Abstract
Description
本出願は、2011年9月15日出願の米国出願第61/535,086号(参照によりその全体が本願明細書中に組み込まれる)の優先権を主張する。
本願明細書と同時提出され、かつ、以下の通り識別されるコンピューター可読のヌクレオチド/アミノ酸の配列表が、本明細書中、参照によりその全体が組み込まれる:2012年8月22日付ファイル名「710922ST25.TXT」、52,162バイトのASCII(テキスト)ファイル1件。
本発明は、a)HLA−A1に関連してメラノーマ抗原ファミリーA(MAGE A)−3、又は、b)HLA−Cw7に関連してMAGE−A12に対しする抗原特異性を有する、単離又は精製されたT細胞受容体(TCR)を提供する。本発明は、関連するポリペプチド及びタンパク質並びに関連する核酸、組換え発現ベクター、宿主細胞及び細胞集団をさらに提供する。本発明のTCRに関連する抗体、又はその抗原結合部分、及び、医薬組成物が、本発明によってさらに提供される。
本発明の実施形態は、a)HLA−A1に関連してメラノーマ抗原ファミリーA(MAGE A)−3(MAGE−3としても知られている)、又は、b)HLA−Cw7に関連してMAGE−A12(MAGE−12としても知られている)に対する抗原特異性を有するT細胞受容体(TCR)を提供する。
本実施例は、T細胞クローンからのTCR遺伝子のクローニングと、TCRコンストラクトの生成を実証する。
本実施例は、HLA−A1+/MAGE−A3+細胞に反応した、抗MAGE−A3 TCR−A10(配列番号13及び14)ならびに抗MAGE−A3 TCR 13−18(配列番号24及び25)を発現する細胞の反応性を実証している。
本実施例は、HLA−Cw*07+/MAGE−A12+細胞との共培養に反応した、抗MAGE−A12 TCR 502(配列番号34及び35)又は抗MAGE−A12 TCR FM8(配列番号44及び45)を発現する細胞の反応性を実証している。
本実施例は、本発明のTCRを使用して治療され得る人口を実証している。
本実施例は、MAGEファミリー由来の、様々なタンパク質のペプチドでパルスした、HLA−Cw*0701又はHLA−Cw*0702を発現する標的細胞との共培養に反応した、TCR502又はTCR FM8を発現する細胞の反応性を実証している。
本実施例は、抗MAGE−A3及び抗MAGE−A12 TCRの特異性を実証している。
本実施例は、抗MAGE−A3及び抗MAGE−A12 TCRの特異性を実証している。
本実施例は、形質導入したT細胞の反応性を実証している。
Claims (35)
- a)HLA−A1に関連してメラノーマ抗原ファミリーA(MAGE A)−3、又は、b)HLA−Cw7に関連してMAGE−A12に対する抗原特異性を有する、単離又は精製されたT細胞受容体(TCR)。
- a)EVDPIGHLY(配列番号2)を含むMAGE−A3エピトープ、又は、b)VRIGHLYIL(配列番号4)を含むMAGE−A12エピトープに対する抗原特異性を有する、請求項1の単離又は精製されたTCR。
- 配列番号5〜10、16〜21、26〜31、又は36〜41のアミノ酸配列を含む、請求項2の単離又は精製されたTCR。
- 配列番号11〜12、22〜23、32〜33、又は42〜43のアミノ酸配列を含む、請求項3の単離又は精製されたTCR。
- 配列番号13〜14、24〜25、34〜35、又は44〜45のアミノ酸配列を含む、請求項4の単離又は精製されたTCR。
- 請求項1〜5のいずれかのTCRの機能部分を含む単離又は精製されたポリペプチドであって、機能部分が、配列番号5〜10、16〜21、26〜31、又は36〜41のアミノ酸配列を含む、ポリペプチド。
- 部分が、配列番号11〜12、22〜23、32〜33、又は42〜43のアミノ酸配列を含む、請求項6の単離又は精製されたポリペプチド。
- 部分が、配列番号13〜14、24〜25、34〜35、又は44〜45のアミノ酸配列を含む、請求項7の単離又は精製されたポリペプチド。
- 請求項6〜8のいずれかのポリペプチドのうち少なくとも1つを含む、単離又は精製されたタンパク質。
- 配列番号5〜7、16〜18、26〜28、又は36〜38のアミノ酸配列を含む第一のポリペプチド鎖;および
配列番号8〜10、19〜21、29〜31、又は39〜41のアミノ酸配列を含む第二のポリペプチド鎖を含む、請求項9の単離又は精製されたタンパク質。 - 配列番号11、22、32、又は42のアミノ酸配列を含む第一のポリペプチド鎖;および
配列番号12、23、33、又は43のアミノ酸配列を含む第二のポリペプチド鎖を含む、請求項10の単離又は精製されたタンパク質。 - 配列番号13、24、34、又は44のアミノ酸配列を含む第一のポリペプチド鎖;および
配列番号14、25、35、又は45のアミノ酸配列を含む第二のポリペプチド鎖を含む、請求項11の単離又は精製されたタンパク質。 - タンパク質が融合タンパク質である、請求項9〜12のいずれか1項のタンパク質。
- タンパク質が組換え抗体である、請求項9〜13のいずれか1項のタンパク質。
- 請求項1〜5のいずれか1項のTCR、請求項6〜8のいずれか1項のポリペプチド、又は請求項9〜14のいずれか1項のタンパク質をコードするヌクレオチド配列を含む、単離又は精製された核酸。
- 配列番号46〜49のいずれかを含むヌクレオチド配列を含む、請求項15の核酸。
- 請求項16の核酸のヌクレオチド配列に対して相補的なヌクレオチド配列を含む、単離又は精製された核酸。
- 請求項15〜17のいずれか1項の核酸のヌクレオチド配列に対し、ストリンジェントな条件下でハイブリダイズするヌクレオチド配列を含む、単離又は精製された核酸
- 請求項15〜18のいずれか1項の核酸を含む、組換え発現ベクター。
- 請求項19の組換え発現ベクターを含む、単離された宿主細胞。
- 細胞が末梢血リンパ球(PBL)である、請求項20の単離された宿主細胞。
- PBLがT細胞である、請求項21の単離された宿主細胞。
- 細胞が、腫瘍浸潤リンパ球(TIL)である、請求項20の単離された宿主細胞。

- 請求項1〜5のいずれか1項のTCRの機能部分に特異的に結合する抗体、又はその抗原結合部分であって、機能部分が、配列番号5〜10、16〜21、26〜31、又は36〜41のアミノ酸配列を含む、抗体、又はその抗原結合部分。
- 請求項1〜5のいずれかのTCR、請求項6〜8のいずれかのポリペプチド、請求項9〜14のいずれかのタンパク質、請求項15〜18のいずれかの核酸、請求項19の組換え発現ベクター、請求項20〜23のいずれかの宿主細胞、請求項24の細胞集団、請求項25の抗体又はその抗原結合部分、および医薬的に許容される担体を含む、医薬組成物。
- 宿主における癌の存在を検出する方法であって:
(i) 癌細胞を含む試料を、請求項1〜5のいずれか1項のTCR、請求項6〜8のいずれか1項のポリペプチド、請求項9〜14のいずれか1項のタンパク質、請求項15〜18のいずれか1項の核酸、請求項19の組換え発現ベクター、請求項20〜23のいずれか1項の宿主細胞、請求項24の細胞集団、請求項25の抗体又はその抗原結合部分を接触させることによって、複合体を形成すること、および
(ii) 複合体を検出すること
を含み、
複合体の検出が、宿主における癌の存在を表す、方法。 - 宿主における癌の治療又は予防の方法であって、宿主に、請求項1〜5のいずれか1項のTCR、請求項6〜8のいずれか1項のポリペプチド、請求項9〜14のいずれか1項のタンパク質、請求項15〜18のいずれか1項の核酸、請求項19の組換え発現ベクター、請求項20〜23のいずれか1項の宿主細胞、請求項24の細胞集団、請求項25の抗体若しくはその抗原結合部分、又は請求項26の医薬組成物を、宿主における癌を治療又は予防するのに有効な量で投与することを含む、方法。
- 癌が、メラノーマ、乳癌、白血病、甲状腺癌、胃癌、膵臓癌、肝臓癌、肺癌、卵巣癌、多発性骨髄腫、食道癌、腎臓癌、頭部癌、頸部癌、前立腺癌、又は尿路上皮癌である、請求項27又は28の方法。
- 宿主細胞が、宿主に対して自己の細胞である、請求項27〜29のいずれか1項の方法。
- 集団の細胞が、宿主に対して自己の細胞である、請求項27〜29のいずれか1項の方法。
- 癌の治療又は予防において使用するための、請求項1〜5のいずれか1項のTCR、請求項6〜8のいずれか1項のポリペプチド、請求項9〜14のいずれか1項のタンパク質、請求項15〜18のいずれか1項の核酸、請求項19の組換え発現ベクター、請求項20〜23のいずれか1項の宿主細胞、請求項24の細胞集団、請求項25の抗体若しくはその抗原結合部分、又は請求項26の医薬組成物。
- 癌が、メラノーマ、乳癌、白血病、甲状腺癌、胃癌、膵臓癌、肝臓癌、肺癌、卵巣癌、多発性骨髄腫、食道癌、腎臓癌、頭部癌、頸部癌、前立腺癌、又は尿路上皮癌である、請求項32のTCR、ポリペプチド、タンパク質、核酸、組換え発現ベクター、宿主細胞、細胞集団、抗体若しくはその抗原結合部分、又は医薬組成物。
