Abstract
a - (N - (b - Diethylaminoethyl)) - aminophenylacetic acid isoamyl ester hydrochloride is prepared by reacting a -bromophenylacetic acid isoamyl ester with b -diethylaminoethylamine and converting the resulting base to its hydrochloride. The reaction is preferably carried out using equimolecular quantities of the reactants in a solvent, but may also be carried out in the absence of a solvent and with cooling. A dehydrobrominating agent such as potassium carbonate or an excess of the amine may be present. The a -bromophenylacetic acid isoamyl ester used as the starting material may be made by brominating phenylacetyl chloride to give a -bromophenylacetyl chloride and converting this into the desired ester by reation with isoamyl alcohol. Examples describe the reaction of a -bromophenyl acetic acid isoamyl ester with b -diethylaminoethylamine to give a -(N - (b - diethylaminoethyl) - aminophenyl-acetic acid isoamyl ester (1) in the absence of a solvent at below 10 DEG C.; (2) by refluxing in acetone in the presence of potassium carbonate; and (3) by refluxing in benzene.
