Abstract
The invention comprises compounds having the formulae <FORM:0924981/IV(a)/1> <FORM:0924981/IV(a)/2> and the corresponding D 1,4, D 1,4,6, and D 4,6 compounds wherein X is hydrogen, fluorine, chlorine and bromine, Y is halogen, alkoxy containing up to 10 carbon atoms, hydroxyl or acylated hydroxyl, the acyl group containing up to 10 carbon atoms and being derived from a mono- or di-carboxylic acid, and R is hydrogen, hydroxyl or acylated hydroxyl, and therapeutic compositions thereof. The 21-halomethyl compounds of the above formulae are obtained by reacting the corresponding 21-methylene compound with anhydrous hydrogen halide in an inert solvent. Reacting the 21-methylene compound with an alkanol results in a 22-alkoxy compound. This may be reduced to a 21-hydroxymethyl by hydrogenation provided the 3-keto group is protected, e.g. by forming a formate ester. The 17- and 21-hydroxyl groups in the resulting compounds may be esterified by the usual methods. The compositions have progestational activity and comprise the above compounds and a pharmaceutical carrier.
