実施例】
以下、実施例等により本発明をさらに具体的に説明するが、本発明の範囲はこれらの実施例等に限定されるものではない。

〔製造例1〕
抽出原料としてサウスレア・インボルクラタ(Saussurea involucrata (Kar.et Kir.) Sch. Blp.)の葉および茎の乾燥・粗砕物300gを、表1に示す抽出溶媒2000mlに投入し、穏やかに攪拌しながら3時間、70℃に保った後、濾過した。濾液を40℃で減圧下に濃縮し、さらに減圧乾燥機で乾燥して抽出物(粉末状)を得た。4種類の抽出溶媒を用いて上記抽出処理を行ったところ、得られた抽出物(試料a〜d)の収率は表1のとおりであった。
なお、抽出溶媒が混合物の場合、以下に示す混合比は重量基準によるものである。

製造例1で用いたものと同じ抽出原料の乾燥・粗砕物300gを1,3−ブチレングリコール−水(1:1混合液)2000mlに投入し、攪拌しながら80℃で3時間保持したのち濾過して、固形分濃度2.0重量%の抽出液(試料e)1560mlを得た。

抽出溶媒をグリセリン−水(1:1混合液)に変更したほかは製造例2と同様にして、固形分濃度1.6重量%の抽出液(試料f)1520mlを得た。

〔試験例1〕
コラーゲン産生促進作用の試験
製造例1〜4による試料a〜gについて、Websterらの方法(Anal.Biochem.,Vol.96, 220, 1979)によりコラーゲン産生促進率を求めた。具体的には、以下のようにして試験を行った。

ヒトの線維芽細胞をプレートに播種し、37℃、5%二酸化炭素下、試料添加培地(試料濃度:200ppm)で数日間培養した後、β−アミノプロピオニトリルと[3H]−プロリンとを添加し、更に24時間培養した。当該培養液全体にペプシン/酢酸溶液を加えて4℃下で16時間消化し、次いでこの消化液にキャリアーを加えて0.7M食塩で沈殿させ、更に中性条件下で再溶解させて、4.2M食塩で再沈殿させた。得られた沈殿物を20%エタノールで洗浄した後、その沈殿物の放射活性を測定した。
