Abstract
Description
本出願は、2017年9月14日に出願された米国特許仮出願第62/558,763号に対する優先権の利益を主張する米国特許出願であり、該米国特許仮出願の全体は参照により本明細書に組み込まれる。
本開示の化合物には、以下の式(I)及び/又は(II)のうちの1つ又は2つ以上の化合物が含まれる:
Xは、O又はNHであり、Yは、O−PG、−O(CR4 2)aCR4 3、−O(CR4 2)bOCR4 3(例えば、OEt、O−メトキシエトキシ及びOCF3)からなる群から選択され、Zは、Hであり、PGは、保護基であり、Bは、天然若しくは未修飾の核酸塩基又は修飾核酸塩基、あるいはそれらの保護された形態であり、各R1は、独立して、C1〜6アルキル又はシクロアルキルであり、R2は、CH2CH2CN又はC1〜6アルキルであり、あるいは1つのR1及びR2が一緒になって、任意選択により置換されたC1〜6シクロアルキルを形成し、R3は、H又はPGであり、R4は、独立して、各場合においてH又はFであり、aは、0〜2の整数(例えば、0、1又は2)であり、bは、1〜3の整数(例えば、1、2又は3)であり、Aは、−(CR’R’)1〜2−であり、R’は、独立して、各場合においてH又はMeである)。
本開示はまた、本開示の化合物と、1つ又は2つ以上の反応物質又は溶媒若しくは不純物と、を含む組成物も包含する。
本開示の組成物は、修飾ヌクレオシドを含むオリゴヌクレオチドの合成前駆体、例えば、それぞれ参照により組み込まれる米国仮出願第62/420801号、同第62/394737号、同第62/394738号及び同第62/394739号に記載されているものとして使用され得る。修飾オリゴヌクレオチドは、当該技術分野における方法により、例えば、脱保護(deblock)、カップリング及び待機工程の修正を伴って記載される93ステップサイクルを使用したABI−394シンセサイザーで合成され得る。固体担体は、3’−NHTr−5’−LCAA−CPGであり得る。各オリゴヌクレオチドは、本明細書に記載の方法を用いて個別に合成される。
本開示の組成物は、以下の実施例に記載されるものなどの合成手順によって合成し得る。
本明細書で使用される用語は、特定の実施形態を説明する目的でのみ使用され、本発明の範囲を制限することを意図しないと理解すべきである。別途記載のない限り以下の定義が適用されるものとする。
脱保護、カップリング及び待機工程の修正を伴って記載される93工程のサイクルを用いて、ABI−394合成装置で修飾オリゴヌクレオチドを合成した。固体支持体は3’−NHTr−5’−LCAA−CPGである。各オリゴヌクレオチドは、本明細書に記載の方法を用いて個別に合成される。
Claims (24)
- 式(I)で表される化合物
Xは、O又はNHであり、
Yは、O−PG、−O(CR4 2)aCR4 3、−O(CR4 2)bOCR4 3及び−O(CR4 2)b−CR4=CR4 2からなる群から選択され、
Zは、Hであり、
PGは、保護基であり、
Bは、天然若しくは未修飾核酸塩基又は修飾核酸塩基、あるいはそれらの保護された形態であり、
各R1は、独立して、C1〜6アルキル又はシクロアルキルであり、
R2は、CH2CH2CN又はC1〜6アルキルであり、
あるいは1つのR1及びR2が一緒になって、任意選択により置換されたC1〜6シクロアルキルを形成し、
R3は、H又はPGであり、
R4は、独立して、各場合においてH又はFであり、
aは、0〜2の整数であり、
bは、1〜3の整数であり、
XがOである場合、Yは、−O(CR4 2)aCR4 3、−O(CR4 2)bOCR4 3又は−O(CR4 2)b−CR4=CR4 2である]。 - Xが、NHであり、Yが、O−PG、OEt又はO−メトキシエトキシである、請求項1に記載の化合物。
- R1が、C2〜3アルキルである、請求項1又は2に記載の化合物。
- R1が、イソプロピルである、請求項1又は2に記載の化合物。
- R3が、トリチル保護基である、請求項1〜4のいずれか一項に記載の化合物。
- R3が、XがNHである場合、モノメトキシトリチル(MMTr)又はトリトリルであり、XがOである場合、4,4’−ジメトキシトリチル(DMTr)又はトリトリルである、請求項1〜4のいずれか一項に記載の化合物。
- Bが、アデニン(A)、グアニン(G)、チミン(T)、シトシン(C)、ウラシル(U)及び5−メチルシトシン(5−me−C)又はそれらの保護された形態から選択される、請求項1〜6のいずれか一項に記載の化合物。
- Bが、6−N−ベンゾイルアデノシン(ABz)、4−N−ベンゾイルシチジン(CBz)及び2−N−イソブチリルグアノシン(GiBu)から選択される、請求項1〜6のいずれか一項に記載の化合物。
- 請求項1〜8のいずれか一項に記載の化合物と、1つ又は2つ以上の反応物質若しくは溶媒又は不純物と、を含む組成物。
- 前記不純物の量が1%未満である、請求項9に記載の組成物。
- 修飾ヌクレオシドを含むオリゴヌクレオチドを製造する方法であって、請求項1〜8のいずれか一項に記載の化合物を反応させて、前記オリゴヌクレオチドを形成することを含む、方法。
- XがNHであり、ZがHである、請求項12に記載の化合物。
- R1が、C2〜3アルキルである、請求項12又は13に記載の化合物。
- R1が、イソプロピルである、請求項12又は13に記載の化合物。
- R3が、トリチル保護基である、請求項12〜15のいずれか一項に記載の化合物
- R3は、XがNHである場合、モノメトキシトリチル(MMTr)又はトリトリルであり、XがOである場合、4,4’−ジメトキシトリチル(DMTr)又はトリトリルである、請求項12〜15のいずれか一項に記載の化合物。
- Bが、アデニン(A)、グアニン(G)、チミン(T)、シトシン(C)、ウラシル(U)及び5−メチルシトシン(5−me−C)又はそれらの保護された形態から選択される、請求項12〜17のいずれか一項に記載の化合物。
- Bが、6−N−ベンゾイルアデノシン(ABz)、4−N−ベンゾイルシチジン(CBz)及び2−N−イソブチリルグアノシン(GiBu)から選択される、請求項11〜17のいずれか一項に記載の化合物。
- 請求項12〜19のいずれか一項に記載の化合物と、1つ又は2つ以上の反応物質若しくは溶媒又は不純物と、を含む組成物。
- 前記化合物が、99%以上の純度を有する、請求項20に記載の組成物。
- 修飾ヌクレオシドを含むオリゴヌクレオチドを製造する方法であって、請求項12〜19のいずれか一項に記載の化合物を反応させて、前記オリゴヌクレオチドを形成することを含む、方法。
- 式(I)で表される化合物
Bは、天然若しくは未修飾核酸塩基又は修飾核酸塩基、あるいはそれらの保護された形態であり、
Zは、Hであり、
Yは、−O(CR4 2)b−CR4=CR4 2であり、R4は、独立して、各々の場合においてH又はFであり、bは、1〜3の整数であり、
Xは、NHであり、
R3は、H又はPGであり、PGは保護基であり、
各R1は、独立して、C1〜6アルキル又はシクロアルキルであり、
R2は、CH2CH2CN又はC1〜6アルキルであり、
あるいは1つのR1及びR2が一緒になって、任意選択により置換されたC1〜6シクロアルキルを形成する]
を製造する方法であって、化合物1を還元した後に化合物2を保護して化合物3を形成することを含む、方法。
