Abstract
Relaxin preparations containing impurities having an isoelectric point higher than that of relaxin are purified by bringing an aqueous solution having a pH greater than the isoelectric point of relaxin, say 7-8.6, into contact with a carboxylic acid type cation exchange resin buffered to the same pH, and washing unabsorbed material from the resin with a buffered solution of pH not greater than that of the buffered resin thereby recovering the purified relaxin. The resin may be regenerated by eluting the absorbed impurities with acid. Relaxin preparations containing impurities of lower isoelectric point are purified by bringing an aqueous solution of lower pH than the isoelectric point of relaxin, say 4.5-7, into contact with the said resin buffered to the same pH, and washing unabsorbed material from the resin, purified relaxin being then obtained by washing the resin with acid. Suitable resins are those formed by condensation of a phenolic carboxylic acid and an aldehyde, or preferably, copolymers of acrylic or methacrylic acid and divinyl benzene wherein the latter constitutes 2 1/2 -5 per cent of the resin composition. Prior to or after purification, electrolytes are removed by dialysis, or by treatment with a sulphonic acid type cation exchange resin (acid form), followed by treatment with an anion exchange resin. The product may be freeze-dried.
