Claims (1)
- 【特許請求の範囲】 1.式I: 〔式中、 R1は、 (a)CH3、 (b)NH2、 (c)NHC(O)CF3、 (d)NHCH3 から成るグループから選択され; Arは、モノ−、ジ−またはトリ置換フェニルまたはピリジニル(またはその N−オキシド)を表し、置換基は、 (a)水素、 (b)ハロ、 (c)C1-4アルコキシ、 (d)C1-4アルキルチオ、 (e)CN、 (f)C1-4アルキル、 (g)C1-4フルオロアルキル から成るグループから選択され; R2は、 (a)F、Cl、Br、I、 (b)CN、 (c)アジド から成るグループから選択される〕の化合物の製造方法であって、 式A1: の化合物を、酸性条件下及び任意に非反応性溶媒の存在下及びアンモニウム試薬 の存在下で化合物A2: と縮合させて、式Iの化合物を製造する段階から成る方法。 2.非反応性溶媒が酢酸である請求項1に記載の方法。 3.Arがモノ−またはジ−置換された3−ピリジニルである請求項1に記載の 方法。 4.R1がCH3またはNH2である請求項1に記載の方法。 5.Arがモノ−またはジ−置換された3−ピリジニルであり、置換基が、 (a)水素、 (b)ハロ、 (c)C1-3アルコキシ、 (d)C1-3アルキルチオ、 (e)C1-3アルキル、 (f)CF3、及び、 (g)CN から成るグルーブから選択される請求項1に記載の方法。 6.R1がCH3またはNH2であり、 Arがモノ−またはジ−置換された3−ピリジニルであり、置換基が、 (a)水素、 (b)ハロ、 (c)C1-3アルキル、 (d)CF3、及び、 (e)CN から成るグループから選択される請求項1に記載の方法。 7.R2がClであり、 R1がCH3またはNH2であり、 Arがモノ−またはジ−置換された3−ピリジニルであり、置換基が、水素及び C1-3アルキルから成るグループから選択される請求項1に記載の方法。 8.炎症及びその他のシクロオキシゲナーゼ−2介在疾患の治療に有効な式I: 〔式中、 R1は、 (a)CH3、 (b)NH2、 (c)NHC(O)CF3、 (d)NHCH3 から成るグループから選択され; Arは、モノ−、ジ−またはトリ置換フェニルまたはピリジニル(またはその N−オキシド)を表し、置換基は、 (a)水素、 (b)ハロ、 (c)C1-4アルコキシ、 (d)C1-4アルキルチオ、 (e)CN、 (f)C1-4アルキル、 (g)C1-4フルオロアルキル から成るグループから選択され; R2は、 (a)F、Cl、Br、I、 (b)CN、 (c)アジド から成るグループから選択される〕の化合物の製造方法であって、 (a)式A2: の化合物を、第二の非反応性溶媒の存在下で強塩基と反応させて式B1: 〔式中、Mはカリウム、リチウムまたはナトリウムを表す〕のエノラートを生成 させる段階と、 (b)式B1: の化合物を第三の非反応性溶媒の存在下で化合物B2:〔式中、R3はトシル、メシルまたはハロのような離脱基を表す〕と反応させ、 次いでアンモニア試薬の存在下で加熱して式Iの化合物を得る段階と、から成る 方法。 9.Arがモノ−またはジ−置換された3−ピリジニルである請求項8に記載の 方法。 10.R1がCH3またはNH2である請求項8に記載の方法。 11.Arがモノ−またはジ−置換された3−ピリジニルであり、置換基が、 (a)水素、 (b)ハロ、 (c)C1-3アルコキシ、 (d)C1-3アルキルチオ、 (e)C1-3アルキル、 (f)CF3、及び、 (g)CN から成るグループから選択される請求項1に記載の方法。 12.R1がCH3またはNH2であり、 Arがモノ−またはジ−置換された3−ピリジニルであり、置換基が、 (a)水素、 (b)ハロ、 (c)C1-3アルキル、 (d)CF3、及び、 (e)CN から成るグループから選択される請求項8に記載の方法。 13.R2がClであり、 R1がCH3またはNH2であり、 Arがモノ−置換された3−ピリジニルであり、置換基が、水素及びC1-3アル キルから成るグループから選択される請求項8に記載の方法。 14.R2がクロロである請求項1に記載の方法。 15.R3がクロロである請求項8に記載の方法。 16.アンモニウム試薬がアンモニア及び酢酸アンモニウムから選択される請求 項1または8に記載の方法。 17.強塩基がリチウムビス(トリメチルシリル)アミドである請求項8に記載 の方法。 18.第三の非反応性溶媒がトルエンである請求項8に記載の方法。 19.式: 〔式中、 R1は、 (a)CH3、 (b)NH2、 (c)NHC(O)CF3、 (d)NHCH3 から成るグループから選択され; Arは、モノ−、ジ−またはトリ置換フェニルまたはピリジニル(またはその N−オキシド)を表し、置換基は、 (a)水素、 (b)ハロ、 (c)C1-4アルコキシ、 (d)C1-4アルキルチオ、 (e)CN、 (f)C1-4アルキル、 (g)C1-4フルオロアルキル から成るグループから選択され; R2は、 (a)F、Cl、Br、I、 (b)CN、 (c)アジド から成るグループから選択される〕の化合物。 20.R1がCH3またはNH2であり、 Arがモノ−またはジ−置換された3−ピリジニルであり、置換基が、 (a)水素、 (b)ハロ、 (c)C1-3アルキル、 (d)CF3、及び、 (e)CN から成るグループから選択される請求項19に記載の化合物。
