前記方法によって得られるか、または得ることができるショ糖エステル組成物もまた提供される。
本願の範囲内で用いられる技術用語および表現には、一般に、植物および分子生物学の関連技術において一般に用いられている意味と同じ意味がそれらに付与されるものとする。以下の用語の定義のすべては、本願の全内容に適用される。語”含んでいる(comprising)”は、他の要素または段階を排除せず、不定冠詞”ある(a)”または”ある(an)”は、複数であることを排除しない。個々の段階は、特許請求の範囲に記載のいくつかの特徴の機能を満たすことができる。所定の数値または数値範囲との関係において、用語”約”、”本質的に”および”おおよそ”は、所定の数値または範囲の20%以内、10%以内、または5%、4%、3%、2%もしくは1%以内である数値または範囲のことを言う。

タバコにおけるショ糖エステル合成を研究するために、2つのモデル変種を選択する。鉄管乾燥Hicks Broadleaf (HBL) および primitive Red Russian (RR)である。National Genetic Resources Program (Germplasm Resources Information Network - (GRIN), National Germplasm Resources Laboratory, Beltsville, Maryland)カタログから入手できる初めのデータは、これらの2つの系統間のβ-メチルバレリル含有ショ糖エステル含量において異なった含量を示唆している。
葉浸出物におけるショ糖エステル組成物を、HBLおよびRR親の交雑によって得られた自殖した136のF2遺伝子型植物からそれぞれ遺伝した8のF3植物の136集団において分析する。マイクロサテライトマーカーとβ-メチルバレリル含有ショ糖エステル表現型とを関連づけるために、量的形質遺伝子座研究を行う。量的形質遺伝子座cartographerを用いてデータを解析する。β-メチルバレリル含有ショ糖エステル形質は、タバコゲノムにおける単一の遺伝子座に関連づけられる。この遺伝子座は、タバコの連鎖群15(染色体A)に位置する。
図3に記載された機能をコードするすべての潜在的遺伝子の中の選択的遺伝子標的を選択するために、トリコームにおけるタバコエクソンの転写レベルを研究する。マイクロアレイを用い、β-メチルバレリル含有ショ糖エステル産生プロフィールに関して選択されたいくつかの変種においてその発現を研究した。
NtIPMS2に対応する発現配列タグは、GT09L09(起源:Galpao[TI1068]トリコーム)およびOT05C20(起源Orinocco[TI81]トリコーム)の形のトリコームESTライブラリーから単離される。両方の分子はヌクレオチド配列が同一であり、従って、発現実験にはOT05C20 cDNAのみを用いる。コンセンサス配列に基づくNtIPMS2の第1エクソンおよび第1イントロンを用いてRNAi構築物が作られる。両方の構築物は、トリコームに特異的なCPS2pプロモーター(CPS2はのみに存在するcisアビエノール合成に関与するコパリルシンターゼ2である)またはMMVpウイルスプロモーター(MMVはミラビリスモザイクウイルスである)のいずれかを含むバイナリーベクターに導入される。それぞれのバイナリーベクターを含むアグロバクテリウム・ツメファキエンスAGL1を用いる恒久的形質転換を、オリエンタルタバコ変種Basma xanthiおよび鉄管乾燥変種K326において行う。
この実施例は、遺伝子の発現を改変するためのツールを開発するための所定のゲノムデータベースと比較して、所定の遺伝子配列内の唯一の標的部位の存在をスクリーニングするためのイソプロピルリンゴ酸シンターゼ遺伝子の検索法を例示する。本開示の方法によって同定される標的部位、配列モチーフ、植物、例えばタバコにおける対応する遺伝子配列の改変における部位またはモチーフのいずれかの使用は本開示に含まれる。
ジンクフィンガーDNA結合ドメインは、3塩基対ヌクレオチド配列を認識する。ジンクフィンガーヌクレアーゼは、1、2、3、4、5、6以上のジンクフィンガーDNA結合ドメインおよびIIS型制限酵素の非特異的ヌクレアーゼを含むジンクフィンガータンパク質を含む。ジンクフィンガーヌクレアーゼは、標的配列に二本鎖切断を導入するために用いることができる。二本鎖切断を導入するためには、一対のジンクフィンガーヌクレアーゼであって、1つが標的配列のプラス(上)鎖に結合し、もう一つが0、1、2、3、4、5、6以上ヌクレオチド離れた同じ標的配列のマイナス(下)鎖に結合する前記ジンクフィンガーヌクレアーゼが必要である。2つの固定した長さの部分文字列DNAモチーフのそれぞれに関して、3の複数用いることによって、所定のスペーサー長さによって離された2つのジンクフィンガータンパク質標的部位を同定するために、このプログラムを用いることができる。

Claims (15)
- 変異植物、天然に存在しない植物またはトランスジェニック植物細胞であって、
(i)イソプロピルリンゴ酸シンターゼをコードし、かつ配列番号1もしくは配列番号10もしくは配列番号12もしくは配列番号14に少なくとも60%の配列同一性を有する配列を含むか、それからなるか、または本質的にそれからなる、少なくとも1つのポリヌクレオチド;または
(ii)前記ポリヌクレオチド(単数または複数)によってコードされるポリペプチド;または(iii)配列番号2もしくは配列番号11もしくは配列番号13もしくは配列番号15に少なくとも60%の配列同一性を有するポリペプチド;または
(iv)前記ポリヌクレオチド配列(単数または複数)を含む構築物、ベクターもしくは発現ベクターであって、場合により、配列番号8に示す配列もしくはそれに少なくとも約60%の同一性を示すそのバリアントを含むか、それからなるか、または本質的にそれからなるプロモーターまたはトリコームプロモーターまたは天然に存在するイソプロピルリンゴ酸シンターゼプロモーターをさらに含む前記構築物、ベクターもしくは発現ベクター

(ii)それらからショ糖エステル(単数または複数)を抽出する段階;および
(iii)場合により、抽出されたショ糖エステル(単数または複数)を単離または精製する段階

- タバコまたはタバコ製品のフレーバーを調節する方法であって、(i)タバコもしくはタバコ製品に、請求項2、4もしくは5のいずれかに記載の変異植物、天然に存在しない植物もしくはトランスジェニック植物からの植物の一部分、好ましくは葉または請求項6に記載の植物材料を加えること;または(ii)タバコもしくはタバコ製品に、請求項8もしくは9の方法によって得られるか、または得ることができるショ糖エステルを含む組成物を加えることを含む前記方法。
- β-メチル吉草酸を製造する方法であって、
(i)請求項2、4もしくは5のいずれかに記載の変異植物、天然に存在しない植物もしくはトランスジェニック植物の少なくとも一部分、請求項6に記載の植物材料または請求項7に記載のタバコ製品を提供する段階;
(ii)段階(i)で提供される材料またはその抽出物を加水分解する段階;および
(iii)場合によりβ-メチル吉草酸を単離または精製する段階
を含む前記方法。 - イソプロピルリンゴ酸シンターゼをコードし、かつ配列番号1または配列番号10または配列番号12または配列番号14に少なくとも60%の配列同一性を有する配列を含むか、それからなるか、または本質的にそれからなる、単離されたポリヌクレオチド。
- 請求項12に記載のポリヌクレオチドによってコードされる単離されたポリペプチド。
- 配列番号2または配列番号11または配列番号13または配列番号15に少なくとも60%の配列同一性を有する単離されたポリペプチド。
- 請求項12に記載の単離されたポリヌクレオチド配列を含む構築物、ベクターまたは発現ベクターであって、場合により、配列番号9に記載の配列もしくはそれに少なくとも約60%の同一性を有するそのバリアントを含むか、それからなるか、もしくは本質的にそれからなるプロモーターまたはトリコームプロモーターまたは天然イソプロピルリンゴ酸シンターゼプロモーターをさらに含む前記構築物、ベクターまたは発現ベクター。
