Abstract
A drug destined for injection is compressed under a pressure of at least 2500 Kg/cm.2 to a solid body which is subdivided to particles having a maximum size of 250 microns and suspended in a parenterally acceptable liquid carrier in which the drug is substantially insoluble. Alternatively, especially if the drug is soluble, it may be mixed with a solid carrier which is sparingly soluble in the liquid medium before compression. Examples of drugs are: heparin-butacaine, glucagon, insulin (amorphous and coupled with salmine), insulin and glucagon, ACTH, ACTH-protamine-zinc, lipoic acid, thyrotropic hormone, antidiuretic hormone, testosterone, procaine-penicillin, heparin, Vitamin B12, atropine tannate. Zinc, aluminium and tricalcium phosphates are specified solid carriers. The liquid carrier may be aqueous or an oil and may contain additional agents which retard the resorption of the active substance, e.g. gelatine, dextrans or other high molecular weight polysaccharides. Metal ions, e.g. zinc, which effect the period of action of certain drugs may also be present. The suspensions may be lyophilized to give a powder which may be resuspended in a suitable liquid carrier before use.
