Abstract
Compounds of ion-exchange resins and ionizable therapeutic agents are prepared by contacting the resin in suitable form with a solution of the therapeutic agent. Suitable resins are copolymers of polystyrene cross-linked with divinyl benzene or aliphatic ester, containing quaternary ammonium or sulphonic acid groups; polystyrene resins containing dibenzylamine groups; and polymerized methacry ic acid resins having a degree of cross-linkage between 1% and 15% and containing carboxyl groups, examples given being those available under the names "Zeokarb" (Registered Trade Mark) 226, 225 and 225H; "Deacidite" (Registered Trade Mark) D, G and FF; "Deminrolit" (Registered Trade Mark). Specified therapeutic agents are b -phenylethylamine derivatives, e.g. ephedrine; dexamphetamine; barbituric acid derivatives, e.g. amylobarbitone; antihistamines; alkaloids e.g. morphine, codeine, hyocy<\>aamine; para-amino-salicylic acid. The resin/drug compounds may be made into pharmaceutical compositions comprising aqueous media, oil in water emulsions, glycerine, cellulose ether, vegetable oil and sucrose esters. Specifications 823,000, 829,245 and 838,952 are referred to.ALSO:Pharmaceutical compositions comprise a carrier liquid substantially free of ions or ionizable material and, dispersed therein, a "resin-drug" compound formed by reacting a cationic or anionic ion-exchange resin with a drug. The carrier liquid may be water, a non-ionizing polyhydric alcohol, e.g. glycerine, or an oil in water emulsion and is preferably an aqueous vehicle or other mixed menstrum in which water is the major component. Non-ionizing sweetening and flavouring agents may be added and the carrier liquid may itself be a sweetening and/or flavouring agent. Non-ionic emulsifying agents, e.g. a sucrose ester, e.g. sucrose distearate, or a soluble cellulose ether may also be present. Specified therapeutically-active substances are b -phenylethylamine derivatives, e.g. ephedrine and dexamphetamine, bartituric acid derivatives, e.g. amylobarbitone, antihistamines, alkaloids, e.g. morphine, codeine, hyoseyamine, hydroxybenzoic acid derivatives, e.g. p-amino-salicylic acid. Specified resins are copolymers of polystyrene with divinyl benzene or aliphatic esters containing quaternary ammonium or sulphonic acid groups; polystyrene resins containing dibenzylamine groups; and polymerized methacrylic acid resins having a degree of crosslinkage between 1 and 15% and containing carboxyl groups, examples being those available under the Registered Trade Marks "Zeokarb" (Nos. 225, 225H and 226), "Deacidite" (D, G and FF), and "Deminrolit" (a mixed anionic and cationic resin). Specifications 823,000, 829,245 and 838,952 are referred to.
