Abstract
Description
リパーゼが有機媒体中でエステルの形成を触媒し得ることは周知である。したがって、リパーゼを用いて、十分な遊離ヒドロキシ置換化合物の存在下でカルボン酸をエステルに変換することができる。
本発明の範囲に関して、パタチンは、天然ジャガイモタンパク質単離物の高分子量(HMW)画分で、ジャガイモタンパク質の約40〜50 wt.%を構成する、30 kDaまたはそれ以上、好ましくは35 kDaまたはそれ以上、およびより好ましくは約43 kDaの分子量、ならびに5.8未満、好ましくは4.8〜5.5の間の等電点を有する糖タンパク質の高度に相同的なファミリーを意味すると理解される。パタチンは、アシルヒドロラーゼ反応性を示す糖タンパク質のファミリーであり、ジャガイモ塊茎中の全可溶性タンパク質の40 wt %までを占める。出願WO2008/069650において、ジャガイモ果汁(PFJ)またはジャガイモ果実水(PFW)からのパタチンの単離の入念な説明が記載されており、これは参照により本明細書に組み入れられる。
・短鎖脂肪酸(SCFA)はC6よりも短い脂肪酸である。
・中鎖脂肪酸(MCFA)はC6〜C12脂肪酸である。
・長鎖脂肪酸(LCFA)はC13〜C21脂肪酸である。
・超長鎖脂肪酸(VLCFA)はC22よりも長い脂肪酸である。
多くの脂質分解プロセスは、有機溶媒中またはそのような溶媒の水性混合物中で行われる。主要な例は、トリグリセリドまたは遊離脂肪酸からのバイオディーゼルの生成である。これらの場合、低MWアルコールが、脂肪酸アルキルエステル(FAAE)を形成するためのアシルアクセプターとして用いられる。アルコールは、反応物と溶媒の両方を兼ねる。得られるバイオディーゼルの価格および品質の理由で、メタノールが好ましいアルコールである。
脱塩水100μL中10 mgのパタチン(Solanic 206P)を、5 mmol(1.0 g)のラウリン酸(Merck 8.05333)、ならびに当モル量のメタノール(Merck 1.06007)、エタノール(Prolabo 83804.360)、n-プロパノール(Alfa Aesar A19902)、イソプロパノール(Prolabo 437423R)、またはn-ブタノール(Merck 1.01990)のいずれかと接触させた。脱塩水を添加して、20%(v:v)アルコール水溶液を作製した。この混合物を、激しい撹拌下において45℃で一晩インキュベートした。
オクタン酸(Merck 8.00192.1000)および20%メタノール(Merck liChrosolv 1.06007.1000)をイソオクタン(SigmaAldrich 360597)中に混合して、オクタン酸とメタノールがおよそ等モル量となるようにした(イソオクタン中にオクタン酸11.0 mLおよび20%メタノール16.8 mL)。500 mgのパタチン(Solanic 206P)および40 mgの水を添加した。パタチンを除外すること以外は同様の方法で、ブランク試料を調製した。反応は閉塞チューブ中で行い、閉蓋を備えた水浴中でチューブを加熱した。溶液を50 Cで4時間インキュベートした。反応容器から分割量を採取し、純粋アセトニトリル(Merck LiChrosolv 1.00030.25007)で10倍希釈し、HPLCによって解析した。HPLCに逆相シリカゲルカラム(C18セファロース)、202 nmで作動するUV検出器(Knauer Smartline UV検出器2600)、および勾配ミキサー(Smartline Manager 5000溶離液ミキサー)を取り付けた。アセトニトリル勾配をHPLC実験に適用した。
異なるアシル鎖長のリン脂質を、0.5%アラビアゴムおよび100 mM Triton X100を含む水溶液中に、30分間超音波処理することによって分散させた。100 mgまでの量のパタチンを添加し、水酸化ナトリウムの消費を、pH 7.5で作動するpHスタット設定によりモニターした。
オクタン酸(Merck 8.00192.1000)の1 mL分割量を、脱塩水または飽和塩化カリウム(SigmaAldrich P9311)溶液のいずれかと混合し、50℃まで短時間加熱した。冷却時に、オクタン酸‐塩溶液混合物は、有機相中に濁りおよび界面上に石鹸カスを示したのに対して、オクタン酸‐水混合物は、透明でかつ石鹸のないままであった。濁りは部分沈殿を示す。結果から、塩の存在が遊離脂肪酸の取り扱いをより困難にすることが示される。
Claims (12)
- 脂肪酸アルキルエステルを選択的に調製するための方法であって、1種または複数種のC4〜C26脂肪酸と、1種または複数種のC1〜C3アルコールと、天然パタチンとを含む液体媒体を提供する段階、および該1種または複数種の脂肪酸を該1種または複数種のアルコールと反応させて脂肪酸アルキルエステルを得る段階を含む、方法。
- パタチンが液体媒体中に自由に懸濁されている、請求項1に記載の方法。
- アルコールがメタノールおよび/またはエタノールであり、脂肪酸がC4〜C12脂肪酸であり、かつ脂肪酸アルキルエステルがC4〜C12脂肪酸のメチルエステルまたはエチルエステルである、請求項1または2に記載の方法。
- 媒体が本質的に、短鎖または中鎖脂肪酸を含む脂肪酸供給原料、およびメタノールまたはエタノールのみを含む、請求項1〜3のいずれか一項に記載の方法。
- 媒体が融解している、請求項4に記載の方法。
- アルコールがメタノールである、前記請求項のいずれか一項に記載の方法。
- バイオディーゼルを生成するための方法であって、パタチンを、1種または複数種のC4〜C26脂肪酸を含む脂肪酸供給原料および1種または複数種のC1〜C3アルコールと混合する段階、ならびに該1種または複数種の脂肪酸を該1種または複数種のアルコールと反応させる段階を含む、方法。
- パタチンが供給原料中に自由に懸濁される、請求項1に記載の方法。
- アルコールがメタノールおよび/またはエタノールであり、脂肪酸がC4〜C12脂肪酸であり、かつ脂肪酸アルキルエステルがC4〜C12脂肪酸のメチルエステルまたはエチルエステルである、請求項7または8に記載の方法。
- 供給原料が本質的に、短鎖または中鎖脂肪酸を含む脂肪酸供給原料、およびメタノールまたはエタノールのみを含む、請求項7〜9のいずれか一項に記載の方法。
- 供給原料が融解している、請求項10に記載の方法。
