Abstract
Description
− 油性物質を、本発明の系を形成することによって前記油性物質と相互作用することが可能な糖の水溶液または懸濁液に加えることからなる工程と、
− 15〜40℃、好ましくは18〜37℃、より詳細には20〜30℃、特に20〜25℃の温度で媒体を適度に攪拌することからなる工程と形成された系を回収するからなる工程と
を包含することを特徴とする。
第1の工程では、全質量の67〜82%を表す水性相に出来るだけ溶解されたシクロデキストリン(α−CD)(3〜6%m/m)がフラスコに導入される。ダイズ油から形成された油性相(15〜30%m/m)が水の表面に加えられる。水性相のpHはpH2〜9.3に調整され得る。カプセル封入されるべき分子が2相のうちの1つに加えられ得る。:水溶性分子は水性相に加えられ得、脂溶性分子は、油性相に加えられ得る。次いで、フラスコは付栓され、その後、サーモスタット水浴中(28℃)において200rpmの速度で攪拌に付される(Rotatest,Bioblock Scientific)。約0.5〜30日の期間の後、ただしより一般的には2〜3日の後、白色の多少の球状のビーズが生ずる。いくつかの中間体状態(流動次いで固形状態)がビーズの形成に先行する。ビーズ形成の速度論は、試験された条件下では、酸性のpHではより遅い。9.5〜10.3のpHでは、相は固形のままである。
有効成分を含むビーズの経口投与の観点において、凍結乾燥されたビーズおよび凍結乾燥されないビーズの安定性の試験が消化液をシミュレートする媒体中で行われ、37℃で攪拌に付された(胃媒体(pH1.2);腸(pH6.8):American Pharmacopoeia USP XXIIIによって記載された媒体)。
興味ある分子として、治療において活性な分子または化粧品(例えば色素または染料、ビタミンEアセテート、ベンゾフェノンまたはイソトレチノイン(isotretinoin))に用いられ得る分子を用いた他は、実施例1に記載されたような手順が実行された。
シクロデキストリン(α)を含む水性相が油性相によりタービン攪拌機を用いて乳化された他は、実施例1に記載されたような手順が実行された。次いで、得られた混合物は、実施例1に記載された攪拌条件に付された。
Claims (18)
- 油性物質および糖から創り出され、結晶構造のスタッキングに相当する実質的に組織化された集合物を形成することを特徴とするマイクロカプセル封入系。
- 六方晶系またはプソイド六方晶系型の結晶構造の形態の組織を示すことを特徴とする請求項1に記載の系。
- 糖が多糖および/またはオリゴ糖および/またはデンプン粉および/またはそれらの誘導体であることを特徴とする請求項1または2に記載の系。
- オリゴ糖がシクロデキストリンであることを特徴とする請求項3に記載の系。
- シクロデキストリンがα−シクロデキストリンであることを特徴とする請求項4に記載の系。
- 油性物質が脂肪酸、モノグリセリド、ジグリセリドまたはトリグリセリドであることを特徴とする請求項1〜5のいずれか1つに記載の系。
- 油性物質がダイズ油、小麦麦芽油、アボガド油またはスウィートアーモンド油等の植物油、オナジャ油等の動物油または合成油または鉱油であることを特徴とする請求項6に記載の系。
- 油性物質が分散された状態にあり、および/または、例えばシクロデキストリン、特にα−シクロデキストリンとの包摂複合体の形態であることを特徴とする請求項1〜7のいずれか1つに記載の系。
- 1以上の興味ある物質をさらに含むことを特徴とする請求項1〜8のいずれか1つに記載の系。
- 興味ある物質が水溶性物質または脂溶性物質であることを特徴とする請求項9に記載の系。
- 前記物質は特に低用量で治療的に活性であることを特徴とする請求項10に記載の系。
- 前記物質は化粧品分野に用いられ得ることを特徴とする請求項10に記載の系。
- 高密度構造の固体ビーズの形態で提供されることを特徴とする請求項1〜12のいずれか1つに記載の系。
- 1ミクロン〜数センチメートル、特に0.1〜8mm、さらには0.1〜5mm、特に0.5〜3mmの粒子サイズを特徴とする請求項13に記載の系。
- 固形相の形態で提供されることを特徴とする請求項1〜14のいずれか1つに記載の系。
- 懸濁液中のビーズ、水性または非水性液体中またはゲル中に再分散されてもされなくてもよい乾燥されたまたは凍結乾燥されたビーズの形態で提供されることを特徴とする請求項1〜15のいずれか1つに記載の系。
- 請求項1〜16のいずれか1つに記載の系を製造する方法であって、
油性物質を、結晶構造、特に六方晶系またはプソイド六方晶系型の構造のスタッキングに相当する実質的に組織化された系を形成することによって該油性物質と相互作用することが可能な糖の水性溶液または懸濁液に加えることからなる工程と、
15〜40℃、好ましくは18〜37℃、より好ましくは20〜30℃、特には20〜25℃の温度で媒体を適度に攪拌することからなる工程と形成された系を回収することからなる工程と
を包含することを特徴とする方法。 - 高密度構造、または固形または流動相の固体ビーズを得ることを可能にする速度および継続時間の条件下に攪拌が実行されることを特徴とする請求項17に記載の方法。
