Claims (1)
- 【特許請求の範囲】 1.低pH環境から脂質層を横切ってより高いpH水性コンパートメントへ化合物 を輸送するための組成物であって、 (a)1つ以上の対の側鎖カルボキシル基を含むポリペプチドであって、ここ で (i)1対の該カルボキシル基が、0、2または3アミノ酸によって分けられ 、 (ii)該ポリペプチドが、約8と約100との間のアミノ酸残基の長さを有し、 (iii)該ポリペプチドが、該低pH環境から該脂質層へ分配するのに効果的な 親油性形態と、該脂質層から該より高いpHの水性コンパートメントへ優先的に分 配するのに効果的な親水性形態との間で、可逆的遷移を受けるのに効果的であり 、そして (iv)該ポリペプチドが、該脂質層への末端領域の侵入を促進するために、該 ポリペプチドの該末端領域にイニシエーター部分を含み、 ここで、該イニシエーター部分が、(i)3〜12アミノ酸残基を有するイニシ エーターポリペプチド配列であって、ここで非酸側鎖残基対酸側鎖残基の比が1 より大きい、配列、(ii)NまたはC末端で正または負電荷を排除するのに効 果的であり、そして該末端付近で極性部位を保護するのに効果的な少なくとも1 つの遠隔の極性基を有する、該NまたはC末端に共有結合した基、(iii)送達 されるべき親油性化合物、および(iv)それらの組み合わせ、からなる群より選 択され; これによって、該ポリペプチドが、該低pH環境から該より高いpHコンパートメ ントへ該脂質層を横切ることが可能である、ポリペプチド、および (b)該ポリペプチドに共有結合した、輸送されるべき化合物、 を含む、組成物。 2.前記イニシエーター部分が、前記イニシエーターポリペプチド配列である 、請求項1に記載の組成物。 3.前記配列が、5〜10アミノ酸残基を有する、請求項2に記載の組成物。 4.前記イニシエーター配列におけるアミノ酸残基が、グルタミン酸、ロイシ ン、ノルバリン、メチオニン、アラニン、α-アミノ酪酸、β-アミノ酪酸、およ びβ-アラニンからなる群より選択され、ここで非グルタミン酸対グルタミン酸 残基の比が1より大きい、請求項2に記載の組成物。 5.前記NまたはC末端に共有結合した前記基が、前記ポリペプチドの合成を 開始するために使用される該C末端でのβ-アラニン部分、該ポリペプチドを終 結するために使用される該N末端でのジカルボン酸、およびこの2つの組み合わ せから選択される、請求項1に記載の組成物。 6.前記ポリペプチドを形成するアミノ酸残基の30〜100パーセントがグルタ ミン酸である、請求項1に記載の組成物。 7.前記ポリペプチドが、前記末端領域にイニシエーター部分を有するポリグ ルタミン酸である、請求項6に記載の組成物。 8.前記イニシエーター部分が、前記ポリペプチドの合成を開始するために使 用される前記C末端でのβ-アラニン部分、該ポリペプチドを終結するために使 用される前記N末端でのジカルボン酸、およびこの2つの組み合わせから選択さ れる、請求項7に記載の組成物。 9.前記輸送された化合物の共有結合に使用されない前記ポリペプチドの非酸 側鎖残基が、該ポリペプチドが低pHα-ヘリカルコンホメーションである場合に カチオン性である部分を含まない、請求項1に記載の組成物。 10.前記非酸側鎖残基が、フェニルアラニン、ロイシン、イソロイシン、メ チオニン、バリン、ノルバリン、アラニン、α-アミノ酪酸、ならびにアスパラ ギン酸およびグルタミン酸の側鎖エステルからなる群より選択される、請求項9 に記載の組成物。 11.前記非酸側鎖残基が、ロイシン、メチオニン、アラニン、およびα-ア ミノ酪酸から選択される、請求項10に記載の組成物。 12.前記ポリペプチドが、約24と100との間のアミノ酸残基の所定の長さを 有する、請求項1に記載の組成物。 13.前記ポリペプチドが、約24と48との間のアミノ酸残基の所定の長さを有 する、請求項12に記載の組成物。 14.前記ポリペプチドが、約8と約100との間のグルタミン酸および非グル タミン酸残基の所定の配列を有する、請求項1に記載の組成物。 15.前記所定の配列のポリペプチドが、50%未満の酸側鎖アミノ酸残基を有 する、請求項14に記載の組成物。 16.化合物/ポリペプチド比が、5:1以下である、請求項1に記載の組成物 。 17.前記組成物が、前記イニシエーター部分を有する末端とは反対のポリペ プチドの末端にまたはその付近に結合した、単一の結合した化合物を含む、請求 項16に記載の組成物。 18.前記化合物が、タキソール、シクロスポリン、アンホテリシンB、およ び配列特異的核酸結合ポリマーからなる群より選択される、請求項1に記載の組 成物。 19.前記化合物が、配列特異的核酸結合ポリマーである、請求項18に記載 の組成物。 20.低pH環境から脂質層を横切ってより高いpH水性コンパートメントへの化 合物の輸送を促進するための方法であって、 1つ以上の対の側鎖カルボキシル基を含むポリペプチドに該化合物を共有結合 する工程であって、ここで (i)1対の該カルボキシル基が、0、2または3アミノ酸によって分けられ 、 (ii)該ポリペプチドが、約8と約100との間のアミノ酸残基の長さを有し、 (iii)該ポリペプチドが、該低pH環境から該脂質層へ分配するのに効果的な 親油性形態と、該より高いpHの水性コンパートメントへ優先的に分配するのに効 果的な親水性形態との間で、可逆的遷移を受けるのに効果的であり、そして (iv)該ポリペプチドが、該脂質層への末端領域の侵入を促進するために、該 末端領域にイニシエーター部分を含み、 ここで、該イニシエーター部分が、(i)3〜12アミノ酸残基を有するイニシ エーターポリペプチド配列であって、ここで非酸側鎖残基対酸側鎖残基の比が1 より大きく、同じ長さのポリグルタミン酸がαヘリックスを形成するpHよりも 高いpHでαヘリックスを形成するのに効果的である、配列、(ii)NまたはC 末端で正または負電荷を排除するのに効果的であり、そして該末端付近で極性部 位を保護するのに効果的な少なくとも1つの遠隔の極性基を有する、該Nまたは C末端に共有結合した基、(iii)送達されるべき親油性化合物、および(iv) それらの組み合わせ、からなる群より選択され; これによって、該ポリペプチドおよび結合した化合物が、該低pHコンパートメ ントから該より高いpHコンパートメントへ該脂質層を横切ることが可能である、 工程 を包含する、方法。 21.前記ポリペプチドの非酸側鎖残基が、フェニルアラニン、ロイシン、イ ソロイシン、メチオニン、バリン、ノルバリン、アラニン、α-アミノ酪酸、な らびにアスパラギン酸およびグルタミン酸の側鎖エステルからなる群より選択さ れる、請求項20に記載の方法。 22.前記ポリペプチドが、該ポリペプチドの合成を開始するために使用され る前記C末端でのβ-アラニン部分、該ポリペプチドを終結するために使用され る前記N末端でのジカルボン酸、およびこの2つの組み合わせから選択されるイ ニシエーター部分を有するポリグルタミン酸である、請求項20に記載の方法。 23.前記ポリペプチドが50%未満の酸側鎖アミノ酸残基を有する所定の配列 のポリペプチドである、請求項20に記載の方法。 24.前記化合物/ポリペプチド比が5:1以下である、請求項20に記載の方 法。 25.前記化合物が、タキソール、シクロスポリン、アンホテリシンB、およ び配列特異的核酸結合ポリマーからなる群より選択される、請求項24に記載の 方法。 26.前記送達されるべき化合物が治療化合物である、所定のpHを有する細胞 外培地から、より高いpHを有する細胞のサイトゾルへ、該化合物を送達するこ とにおける使用のための、請求項20に記載の方法。 27.前記細胞外培地が胃液であり、前記細胞がH.pylori細菌細胞であり、 そして前記結合した化合物が抗細菌剤である、H.pylori感染の処置での使用の ための、請求項26に記載の方法。 28.前記細胞が酸性の細胞外培地に存在する腫瘍細胞であり、前記結合した 化合物が抗新生物剤である、抗腫瘍治療での使用のための、請求項26に記載の 方法。 29.前記細胞が酸生成カリエス誘発性細菌細胞であり、そして前記結合した 化合物がカリエス誘発性細菌に対して効果的な抗細菌剤である、歯の崩壊の処置 および予防での使用のための、請求項26に記載の方法。 30.前記脂質層が角質層の細胞外マトリクスを含む、角質層を通る化合物の 経皮送達での使用のための、請求項20に記載の方法。
