海泥1kgに、精製水3Lを加え、100℃で1時間加熱後、ろ過した。ろ液を、減圧濃縮後、凍結乾燥し、水を除去し、海泥熱水抽出物した。このときの海泥に対する収率は4%であった。また、海泥熱水抽出物のチロシナーゼ阻害活性は、2mg/mLの濃度で74%であった。

(2)(1)で得た海泥熱水抽出物を水で溶解し、シリカゲル〔シリカゲル60N(63−210μm;関東化学株式会社製)〕カラムクロマトグラフィー(直径6cm、長さ50cm)にて、クロロホルム/メタノール/水〔6:4:1(V/V)〕混合溶媒を用いてまず溶出し、各画分の溶出液は、これの一部をTLCで、クロロホルム/メタノール/水〔6:4:1(V/V)〕混合溶媒を用いて展開した。ニンヒドリン試薬で呈色したときのRf値が0.6〜1のものを画分a〔海泥熱水抽出物に対する収率(以下、単に収率と略称する。):0.8%〕、Rf値が0.5〜0.6のものを画分b(収率:6.56%)及びRf値が0.1〜0.2のものを画分c(収率:43.9%)として得た。各画分の1mg/mLの水溶液濃度のチロシナーゼ阻害率は、それぞれ、40.0%、58.6%、63.3%であった。次いで上記シリカゲルクロマトグラフィーをメタノールで溶出する。該メタノールで溶出される溶出液を回収し、画分d(収率:20.8%)とした。該画分dの1mg/mLの濃度のチロシナーゼ阻害率は89.%であった。なお、画分a及び画分bについて、さらにODSカラムクロマトグラフィー、及びシリカゲルカラムクロマトグラフィー〔溶出液:ヘキサン/酢酸エチレン=1:2(V/V))で分離精製を試み、3−ヒドロキシブタン酸を分取した。3−ヒドロキシブタン酸100μg/mLのチロシナーゼ阻害率は約40%であった。 (2) The sea mud hot water extract obtained in (1) is dissolved in water, and silica gel [silica gel 60N (63-210 μm; manufactured by Kanto Chemical Co., Ltd.)] column chromatography (diameter 6 cm, length 50 cm). (4) チロシナーゼ阻害活性の高い画分d3は、さらにシリカゲルクロマトグラフィー(直径2cm、長さ15cm)にて、クロロホルム/メタノール/水〔5:4:1(V/V)〕混合溶媒を用いて溶出した。溶出液は、TLCで、クロロホルム/メタノール/水〔2:4:1(V/V)〕混合溶媒を用いて展開し、ニンヒドリン試薬で呈色により3つの画分を得た。それぞれ画分d3−1(ニンヒドリン試薬で呈色しない画分、収率:0.04%)、d3−2(Rf値0.3、収率:0.11%)、d3−3(Rf値0.2及び0.4、収率:0.046%)とした。各画分の100μg/mLのチロシナーゼ阻害率はそれぞれ、33%、45.2%及び90.6%であった。
ついで、溶出液をクロロホルム/メタノール/水〔4:4:1(V/V)〕混合溶媒に換えさらに溶出する。クロロホルム/メタノール/水〔4:4:1(V/V)〕混合溶媒で溶出される画分を回収し、画分d3−4とした。画分d3−4の収率は0.77%で、100μg/mLのチロシナーゼ阻害率は84.8%であった。
海泥熱水抽出物を精製し、100μg/mLのチロシナーゼ阻害率が80%以上である
海泥エキスである画分d3−3及び画分d3−4を得た。これら海泥エキスは、海泥熱水抽出物に比較して約1/20の量で、海泥熱水抽出物と同等のチロシナーゼ阻害活性を有するまで精製されたことが分かった。

The yield of fraction d3-4 was 0.77%, and the tyrosinase inhibition rate at 100 μg / mL was 84.8%.
The sea mud hot water extract was purified to obtain fraction d3-3 and fraction d3-4, which are sea mud extracts having a tyrosinase inhibition rate of 100% or more at 80% or more. It was found that these sea mud extracts were purified to have a tyrosinase inhibitory activity equivalent to that of the sea mud hot water extract in an amount of about 1/20 that of the sea mud hot water extract.
