Abstract
The invention comprises quaternary ammonium salts of N-substituted 10-(N,N-dimethyl)-b -amino-propyl-phenothiazine derivatives having the general formula <FORM:0881379/IV (b)/1> in which n is either 1 or 2 and X denotes the chloride ion or the methylsulphate radical and processes for the preparation thereof by (a) reacting a tertiary amine of the general formula <FORM:0881379/IV (b)/2> with either methyl chloride or dimethyl sulphate or (b) reacting N,N-dimethyl-N([a -methyl-b -phenothiazinyl-10]-ethyl)-amine with either b -hydroxy - ethyl chloride or g - hydroxy - propyl chloride, and if desired, the further step of converting the quaternary ammonium chloride, obtained as product, into the corresponding methylsulphate by treatment with dimethyl sulphate. Illustrative products of the invention are N,N-dimethyl-N-([a -methyl-b -phenothiazinyl - 10] - ethyl) - N - (b - hydroxy - ethyl) - ammonium chliride, N,N-dimethyl-N-([a -methyl-b - phenothiazinyl - 10] - ethyl) - N - (g - hydroxypropyl) - ammonium chloride and N,N - dimethyl - N - ([a - methyl - b - phenothiazinyl - 10] - ethyl) - N - (g - hydroxy - propyl) - ammonium methylsulphate. The Provisional Specification describes also N,N-dialkyl derivatives wherein the alkyl groups have preferably 1-3 carbon atoms, and states that the hydroxyalkyl group may have a branched chain.ALSO:Anti-allergic pharmaceutical compositions comprise quaternary ammonium salts of N-substituted 10-(N,N-dimethyl)-b -amino-propyl phenothiazine derivatives having the general formula <FORM:0881379/VI/1> in which n is either 1 or 2 and X denotes the chloride ion or the methylsulphate radical, dispersed or disposed in a pharmaceutical vehicle. Such compositions are administered orally in the form of a tablet, pellet, pill, dragee, lozenge, or capsule or parenterally. An example describes the preparation of tablets containing N,N-dimethyl - N - ([a - methyl - b - phenothiazinyl-10] - ethyl) - N - (b - hydroxyethyl) ammonium chloride.
