Abstract
A reagent combination for the colorimetric analysis of urea nitrogen in biological fluids comprises (1) a vial of an anhydrous mixture of phenol and an alkali metal nitroprusside; (2) a vial of an alkaline aqueous solution of alkali metal hypochlorite; and (3) a vial of buffered urease, there being a sufficient excess of alkali metal cations in said second vial to convert the whole of the phenol in the said first vial to an alkali metal phenate. The alkali metal hypochlorite may be NaClO and said excess alkali in the aqueous solution may be: NaOH, K2CO3, K3PO4 or KOH. The solid nitroprusside may be in the form of a single crystal or particulate and may be added to the melted phenol or the two mixed as solids under anhydrous conditions. To the mixture of buffered urease and urea the nitroprusside and phenol are added either sequentially or together, followed, after up to 2 hours, by the alkaline hypochlorite and the colour measured e.g. on a spectrophotometer. The urease may be buffered with: ethylene diamine tetraacetic acid, KH2PO4 or Na2HPO4.ALSO:A reagent for use in determining ammonia nitrogen in solution comprises an anhydrous mixture of phenol and an alkali metal nitroprusside, and may be part of a reagent combination for the colorimetric analysis of urea nitrogen in biological fluids comprising (1) a vial of an anhydrous mixture of phenol and an alkali metal nitroprusside, (2) a vial of an alkaline aqueous solution of alkali metal hypochlorite, and (3) a vial of buffered urease, there being a sufficient excess of alkali metal cations in said second vial to cnvert the whole of the phenol in the said first vial to an alkali metal phenate. The alkali metal hypochlorite may be NaClO and said excess alkali in the aqueous solution may be: NaOH, K2CO3, K3PO4 or KOH. The solid nitroprusside may be in the form of a single crystal or particulate and may be added to the melted phenol or the two mixed as solids under anhydrous conditions. To the mixture of buffered urease and urea the nitroprusside and phenol are added either sequentially or together, followed, after up to 2 hours, by the alkaline hypochlorite and the colour measured, e.g., on a spectrophotometer. The urease may be buffered with: ethylene diamine tetraacetic acid, KH2PO4 or Na2HPO4.
