Claims (1)
- 【特許請求の範囲】 1.増幅されたプラスミドベクターであって、反復DNA整列(array)からなる 配列を含み、該整列が、反復DNAユニットを含み、ここで、該整列が約20キロ塩 基長より大きい、ベクター。 2.前記DNAがセントロメアDNAである、請求項1に記載のベクター。 3.前記セントロメアDNAがαサテライトDNAである、請求項2に記載のベクタ ー。 4.前記αサテライトDNAの前記整列が100kb長より長い、請求項3に記載のベ クター。 5.前記αサテライトDNAの前記整列が140kb長より長い、請求項4に記載のベ クター。 6.前記αサテライトDNAがヒトαサテライトDNAである、請求項2に記載のベ クター。 7.請求項1〜6のいずれか一項に記載のベクターで安定に形質転換された、 宿主細胞。 8.前記宿主細胞が原核生物細胞である、請求項7に記載の宿主細胞。 9.前記原核生物細胞がE.coliである、請求項8に記載の宿主細胞。 10.前記プラスミドベクターがF因子ベースである、請求項1に記載のプラ スミドベクター。 11.前記プラスミドベクターがBACベクターである、請求項10に記載のプ ラスミドベクター。 12.請求項1〜6、10、および11のいずれか一項に記載のプラスミドベ クターを増幅する方法であって、 (a)該ベクターを宿主細胞に導入する工程;および (b)該ベクターを該宿主細胞中で複製させる工程、 を包含する、方法。
