1379677 1-Isopropyl-4-(4<SP>1</SP>-fluorophenyl)-7- methyl-2(1H)-quinazolinone SANDOZ Ltd 21 June 1972 [24 June 1971 18 Nov 1971] 29063/72 Heading C2C Novel 1 - isopropyl - 4 - (4<SP>1</SP> - fluorophenyl)- 7-methyl-2(1H)-quinazolinone is prepared (a) by reacting an alkyl (C 1-4 ) N-isopropyl-N-[2- (4<SP>1</SP> - fluorobenzoyl) - 5 - methyl - phenyl] carbamate with an ammonium salt of a C 1-4 aliphatic mono- or di-carboxylic acid or (b) by cyclizing 2 - N - isopropylamino - 4 - methyl- 4<SP>1</SP>-fluorobenzophenone with urea. Alkyl (C 1-4 ) N-isopropyl-N-[2-(4<SP>1</SP>-fluorobenzoyl) - 5 - methyl - phenyl]carbamates are prepared by reacting 2-N-isopropylamino-4- methyl-4<SP>1</SP>-fluorobenzophenone with a C 1-4 alkyl chloro- or bromo-formate. Pharmaceutical compositions having antiinflammatory and analgesic activity comprise, as active ingredient, 1-isopropyl-4-(4<SP>1</SP>-fluorophenyl - 7 - methyl - 2(1H) - quinazolinone, in association with a pharmaceutically acceptable diluent or carrier. Reference has been directed by the Comptroller to Specifications 1,248,430 and 1,181,570. [GB1379677A]

Claims (12)

39743/2 CLAIMS:
1. A process for the production of the compound formula I, characterised by a) reacting a compound of formula 39743/2 in which R signifies an alkyl radical of 4 carbon atoms, with an ammonium salt of a C^_^ aliphatic mono- or carboxylic acid, or b) cyclising the compound of formula III, with urea.
2. A process for the production of the compound of formula I, stated in Claim 1, characterised by reacting a compound of formula II, stated in Claim 1, with an ammonium salt of a C^_^ aliphatic mono- or di-^ carboxylic acid.
3. A process for the production of the compound of formula I, stated in Claim 1, characterised by cyclising the compound of formula III, stated in Claim 1, with urea. 39743/2
4. A process according to Claim 2, in which the substituent R of the compound of formula II, signifies methyl or ethyl.
5. A process according to Claim 2 or 4 , in which the ammonium salt is ammonium formate, ammonium acetate, ammonium oxalate or ammonium succinate..
6. A process according to any one of Claims 2, 4 or 5, in which the reaction is effected at a temperature of from 70° to 180°C.
7. A process according to Claim 3, in which the reaction is effected in the presence of a carboxylic acid of 2 to 4 carbon atoms and using at least 3 moles of urea per mole of compound III.
8. A process according to Claim 3 or 7 , in which the reaction is carried out at a temperature of from 80° to 160°C.
9. A process, for the production of the compound of formula I, stated in Claim 1, substantially as herein described with reference to either of the Examples.
10. The compound of formula I, stated in Claim 1, whenever produced by a process according to any one of Claims 1 to 9.
11. The compound of formula I, stated in Claim 1.
12. A pharmaceutical composition comprising the compound of formula I, stated in Claim 1, in association with a pharmaceutically acceptable diluent or carrier.
