Abstract
The invention comprises racemic compounds of the general formula <FORM:0981533/C2/1> (wherein R1 is a hydrogen atom or a methyl group, R* is a C1- 5 alkyl group, R0 is a hydrogen atom or a C1- 5 alkyl group and X is an OR1 group in the 4- or 5-position, being identical to the OR1 group in the 2-position) and non-toxic acid addition salts thereof, pharmaceutical preparations containing them, and their preparation by reacting a compound of the formula <FORM:0981533/C2/2> (wherein X0 is a methoxy group in the 4- or 5-position) with ammonium carbonate and a water-soluble cyanide salt to form a compound of the formula <FORM:0981533/C2/3> reacting the latter compound with a base to form a salt of a racemic compound of the formula <FORM:0981533/C2/4> and acidifying to produce the free acid form of that compound, optionally followed by one or more of the steps (a) demethylation with an aqueous hydrohalic acid, (b) esterification with a C1- 5 alkanol, and (c) acid addition salt formation. The starting materials are prepared by reacting a compound of the formula <FORM:0981533/C2/5> with R*CH2NO2 and reducing the product with iron and hydrochloric acid. The compounds of the invention are useful in the treatment of hypertension, and may be administered orally or parenterally in the form of pharmaceutical preparations (e.g. capsules, tablets, solutions and suspensions) containing them together with a diluent or carrier.
